Recent studies suggest that IGF-I is a crucial regulatory factor in follicular growth during early post-partum period. The aim of the present study was to determine in detail the changing profiles of metabolic and reproductive hormones in relation to ovulation of the dominant follicle (DF) of the first follicular wave post-partum in high-producing dairy cows. Plasma concentrations of related hormones in 22 multiparous Holstein cows were measured from 4 weeks pre-partum to 3 weeks post-partum, and the development of DF was observed with colour Doppler ultrasound. Thirteen cows showed ovulation by 15.2 days post-partum. Anovulatory cows showed higher GH and lower IGF-I levels than those in ovulatory cows during the peri-partum period. Each DF developed...

Association of rumen fill score and energy status during the close-up dry period with conception at first artificial insemination in dairy cows OAK
Kawashima, Chiho; Karaki, Chihiro; Munakata, Megumi; Matsui, Motozumi; Shimizu, Takashi; Miyamoto, Akio; Kida, Katsuya.
Recent studies have shown significant associations between prepartum energy status and postpartum fertility in dairy cows; therefore, the assessment of energy status by blood metabolites and metabolic hormones and suitable improvement of management during the prepartum period may enhance reproductive performance. Rumen fill score (RFS) is associated with feed intake; however, it is unknown whether RFS is also related to blood parameters. Therefore, this study investigated the relationship between RFS and energy status during the prepartum period, and their associations with conception at first artificial insemination (AI) after parturition. In 42 multiparous Holstein cows, RFS assessment and blood sampling were carried out twice a week during 3 weeks of...

The present study aimed to examine the possibility that bovine somatotropin (BST) treatment after artificial insemination (AI) may improve the fertility by the stimulation of corpus luteum (CL) function in seasonally anestrous ewes. The experiment was conducted at the Tawa Field Station, Shibecha-cho in Hokkaido, Japan, during the non-breeding season (May, 1995). Eighty-nine mature (2 to 3 years old) Suffolk and Suffolk-crossed ewes were treated with controlled internal drug release devices containing 0.3 g progesterone (CIDR-G) for 12 days. Ewes were injected intramuscularly with 500 IU PMSG the day before CIDR removal. Eighty-seven ewes were assigned to an insemination of frozen semen on a fixed-time basis (42 to 50 h) after removal of CIDR. On the 11th...

Kawashima, Chiho; Sakaguchi, Minoru; Suzuki, Takahiro; Sasamoto, Yoshihiko; Takahashi, Yoshiyuki; Matsui, Motozumi; Miyamoto, Akio.
Metabolic hormones affect ovarian function in the cow. However, the relationship between metabolic factors and ovarian function is not clear in the postpartum primiparous cow because they are still growing. The aim of the present study was to investigate in detail the time-dependent profile of the metabolic hormones, metabolites, and milk yields of ovulatory and anovulatory primiparous cows during the first follicular wave postpartum. We used 16 primiparous Holstein cows and obtained blood samples for the profiles of metabolites (glucose; non-esterified fatty acid, NEFA; ketone body; total cholesterol; and aspartate aminotransferase), metabolic hormones (growth hormone, GH; insulin-like growth factor-I, IGF-1; and insulin), and progesterone every other day...

The activity of α3-D-mannoside-β-1,2-N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was detected in bovine follicular fluid (bFF). The GnT I activity in bFF had a pH optimum of 5.8 and an absolute requirement for either Co2+, Mn2+, or Mg2+, the activity being stimulated by these cations in the above order. The apparent Km value for α1-3α1-6 mannopentaose of GnT I in bFF was 2.17 mM. The substrate specificity for the GnT I activity decreased in the following order: α1-3α1-6 mannopentaose> α1-3α1-6 mannotriose>α1-3 mannobiose. The GnT I activity in bFF from large atretic follicles was significantly higher than in that from large...

An autoregressive logistic model to predict the reciprocal effects of oviductal fluid components on in vitro spermophagy by neutrophils in cattle OAK
Kowsar, Rasoul; Keshtegar, Behrooz; Marey, Mohamed. A.; Miyamoto, Akio.
After intercourse/insemination, large numbers of sperm are deposited in the female reproductive tract (FRT), triggering a massive recruitment of neutrophils (PMNs) into the FRT, possibly to eliminate excessive sperm via phagocytosis. Some bovine oviductal fluid components (BOFCs) have been shown to regulate in vitro sperm phagocytosis (spermophagy) by PMNs. The modeling approach-based logistic regression (LR) and autoregressive logistic regression (ALR) can be used to predict the behavior of complex biological systems. We, first, compared the LR and ALR models using in vitro data to find which of them provides a better prediction of in vitro spermophagy in bovine. Then, the best model was used to identify and classify the reciprocal effects of BOFCs in...

Immunoreactivity for Gonadotropin-Releasing Hormone in Microdialyzed Perfusates of Bovine Mature Follicles In Vitro OAK
Ozawa, Takahiro; Acosta, Tomas J.; Nakamura, Tadashi; Miyamoto, Akio; 中村, 正; 宮本, 明夫.
The presence of gonadotropin-releasing hormone (GnRH)-like protein has been previously demonstrated in the bovine follicle. This GnRH-like protein was purified and concluded to be histone H2A. However, neither GnRH peptide nor specific GnRH-immunoreactivity (GnRH-IR) has been demonstrated in the bovine follicle so far. Thus, this study focused on the detection of specific GnRH-IR using the second-antibody enzyme immunoassay in isolated bovine mature follicles, and on an examination of the direct effect of luteinizing hormone (LH), endothelin-1 (ET-1), and cytokines on the GnRH-IR using an in vitro microdialysis system (MDS). We further examined a cross-reactivity of the GnRH antibody with bovine histone H2A. GnRH-IR was detected in microdialyzed perfusate...

Sperm Binding to Oviduct Epithelial Cells Enhances TGFB1 and IL10 Expressions in Epithelial Cells as Well as Neutrophils In Vitro: Prostaglandin E-2 As a Main Regulator of Anti-Inflammatory Response in the Bovine Oviduct OAK
Yousef, Mohamed Samy; Marey, Mohamed Ali; Hambruch, Nina; Hayakawa, Hiroyuki; Shimizu, Takashi; Hussien, Hassan Ali; Abdel-Razek, Abdel-Razek Khalifa; Pfarrer, Christiane; Miyamoto, Akio.
Sperm are allogenic to the female genital tract; however, oviducts provide optimal conditions for survival and capacitation of these non-self cells until fertilization. Recently, we showed that oviduct-conditioned media and prostaglandin E-2 (PGE(2)) suppress sperm phagocytosis by polymorphonuclear neutrophils (PMNs) under physiological conditions. We hypothesized that sperm binding to bovine oviduct epithelial cells (BOECs) could change the local innate immunity via PGE2. As the first step to obtain basic information, sub-confluent BOEC monolayers were co-cultured with swim-up sperm for 2 h. BOECs with viable bound sperm were cultured for an additional 3, 6, 12, or 24 h. Then, we confirmed the impact of the sperm-BOEC binding on both BOECs and PMN gene...

Effect of intraluteal injection of endothelin type A receptor antagonist on PGF2alpha-induced luteolysis in the cow. OAK
Watanabe, Sho; Shirasuna, Koumei; Matsui, Motozumi; Yamamoto, Dai; Berisha, Bajram; Schams, Dieter; Miyamoto, Akio; 白砂, 孔明; 松井, 基純; 宮本, 明夫.
Endothelin-1 (ET-1) is a luteolytic mediator in the bovine corpus luteum (CL), and its action appears to be via endothelin type A receptor (ETR-A). Thus, the aim of the present study was to determine the effect of ETR-A antagonist on PGF(2)alpha-induced luteolysis in the cow. Cows on days 1012 of the estrous cycle were subjected to five intraluteal injections of the ETR-A antagonist LU 135252 in saline or only saline at -0.5, 2, 4, 6, and 8 h after PGF(2)alpha administration (=0 h). Serial luteal biopsies were conducted to determine the expression of mRNA in the luteal tissue. There were no significant differences in the decrease in plasma progesterone (P) concentrations and the mRNA expressions of steroidogenic acute regulatory protein and 3...
