Gill transcriptomic analysis in fast- and slow-growing individuals of Mytilus galloprovincialis ArchiMer
Prieto, Daniel; Markaide, Pablo; Urrutxurtu, Iñaki; Navarro, Enrique; Artigaud, Sebastien; Fleury, Elodie; Ibarrola, Irrintzi; Urrutia, Miren Bego.
The molecular basis underlying the mechanisms at the origin of growth variation in bivalves is still poorly understood, although several genes have been described as upregulated in fast-growing individuals. In the present study, we reared mussel spat of the species Mytilus galloprovincialis under diets below the pseudofaeces threshold (BP) and above the pseudofaeces threshold (AP). After 3 months, F and S mussels from each condition were selected to obtain 4 experimental groups: FBP, SBP, FAP and SAP. We hypothesized that the nurturing conditions during the growing period would modify the molecular basis of their growth rate differences. To decipher the molecular mechanisms underlying the growth variation, the gill transcriptomes for the four mussel groups...

Characterization of two new genes implicated in mineralization process of Pinctada margaritifera ArchiMer
Cochennec-laureau, Nathalie; Fleury, Elodie; Belliard, Corinne; Levy, Peva.
A transplant is a complex surgical procedure performed to obtain a pearl after about 12 to 18 months. One of the key steps in this procedure is the transfer of the graft (piece of mantle) from the donor oyster. A functional relationship exists between the coating nature of the pearl and the cellular structure of the pearl bag portion of the graft. We have identified two genetic markers of biomineralisation in Pinctada margaritifera, the pearl oyster: Perline and Calcine, responsible for secretion of aragonite and calcite, respectively. After isolating and sequencing a portion of each of these genes, we verified the specificity of these sequences using molecular methods (RT-PCR, in situ hybridisation) and biocomputing methods (alignments, phylogenetic...

RESCO - Réseau d'observations Conchylicoles : Rapport annuel Campagne 2014. ArchiMer
Fleury, Elodie.

Double-stranded RNA (dsRNA)-mediated genetic interference (RNAi) is a widely used reverse genetic tool for determining the loss-of-function phenotype of a gene. Here, the possible induction of an immune response by long dsRNA was tested in a marine bivalve (Crassostrea gigas), as well as the specific role of the subunit 2 of the nuclear factor kappa B inhibitor (I kappa B2). This gene is a candidate of particular interest for functional investigations in the context of oyster mass mortality events, as Cg-I kappa B2 mRNA levels exhibited significant variation depending on the amount of ostreid herpesvirus 1 (OsHV-1) DNA detected. In the present study, dsRNAs targeting Cg-I kappa B2 and green fluorescent protein genes were injected in vivo into oysters...

Fleury, Elodie.
This work contributed to the development of specific genomic tools for the Pacific oyster Crassostrea gigas, aiming to identify molecular markers associated with summer survival of this species. To do so, 29745 ESTs obtained from different sequencing programs were grouped in a unique database. 9058 unigenes have been used to produce a cDNA microarray, to compare lines selected to be Resistant (R) or Sensitive (S) to summer mortality. 34 genes were observed as differentially expressed before the mortality event. Most of these genes are associated with reproduction and oxidative stress. This underlines the implication of the cost of reproduction and the importance of oxidative stress defenses in development of an immune weakness before the mortality event....

Farmer monitoring reveals the effect of tidal height on mortality risk of oysters during a herpesvirus outbreak ArchiMer
Pernet, Fabrice; Gachelin, Sonia; Stanisière, Jean-yves; Petton, Bruno; Fleury, Elodie; Mazurié, Joseph.
The intertidal zone is characterized by a sharp vertical gradient of environmental stress, which structures species distribution and their interactions. Few studies, however, have examined the influence of tidal height on host–pathogen interactions. Here, we investigated how the tidal height influence outbreak of the Ostreid herpesvirus type 1 (OsHV-1) affecting the Pacific oyster. A volunteer network composed of 20 oyster growers monitored the survival of 28 batches of oysters during an epizootic event in Southern Brittany, France. Oysters were spat from wild collection or hatchery production. The sampling sites were spread over a 150-km2 area with a tidal height ranging from 0.98 to 2.90 m. Concomitantly, we followed survival of oyster spats in relation...

NPY signalling in the gonad of the oyster Crassostrea gigas : involvement in summer mortality? ArchiMer
Corporeau, Charlotte; Fleury, Elodie; Moal, Jeanne; Boudry, Pierre; Favrel, Pascal; Huvet, Arnaud.
In summer, oysters suffer mortality that decimates this year about 60% of the population in France. We previously demonstrated a genetic basis for summer survival thatallowed us to produce resistant (R) and susceptible (S) oyster familiesby divergent selection (Samainet al., 2007). At the physiological level, R families can survive summer mortality because they are reproductively less active than S families (Fleury et al., 2008).In order to identify the molecular basis of the summer survival,a transcriptomicapproach on R and S families was performed using microarray analysis and 34 differentially-expressed genes were identified between R and S in the gonad (Fleury et al., in prep). Among these genes, we found an EST encoding a neuropeptide Y related...

Characterization of a gonad-specific transforming growth factor-β superfamily member differentially expressed during the reproductive cycle of the oyster Crassostrea gigas ArchiMer
Fleury, Elodie; Fabioux, Caroline; Lelong, Christophe; Favrel, Pascal; Huvet, Arnaud.
To study the complex and non identified host causes of summer mortality in C. gigas, divergent selection criteria based upon summer survival have been applied to produce resistant (R) and susceptible (S) oysterfamilies(Samain et al., 2007). A previous analysis by suppression-subtractive hybridization has characterized 46 differentially-expressedcDNAsbetween R and S families(Huvet et al., 2004). The present study reports the screening, using nylon array, of the 46 oyster SSHcDNAsfrom R and S selected families. Among the few differentially expressed genes characterized between R and S samples, one encodes a Transforming-Growth-Factor-β-related (TGFβ) protein. Members of the TGFβsuperfamily are critical growth factors regulating a variety ofimportant...

Determination of risk factors for herpesvirus outbreak in oysters using a broad-scale spatial epidemiology framework ArchiMer
Pernet, Fabrice; Fuhrmann, Marine; Petton, Bruno; Mazurie, Joseph; Bouget, Jean-francois; Fleury, Elodie; Daigle, Gaetan; Gernez, Pierre.
Marine diseases have major impacts on ecosystems and economic consequences for aquaculture and fisheries. Understanding origin, spread and risk factors of disease is crucial for management, but data in the ocean are limited compared to the terrestrial environment. Here we investigated how the marine environment drives the spread of viral disease outbreak affecting The Pacific oyster worldwide by using a spatial epidemiology framework. We collected environmental and oyster health data at 46 sites spread over an area of 300 km2 along an inshore-offshore gradient during an epizootic event and conducted risk analysis. We found that disease broke out in the intertidal farming area and spread seaward. Mortalities and virus detection were observed in oysters...
