Paralytic shellfish poison accumulation yields and feeding time activity in the Pacific oyster (Crassostrea gigas) and king scallop (Pecten maximus) ArchiMer
Bougrier, Serge; Lassus, Patrick; Bardouil, Michele; Masselin, Pierre; Truquet, Philippe.
Pacific oysters and king scallops placed individually in a recirculating flume were fed for 2 weeks with a constant concentration (120 cell ml(-1)) of a toxic strain of Alexandrium minutum. Fluorescence at the outlet of each experimental unit was measured continuously, and biodeposits were recovered twice daily to evaluate feeding time activity (FTA) and rates of organic filtration (OFR), ingestion (OIR) and organic absorption (OAR). Ion-pairing high performance liquid chromatography (IP-HPLC) was performed concurrently to quantify paralytic phycotoxins both (i) individually in shellfish at the end of the contamination period and (ii) in A. minutum cultures to estimate cellular toxin concentration. These data allowed comparison of the actual tissue toxin...

Morlaix, Martine; Lassus, Patrick.
Very few attempts have been made in order to investigate the role of nutrients on cellular growth and toxin production in dinoflagellates associated with DSP (Diarrhetic Shellfish Poison). Some preliminary results are presented here, aiming at a better knowledge of inorganic nitrogen and organic phosphorus effects upon division rate and toxin content of Prorocentrum lima (PL2V spanish strain). Nitrogen have a positive effect on growth rate, up to a threshold value of 880muM.l-1. Above this threshold the rate decreases. There is no effect on toxin content since okadaic acid and DTX1 cell contents remain low (4.8 +/- 0.5 pg.cell.-1) and steady. Phosphorus effect upon division rate follows a non-linear relationship, whereas cellular toxic content is inversely...

ArchiMer
Lassus, Patrick; Martin, A; Maggi, Pierre; Berthome, Jean-paul; Langlade, Aime; Bachere, Evelyne.
Discolored waters and human diseases by shellfishes were dominant features of summer 1983 in south coast of Brittany. Vilaine Bay, the most damaged area, was successively affected by brown waters in early June, and diarrhetic intoxications by harvested mussels (July and August), just as DPS producing dinoflagellate (D. acuminata ) reached higher water or mussel guts concentrations than years before. Outspreading in the whole south coast of Brittany and possible relation with hydrological or biological parameters are analyzed.

Paralytic phycotoxin uptake by scallops (Pecten maximus) ArchiMer
Lassus, Patrick; Bardouil, Michele; Ledoux, Martial; Murail, Ingrid; Bohec, Madeleine; Truquet, Philippe; Frémy, Jean-marc; Rohmer, Véronique.
It is difficult to apply sanitary standards for mollusc contamination by paralytic shellfish poisoning to animals not consumed whole. This problem is illustrated by the 1990 embargo on a Japanese shipment of frozen scallop muscle and gonad: the sanitary threshold applied to digestive gland (in Japan) does not guarantee that muscle and especially gonad are toxin-free. Accordingly, we performed experimental contaminations of scallops (Pecten maximus) from Port-en-Bessin (Normandy, France) using a toxic Japanese strain of Alexandrium tamarense. During the contamination/decontamination experiment on different tissues (digestive gland, muscle, gonad), extracts were obtained using cold acetic acid 0.1 N to maintain the perfect integrity of the toxin profile....

Lassus, Patrick; Maggi, Pierre; Proniewski, F.; Truquet, Philippe; Nizard, G..
Time-related Dinophysis spp vertical profiles have been studyed during a 48 hours cycle in a submerged cylindric tank. A coïncidental Gymnodinium sp. (Scrippsiella trochoidea and Prorocentrum triestinum as companion species) red tide was believed to have strong effect on Dinophysis distribution in the upper layer during the tirst 24 hours cycle of the period of investigation (August 30-31, 1991). Between 24 and 48 hours, the sharp decrease of Dinophysis densities as well as the graduai decline observed in the red tide and in the vertical thermal gradient have resulted in a migration of dominant species : D. sacculus and D. cf. acuminata toward subsuperficiallevels (5 to 7 m.d) with a higher range in cells vertical movements. This behavior is very similar...

Maggi, Pierre; Gruet, Yves; Lassus, Patrick; Kalifa, Francis.

Effects of Alexandrium minutum exposure upon physiological and hematological variables of diploid and triploid oysters, Crassostrea gigas ArchiMer
Haberkorn, Hansy; Lambert, Christophe; Le Goic, Nelly; Gueguen, Marielle; Moal, Jeanne; Palacios, Elena; Lassus, Patrick; Soudant, Philippe.
The effects of an artificial bloom of the toxin-producing dinoflagellate, Alexandrium minutum, upon physiological parameters of the Pacific oyster, Crassostrea gigas, were assessed. Diploid and triploid oysters were exposed to cultured A. minutum and compared to control diploid and triploid oysters fed T. Isochrysis. Experiments were repeated twice, in April and mid-May 2007, to investigate effects of maturation stage on oyster responses to A. minutum exposure. Oyster maturation stage, Paralytic Shellfish Toxin (PST) accumulation, as well as several digestive gland and hematological variables, were assessed at the ends of the exposures. In both experiments, triploid oysters accumulated more PSTs (approximately twice) than diploid oysters. Significant...

Maggi, Pierre; Mastouri, Abderrazak; Lassus, Patrick; Soulard, Louis; Cadiou, Yvon; Le Paul, Christine.

Lassus, Patrick; Amzil, Zouher; Bardouil, Michele; Baron, Regis; Bohec, Madeleine; Bougrier, Serge; Garen, Pierre; Laabir, Mohamed; Leguay, Didier; Masselin, Pierre; Truquet, Philippe; Sechet, Veronique; Bablon, Clotilde; Berard, Jean-baptiste; Carre, Camille; Cheignon, Valerie; Lecamp, Sylvain; Mabit, Marie.

Heral, Maurice; Lassus, Patrick; Halguand, D..

Lassus, Patrick; Maggi, Pierre; Proniewski, F.; Truquet, Philippe; Bardouil, Michele.
Since 1987 Antifer oil tanker terminal (Normandy, France) was weIl known, as a preferential area for summer highest blooms of Dinophysis cf acuminata, a toxic dinoflagellate. Three oceanographic cruises were made in August 1989, before, in the course of and after the seasonal max. of this species. Results obtained from these cruises (hydrological parameters and phytoplankton) as weIl as from a daily survey at a reference station in Antifer harbour, corrobora te the prevailing role of coastal up flowing of Seine estuarine waters, pushed to the coast by a dominant S - SW wind, on intensity and duration of Dinophysis bloom. Such observations are in agreement with those of Summer 1987 at reference station.

Lassus, Patrick; Le Dean, Loic.

ArchiMer
Andresen Leitao, Maria Da Paz; Lassus, Patrick; Maggi, Pierre; Le Baut, Claire; Chauvin, Jacky; Truouet, Philippe.
Following 1978 and 1981 human poisoning by mussels in Vilaine Bay (France) a survey of potentially toxic dinoflagellates was conducted in that area in 1982. At the same time paralytic shellfish poison (PSP) analyses were made, the authors studied the dinoflagellates encountered in the intestinal guts of mussels from January to August 1982. Results failed to demonstrate an occurrence of PSP producing species, either by toxicological analysis or dinoflagellate persistency in the mussel guts.

Utilisation du filet à plancton du type "Bongo" ArchiMer
Heral, Maurice; Woehrling, Daniel; Halgand, Daniel; Lassus, Patrick.
The "Bongo" net is an American plankton net whose use is coming into general use for studies of ichthyoplankton. Its dimensions and its power of filtration make it an efficient device for researching groups with unevenly spaced micro distribution as well as rare species, especially fish eggs and larvae. It is recommended by the ICNAF (Standard Techniques for Pelagic Fish Eggs and Larvae Surveys) and will soon be recommended by the FAO. The ISTPM, for its part, has been using this net for some years in several campaigns conducted by the Thalassa, the Cryos, La Pélagia, the Roselys and the Ichthys (ISTPM 1971, 1975, ALDEBERT 1975, ARBAULT and LACROIX 1975). In 1975, we decided to use this net in some studies in coastal waters that the ISTPM is conducting in...

Application of a digital pattern recognition system to Dinophysis acuminata and D-sacculus complexes ArchiMer
Truquet, Philippe; Lassus, Patrick; Honsell, Giorgio; Le Dean, Loic.
An image discrimination technique was developed to improve specific identification of some toxic Dinophysis cells (marine dinoflagellates involved in diarrhoeic shellfish poisoning), especially the ''acuminata'' and ''sacculus'' groups, which can be present at different ratios in natural sea-water samples collected during toxic episodes. This work was performed with image analysis software SAMBA (TITN Alcatel) using preserved cells directly observed through an inverted microscope and recorded with a video camera before further processing. All morphometric parameters used for discrimination of 7 different species or morphotypes of Dinophysis were tested with discriminant analysis. This study indicates that Dinophysis sp. and D. pavillardi seem well...

Improving detoxification Efficiency of PSP-contaminated oysters (Crassostrea gigas Thunberg) ArchiMer
Lassus, Patrick; Bardouil, Michele; Baron, Regis; Berard, Jean-baptiste; Masselin, Pierre; Truquet, Philippe; Pitrat, Jp.
Shellfish production, and production of marine bivalve molluscs in particular, is the main form of marine aquaculture in the European Union, with a production estimated at 1,200,000 tons in 1999 (Food and Agricultural Organization). Moreover, aquaculture production is rapidly expanding throughout Europe and is becoming a major economic resource in developing areas such as Northern Greece, Ireland, and northwestern Spain. These shellfish, mainly mussels, oysters, and clams, are produced in a natural environment. Due to variations in their environment, these otherwise edible species may however become unsafe for human consumption. Potential problems are amplified due to the nature of shellfish themselves: as they are filter feeders, they consume large...
