The identification of genus and species is made by systematic, by noticing morphological and anatomical characteres which were the phenotype expression. However, changes in one or more gene bases can result in phenotypes similarities between different species, inducing an identification error. Alternatively, molecular appliances based in mithocondrial and ribosomal genes appeared. This study aims to compile information about the molecular identification and present the South American Catfish (Rhamdia quelen) case, for which there is controversy generated by taxonomy identification. The different genes used to identification have their particularities, like insertions and deletions on the case of 16S rRNA and the 18S rRNA. Besides these ribosomal genes, mitochondrial genes are used, like the cytochrome b gene, whose problematic focused on the primer design, and the cytochrome C oxidase subunit 1 gene, which is the best option to make the identification, by generating a genetic barcode. DNA barcoding studies help on the molecular identification of species and allow evolutionary studies in special, regarding the speciation process, revealing mechanisms that may have contributed to the adaption in Brazilian different climes.
A identificação de gêneros e espécies é feita pela sistemática, observando-se caracteres morfológicos e anatômicos, os quais são a expressão do fenótipo. Entretanto, mudanças de uma ou mais bases em genes podem resultar em semelhanças fenotípicas entre espécies diferentes, levando a um erro de identificação. Como alternativa, surgiram ferramentas moleculares baseadas em genes ribossomais e mitocondriais. O presente trabalho tem como objetivo compilar informações sobre a identificação molecular e apresentar o caso do jundiá (Rhamdia quelen), no qual existem controvérsias geradas pela identificação taxonômica. Os diferentes genes utilizados para identificação apresentam suas peculiaridades, como inserções e deleções no caso do 16S rRNA e do 18S rRNA. Além destes genes ribossomais, há o uso de genes mitocondriais, como o gene do citocromo b, cuja problemática se concentra no desenho de primers, e o gene da subunidade 1 do citocromo C oxidase, sendo este último a melhor alternativa para identificação, gerando um Barcode genético. Estudos de DNA barcoding auxiliam na identificação de espécies em nível molecular, bem como, permitem estudos evolutivos em particular, no que tange a respeito da especiação, nos revelando mecanismos que podem ter contribuído para a adaptação nos mais diversos climas brasileiros.
