Bacterial blight (BB) disease of rice, caused by Xanthomonas oryzae pv. oryzae (Xoo), is one of the most serious diseases in all major rice-growing areas of Thailand. The most effective approach to control BB is the use of resistant varieties. However, the selection pressure from the usage of resistant cultivars may result in mutation of the pathogen population to overcome the resistant genes. Therefore, using genetic resources conferring different resistant genes are more advantage in rice improvement. The aim of this research was to survey and screen for BB resistant genes in Thai local rice varieties from the National Rice Seed Storage Laboratory for Genetic Resources to gain more genetic diversity for the resistance. In this study, the Thai Local rice cultivars were screened for bacterial blight reaction against Xoo. One hundred fourteen cultivars were tested in a greenhouse at Ubon Ratchathani Rice Research Center in wet season 2009. There were 3, 93 and 18 of the tested cultivars showing highly resistant (HR), resistant (R) and moderately resistant (MR) to BB, respectively. Those Thai local cultivars showing resistant to BB were analyzed with molecular markers including PASAxA5F1/R1 and PASAxA5F2/R2, STSBB7_6F/R, STSBB13_8F/R and STSBB21_11F/R, which are linked to xa5, Xa7, xa13 and Xa21 gene, respectively. Forty-eight cultivars, such as Khao Pong Kai and Puang Ngern, having Xa7 gene were identified. Nine cultivars like Pra Than Bahn Tung and Khao Luang were found carrying Xa21 gene. Interestingly, five cultivars consisting of Leuang 11, Loi Khao, Rootj ampia, Look Daeng Pattani and Leuang Yai, were found carrying both Xa7 and Xa21 gene.
โรคขอบใบแห้งของข้าว (Bacterial blight) เกิดจากเชื้อแบคทีเรีย Xanthomonas oryzae pv. oryzae (Xoo) พบระบาดรุนแรงและทำความเสียหายในแหล่งปลูกข้าวที่สำคัญทุกภาคของประเทศไทย การใช้พันธุ์ต้านทานโรคขอบใบแห้งเป็นวิธีการที่มีประสิทธิภาพ อย่างไรก็ตาม เชื้อสาเหตุโรคสามารถเข้าทำลายพันธุ์ต้านทานได้ เนื่องจากการกลายพันธุ์ของเชื้อทำให้ได้สายพันธุ์เชื้อใหม่ๆ ที่เข้าทำลายยีนต้านทานได้ ดังนั้นการปรับปรุงพันธุ์ข้าวให้ต้านทานต่อโรคขอบใบแห้งจึงจำเป็นต้องหาแหล่งพันธุกรรมยีนต้านทานโรคที่หลากหลายมาใช้ในการปรับปรุงพันธุ์ การใช้เครื่องหมายโมเลกุลที่วางตำแหน่งอยู่ใกล้กับยีนต้านทานโรคขอบใบแห้งมาคัดเลือกหาแหล่งพันธุกรรมยีนต้านทานที่หลากหลายต่อโรคขอบใบแห้ง จะช่วยในการปรับปรุงพันธุ์ข้าวให้มีความต้านทานต่อโรคได้อย่างมีประสิทธิภาพ งานวิจัยนี้มีวัตถุประสงค์เพื่อสำรวจและคัดเลือกข้าวพันธุ์พื้นเมืองไทยในศูนย์ปฏิบัติการและเก็บเมล็ดเชื้อพันธุ์ข้าวแห่งชาติที่มียีนต้านทานต่อโรคขอบใบแห้ง ที่หลากหลายสำหรับใช้ในงานปรับปรุงพันธุ์ข้าว ดำเนินการคัดเลือกพันธุ์ข้าวต้านทานต่อโรคขอบใบแห้งในสภาพเรือนทดลอง ที่ศูนย์วิจัยข้าวอุบลราชธานี ฤดูปลูกปี 2552 จำนวน 114 พันธุ์ ผลการทดลองพบว่าข้าวพันธุ์พื้นเมืองไทยแสดงปฏิกิริยาต้านทานต่อโรคขอบใบแห้ง ระดับ HR จำนวน 3 พันธุ์ ระดับ R จำนวน 93 พันธุ์ และ ระดับ MR จำนวน 18 พันธุ์ หลังจากนั้นนำข้าวพันธุ์พื้นเมืองไทยที่แสดงปฏิกิริยาความต้านทานทุกพันธุ์ (HR, R และ MR) มาวิเคราะห์หายีนต้านทานต่อโรคขอบใบแห้งโดยการใช้เครื่องหมายโมเลกุล PASAxA5F1/R1, PASAxA5F2/R2, STSXa7_6F/R, STSBB13_8F/R และ STSBB21_11F/R ซึ่งวางตำแหน่งใกล้กับยีน xa5, Xa7, xa13 และ Xa21 ผลการวิเคราะห์ พบข้าวพื้นเมืองไทย จำนวน 48 พันธุ์ที่ตรวจพบยีนต้านทาน Xa7 เช่น พันธุ์ขาวโป่งไคร้ และพวงเงิน พบข้าวจำนวน 9 พันธุ์ที่ตรวจพบยีนต้านทาน Xa21 เช่น ประทานบ้านทุ่ง และ ขาวหลวง นอกจากนี้ยังตรวจพบยีนต้านทานทั้ง 2 ยีน คือ Xa7 และ Xa21 ในข้าวจำนวน 5 พันธุ์ ได้แก่ ข้าวพันธุ์เหลือง 11 ลอยขาว รูจจำเปี๊ยะ ลูกแดงปัตตานี และ เหลืองใหญ่
