受精卵クローン技術は一卵性多子を生産する技術であり,種々の比較試験や能力検定に利用することができる.本研究では,同一受精卵由来の5つ子について,その枝肉を画像解析により詳細に調査し,筋肉形状,脂肪の割合ならびに肉色について類似性を検討することである.黒毛和種供卵牛から受精卵を回収し,ホルスタイン等の卵巣から得た未受精卵子(除核済み)を電気刺激により細胞融合し,核移植を行い,7頭のクローン雄子牛を生産した.そのうち5頭を和牛産肉能力検定間接法にしたがって21ヵ月齢まで肥育した.画像解析により胸最長筋,僧帽筋,菱形筋,頭半棘筋,背半棘筋,前背鋸筋および腸肋筋に関して,その面積,各筋肉内に占める脂肪の面積割合(以下,脂肪面積比)などを算出した.また,脂肪交雑粒子のあらさならびにBCSナンバーを画像解析により数値化した.5頭の供試牛の枝肉重量ならびに胸最長筋の面積の範囲は,それぞれ344kg~400kg,38.5cm2~50.4cm2の範囲にあり,類似性はそれほど高くなかった.胸最長筋内の脂肪面積比は,27.2%から28.4%の範囲にあり,類似性はきわめて高く,他の筋肉においても同様の傾向が認められた.脂肪交雑粒子のあらさについては,低い類似性を示すことが確認された.画像解析により推定したBCSナンバーは,2.92から3.26の範囲であり,比較的類似性は高かった.肥育期間中のコンディションの差異が,肉量に関わる形質に大きな影響を及ぼすものの,脂肪の割合および肉色については,類似性が高いことが確認された. Embryonic cloning is a technique to produce multiple offspring from one embryo and could be used for a diverse range of comparative tests and performance tests. In this study,similarities of muscle shape,fat area ratio to muscle area and meat color were investigated by computer image analysis on quintuplet Japanese Blacks derived from an embryo. Blastomeres obtained from a Japanese Black donor were collected and 7 male clones were produced by cell fusion with electro stimulation using enucleated oocytes from the ovaries of a Holstein. Five of the 7 clones were fattened according to the method of Wagyu progeny testing for 21 mo. For M. longissimus thoracis, M. trapezius, M. rhomboideus, M. semispinalis caρitis, M. semispinalis dorsi, M. anterior serratus and M. iliocostalis, the area and fat area ratio to muscle area (FATPER) were measured by image analysis. The Coarsenesses of marbling particles and the Beef Color Standard number were calculated by image analysis. The range of carcass weight and area of M. longissimus thoracis were 344 to 400 kg and 38.5 to 50.4 cm² respectively.Similarity of FATPER for M. longissimus thoracis was extremely high,ranging from 27.2 to 28.4%, and almost the same result was obtained for other muscles.Low similarity was recognized for the coarseness of marbling particles. Similarity of BCS number by image analysis was high ranging from 2.92 to 3.26.High similarities for FATPER and meat color were confirmed among clones,although meat quantity traits might be affected by conditions during the fattening period.
