Benabdelmouna, Abdellah; Maurouard, Elise; D'Amico, Florence; Seugnet, Jean-luc; Grizon, James.

Travers, Marie-agnes; Degremont, Lionel; De Lorgeril, Julien; Azema, Patrick; Montagnani, Caroline; Benabdelmouna, Abdellah; Nicolas, Jean-louis; Le Roux, Frederique.

Parental exposure to the herbicide diuron results in oxidative DNA damage to germinal cells of the Pacific oyster Crassostrea gigas ArchiMer
Barranger, Audrey; Heude-berthelin, Clothilde; Rouxel, Julien; Adeline, Beatrice; Benabdelmouna, Abdellah; Burgeot, Thierry; Akcha, Farida.
Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. In order to explore this question, the levels and cell distribution of the oxidized base lesion 8-oxodGuo were studied in the gonads of exposed genitors. High-performance liquid chromatography...

Benabdelmouna, Abdellah; Hemissi, Issam.

Single or dual experimental infections with Vibrio aestuarianus and OsHV-1 in diploid and triploid Crassostrea gigas at the spat, juvenile and adult stages ArchiMer
Azema, Patrick; Travers, Marie-agnes; Benabdelmouna, Abdellah; Degremont, Lionel.
French production of the Pacific cupped oyster, Crassostrea gigas, is currently threatened by two pathogens, OsHV-1 and V. aestuarianus. While oysters selected for their higher resistance to OsHV-1 are now available for the industry, the impact of V. aestuarianus on such oysters is unknown, especially for triploids. In addition, experimental infection has used the virus or the bacteria alone, but there have been no investigations of dual exposure to these pathogens. This study is the first report of single or dual exposure in spat (Spat1 and Spat2), juvenile and adult naïve oysters. For each of the two stocks evaluated, unselected oysters and oysters selected for their higher resistance to OsHV-1 infection were tested, as well as their triploid siblings of...

Autotetraploid Pacific oysters (Crassostrea gigas) obtained using normal diploid eggs: induction and impact on cytogenetic stability ArchiMer
Benabdelmouna, Abdellah; Ledu, Christophe.
We describe two methods of producing viable and fertile autotetraploid Pacific oyster (Crassostrea gigas Thunberg) based on the use of normal-sized oocytes produced by normal diploid females. Our methods showed that the oocyte size is not a limiting factor for the success of the induction to autotetraploidy. These methods offer means of direct introgression of genetic progress from elite diploid lines to tetraploids used as broodstock, avoiding a triploid step with the risk of transferring undesirable traits from highly fecund triploids. High variability in the level of cytogenetic stability was found among the different tetraploid oysters tested showing that induction method has an important impact on the long-term cytogenetic stability of the...

The mass mortality of blue mussels (Mytilus spp) from the Atlantic coast of France is associated with heavy genomic abnormalities as evidenced by flow cytometry ArchiMer
Benabdelmouna, Abdellah; Ledu, Christophe.
Since 2014, France’s blue mussel industry has been facing heavy mortality outbreaks (90-100%) affecting both juveniles and adults. This report presents evidence of heavy genomic abnormalities associated with mortality outbreaks in blue mussels, Mytilus edulis-galloprovincialis, from the Atlantic coast of France. In this study, ploidy characteristics of hemic cells were investigated using Flow CytoMetry (FCM), revealing an unusual, broad continuum of ploidy distribution from hypodiploidy to tetraploidy. FCM was additionally used to evaluate, at individual and populations levels, different thresholds of genomic abnormality (GA%) using the percentage of non-diploid nuclei. Individual mussels were considered to be abnormal when more than 10% of hemocytes in...

Ostreid herpesvirus type 1 replication and host response in adult Pacific oysters, Crassostrea gigas ArchiMer
Segarra, Amelie; Baillon, Laury; Tourbiez, Delphine; Benabdelmouna, Abdellah; Faury, Nicole; Bourgougnon, Nathalie; Renault, Tristan.
Since 2008, massive mortality outbreaks associated with OsHV-1 detection have been reported in Crassostrea gigas spat and juveniles in several countries. Nevertheless, adult oysters do not demonstrate mortality in the field related to OsHV-1 detection and were thus assumed to be more resistant to viral infection. Determining how virus and adult oyster interact is a major goal in understanding why mortality events are not reported among adult Pacific oysters. Dual transcriptomics of virus-host interactions were explored by real-time PCR in adult oysters after a virus injection. Thirty-nine viral genes and five host genes including MyD88, IFI44, IkB2, IAP and Gly were measured at 0.5, 10, 26, 72 and 144 hours post infection (hpi). No viral RNA among the 39...

Genitors of the Pacific oyster Crassostrea gigas were submitted during gametogenesis to a short pulse exposure to the herbicide diuron at a realistic environmental concentration. Histological analysis showed no effect of diuron on gametogenesis course, sex ratio and reproductive effort. A non-significant increase in testosterone and progesterone levels was observed in genitors exposed to the herbicide. At cell level, diuron exposure was shown to modulate the phagocytic activity of circulating hemocytes. The results of a transcriptional analysis showed that diuron affected the expression of genes belonging to functions known to play a major role during oyster gametogenesis such as gene transcription regulation, DNA replication and repair, DNA methylation...

Mortality associated with OsHV-1 in spat Crassostrea gigas: role of wild-caught spat in the horizontal transmission of the disease ArchiMer
Degremont, Lionel; Benabdelmouna, Abdellah.
The French oyster production of Crassostrea gigas is based on two sources of spat: wild-caught (WC) and hatchery-produced (HP). Massive mortality related to the ostreid herpesvirus type 1 (OsHV-1) has affected both sources in France since 2008. We investigated the mortality in juvenile C. gigas due to the horizontal transmission of OsHV-1 within (separated condition) and between (mixed condition) the two spat sources in three environments from April to June 2010. In the separated condition, no mortality was observed in the HP batches, while the WC batches experienced moderate to high mortality (40–80 %). In contrast, the WC and HP batches experienced high mortality in all tested environments for the mixed condition. At the beginning of the trial, the HP...

Study of genetic damage in the japanese oyster induced by an environmentally-relevant exposure to diuron: evidence of vertical transmission of dna damage ArchiMer
Barranger, Audrey; Akcha, Farida; Rouxel, Julien; Brizard, Raphael; Maurouard, Elise; Pallud, Marie; Menard, Dominique; Tapie, N.; Budzinski, Helene; Burgeot, Thierry; Benabdelmouna, Abdellah.
Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Commercially-raised bivalves are particularly exposed to pollutants, among them pesticides, as shellfish farming zones are subject to considerable pressure from agricultural activities on the mainland. The aims of this study were to determine (1) the genotoxic effects of diuron exposure on oyster genitors and (2) the possible transmission of damaged DNA to offspring and its repercussions on oyster fitness. To investigate these points, oysters were exposed to concentrations of diuron close to those detected in the Marennes-Oleron Basin (two 7-day exposure pulses at 0.4 and 0.6 μg...

Effect of ploidy on the mortality of Crassostrea gigas spat caused by OsHV-1 in France using unselected and selected OsHV-1 resistant oysters ArchiMer
Degremont, Lionel; Ledu, Christophe; Maurouard, Elise; Nourry, Max; Benabdelmouna, Abdellah.
The effect of ploidy on the mortality of Crassostrea gigas spat caused by the ostreid herpesvirus (OsHV-1) genotype μVar was investigated at five sites along the Atlantic coast in France in 2011. Sibling diploids and triploids were produced using either unselected or selected OsHV-1-resistant oysters. No significant interactions were found between the factors of environment, genotype and ploidy at the endpoint dates. The mean mortality rates at the sites were 62 and 59% for diploids and triploids, respectively, and the two rates were not significantly different. The mean mortality rates were 33 and 32% for sibling diploids and triploids, respectively, when OsHV-1-resistant parents were used, and 91 and 85%, respectively, when unselected parents were used....

Synthesis of the “PLAN DE SAUVEGARDE” using selected all-triploid oysters to reduce the shortage of spat in France due to OsHV-1–associated mortality in Crassostrea gigas ArchiMer
Dégremont, Lionel; Maurouard, Elise; Ledu, Christophe; Benabdelmouna, Abdellah.
Due to massive mortality of Crassostrea gigas spat in France since 2008, a “plan de sauvegarde” was set up from 2011 to 2014 (hereafter referred to as PS1 to PS4), in order to reduce the shortage of spat. This plan involved the participation of commercial hatcheries, the French Research Institute for Exploitation of the Sea (Ifremer), and the Direction des Pêches Maritimes et de l'Aquaculture (DPMA) of the French Ministry of Agriculture. It was based on selecting diploid lines of C. gigas for their higher resistance to the oyster herpesvirus OsHV-1 (2nR group), and one of these lines was subsequently tetraploidized (4nR group). Both the 2nR and 4nR groups were produced by Ifremer, and then transferred to commercial hatcheries that produced the selected...

Parental exposure to environmental concentrations of diuron leads to aneuploidy in embryos of the Pacific oyster, as evidenced by fluorescent in situ hybridization ArchiMer
Barranger, Audrey; Benabdelmouna, Abdellah; Degremont, Lionel; Burgeot, Thierry; Akcha, Farida.
Changes in normal chromosome numbers (i.e. aneuploidy) due to abnormal chromosome segregation may arise either spontaneously or as a result of chemical/radiation exposure, particularly during cell division. Coastal ecosystems are continuously subjected to various contaminants originating from urban, industrial and agricultural activities. Genotoxicity is common to several families of major environmental pollutants, including pesticides, which therefore represent a potential important environmental hazard for marine organisms. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to short-term exposure to the herbicide diuron at environmental concentrations during gametogenesis. In this paper, Fluorescent in...

Fluorescence in situ hybridization (FISH) or rDNA genes (5S and 18-5.8-28S) as a tool for chromosomal tagging and to assess the occurrence of somatic and sperm aneuploidy in Pacific oyster Crassostrea gigas ArchiMer
Benabdelmouna, Abdellah; Nadolna, Katarzyna; Ledu, Christophe.
Aneuploidy (or alteration of chromosome number) has been frequently described in diploid, triploid and tetraploid C. gigas. In diploid Pacific oysters, aneuploidy affects 1 to 3 chromosomes in up to 35% of the somatic cells and is positively correlated with inferior growth of the animal and thus could be associated with significant economic loss in this major aquaculture species. In polyploid Pacific oysters, aneuploidy was shown to be more heavily frequent and is linked to reversion to lower ploidy levels and is at the origin of aberrant gametes formation. In this context, identification of individual chromosomes involved in aneuploidy events both in somatic and germinal cells would be an important step towards the study of the exact occurrence of this...

Gamete quality in triploid Pacific oyster (Crassostrea gigas) ArchiMer
Suquet, Marc; Malo, Florent; Quere, Claudie; Ledu, Christophe; Le Grand, Jacqueline; Benabdelmouna, Abdellah.
Triploidy induction in shellfish aims to obtain faster growth and sterility of reared individuals. Triploid Pacific oysters are most often not totally sterile, but have greatly reduced reproductive capacities compared to those recorded in diploid individuals. The description of gamete output in triploid aquatic animals is most often confined to the assessment of fecundity in females. The present work aims at further describing gamete quality of triploid Pacific oysters and comparing it to values observed in diploid individuals. Diploid and triploid oysters (produced by mating tetraploid males × diploid females) were reared in standard conditions and then transferred to the wild to allow the offspring to attain reproductive maturity.
