Ecological approach of macro-fouling prevention and control in cooling systems OMA
Denayer, B.; De Ranter, K.; Van Poele, J.; Ollevier, F.P..
Biofouling is a very frequent and serious problem in the cooling systems of electrical power stations. An investigation is being carried out at the Doel nuclear power station to discover ways of combating micro-fouling. Micro-fouling is combated preventively and curatively using a combination of chlorine, biofilm wetting agent and dispersing agent. This treatment removes the slime layer, which can act as a primary attachment point for macro-organisms.In the study of the macro-fouling in the cooling circuits, a monitoring method was developed which uses artificial substrates to provide direct data on the colonization and growth of these organisms. On the basis of the data obtained, the treatment frequency for the prevention or combating of biofouling was...

Mode of infection and spread of Vibrio anguillarum in turbot Scophthalmus maximus larvae after oral challenge through life feed OMA
Grisez, L.; Chair, M.; Sorgeloos, P.; Ollevier, F.P..
The infection route of the marine fish pathogen Vibrio anguillarum was studied after oral challenge of the juvenile turbot Scophthalmus maximus L. through a life feed. Artemia nauplii were incubated in a suspension of V. anguillarum cells, and subsequently fed twice to the fish. All challenged fish died within 4d after the first challenge, while no mortality occurred in the non-challenged controls. The results of an immunohistochemical examination of the sectioned fish samples clearly demonstrated that V. anguillarum cells were ingested by the Artemia and that the latter were ingested by the fish. Bacteria were released from the...

The use of API 20E for the identification of Vibrio anguillarum and V. ordalii OMA
Grisez, L.; Ceusters, R.; Ollevier, F.P..
Sixty-eight strains of Vibrio anguillarum, five of V. ordalii and the type strains of V. alginolyticus, V. carchariae, V. damsela and V. parahaemolyticus were compared using the API 20E gallery. Within the V. anguillarum strains, distinct groups could be separated mainly on the basis of their reaction on indole production and the fermentation of amygdalin and arabinose. Vibrio ordalii, the former V. anguillarum biotope 2, could easily be separated from V. anguillarum and from the other fish pathogenic Vibrio spp.

Hatching, survival, activity and penetration efficiency of second-stage larvae of Anguillicola crassus (Nematoda) OMA
Thomas, K.M.; Ollevier, F.P..
Hatching of eggs of Anguillicola crassus was studied at temperatures between 5 and 30°C. Hatching occurred between 10 and 30°C and the incubation time was inversely related to temperature. Survival, spontaneous and excitatory rate of activity of 2nd-stage larvae of A. crassus were also affected by temperature. The percentage of larvae eaten by Paracyclops fimbriatus was not always influenced by the activity of the larvae. Penetration efficiency into the haemocoel of the intermediate host was highest for freshly hatched larvae and declined gradually with age. Completely non-active larvae were still able to penetrate, although penetration efficiency was lower.

Comparative serology of the marine fish pathogen Vibrio anguillarum OMA
Grisez, L.; Ollevier, F.P..
The different serotyping systems, based on thermostable O antigens, reported for Vibrio anguillarum and V. ordalii were compared by quantitative agglutination, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and subsequent silver staining or Western blotting (immunoblotting) of purified lipopolysaccharide (LPS), using polyclonal rabbit antisera. The results demonstrate that 16 different serotypes within V. anguillarum (designated 01 to 016) can be distinguished. Each of these serotypes is characterized by a distinct polysaccharide banding pattern, as revealed by silver-stained gels of purified LPS. The comparative analysis allowed a complete alignment of the different stereotypes for the...

Vibrios associated with routine productions of Brachionus plicatilis OMA
Verdonck, L.; Grisez, L.; Sweetman, E.; Minkoff, G.; Sorgeloos, P.; Ollevier, F.P.; Swings, J..
Surveys for bacteriological analysis were carried out in 1990-1991 in two different marine fish hatcheries located in Greece and northern Spain. Samples were taken from 12 different routine productions of Brachionus plicatilis before and during enrichment. Isolates were characterized by whole cell fatty acid methyl ester analysis and by BIOLOG metabolic fingerprinting. Vibrio anguillarum and/or Vibrio alginolyticus were dominant Vibrio species in the rotifer samples taken after enrichment and rinsing, before being delivered to the fish larvae. None of the Vibrio anguillarum strains belonged to the pathogenic serotypes for juvenile and adult fish.

Endoparasitic helminths of the whitespotted rabbitfish (Siganus sutor (Valenciennes, 1835)) of the Kenyan coast: distribution within the host population and microhabitat use OMA
Geets, A.; Ollevier, F.P..
The parasitic fauna of the alimentary tract of adult whitespotted rabbitfish, Siganus sutor, sampled in December 1990 at the Kenyan coast, was investigated. Five endoparasites were found: the digenean trematodes Opisthogonoporoides hanumanthai, Gyliauchen papillatus and Hexangium sigani, the acanthocephalan Sclerocollum rubrimaris and the nematode Procammalanus elatensis. No uninfected fish, nor single species infections occurred. Parasite population data showed very high prevalences for all endoparasites, ranging from 68.18% to 100%. G. papillatus occurred with the highest mean intensity, 201.68 ± 12.54 parasites per infected fish. The effect of Artemia -supplementation of dry foods on growth and survival of Clarias gariepinus (Burchell 1822) larvae OMA
Van Damme, P.; Pacolet, W.; Sulem Yong, S.; Fremaut, D.; Ollevier, F.P..
Two experiments were performed in order to determine the effect of supplementation of dry foods with live Artemia nauplii on the growth, survival, rate of cannibalism, and length-weight relationships of Clarias gariepinus larvae. It was found that Artemia -supplementation resulted in a significantly higher growth rate. The "condition", expressed as a length-weight relationship, was likewise altered after supplementation. The survival and rate of cannibalism were not influenced by supplementation. Artemia -supplementation of dry foods was compared with other approaches which are based on the concept of the 'adaptation weight'.

Protein digestion in juvenile turbot (Scophthalmus maximus) and effects of dietary administration of Vibrio proteolyticus OMA
De Schrijver, R.; Ollevier, F.P..
The trial was made to study the protein digestion in juvenile turbot during passage of digesta from stomach to rectum. Moreover, diet supplementation with the potential probiotic bacteria, Vibrio proteolyticus, was evaluated with regard to protein digestion. For a 3-week period, fish (25-30 g) were fed by oral intubation and received either a liquid mixture consisting of 40% nonpurified control diet and 60% water or this mixture supplemented with 10 10 viable V. proteolyticus ml -1. Daily dry matter was 1.5% body weight. As digesta progressed from the stomach to the foregut, hindgut and rectum, the increase in apparent nitrogen digestibility was accompanied by higher...

Search for beneficial bacterial strains for turbot (Scophthalmus maximus L.) larviculture OMA
Huys, L.; Dhert, P.; Robles, R.; Ollevier, F.P.; Sorgeloos, P.; Swings, J..
The aerobic bacterial flora in the gut of turbot larvae and their influence on larval survival was examined. Two turbot experiments were run with six replicates each time. Large variation, from 0% up to 44%, was observed in the survival percentage of turbot larvae. There was no correlation between the number of bacteria present in the gut of turbot larvae and the larval survival rate. During both experiments, all replicates followed nearly the same rate of bacterial development in the gut of turbot larvae going from circa 102 CFU larva-1 just before first feeding at day 3 post hatch to 105 CFU larva-1 at day 9 post hatch. In total, 127 bacterial isolates from 12...

The life cycle of Anguillicola crassus OMA
De Charleroy, D.; Grisez, L.; Thomas, K.; Belpaire, C.; Ollevier, F.P..
For some years now the parasitic swim bladder nematode Anguillicola crassus of the European eel Anguilla anguilla L., has been reported from several European countries. The entire life history of this parasite has recently been elucidated in our laboratory. Young larvae leave the swim bladder of the host via the pneumatic duct and reach the water through the digestive tract. They are ingested by small copepods (Cyclopoida), which act as intermediate hosts. Larvae remain in the hemocoel until the copepods are eaten by the final host, the European eel. Larvae penetrate through the intestinal wall and reach the swim bladder where they develop into adults. When infected copepods are eaten by other small fish, such as carp...

Extended serotyping scheme for Vibrio anguillarum with the definition and characterization of seven provisional O-serogroups OMA
Pedersen, K.; Grisez, L.; van Houdt, R.; Tiainen, T.; Ollevier, F.P.; Larsen, J.L..
The present paper summarizes the serotyping scheme of the fish pathogenic bacterium Vibrio anguillarum and defines seven additional O-serogroups. Strains, collected in our laboratories that were nontypable with antisera against the previously defined 16 O-serotypes, were used for generating new antisera and were characterized further by means of LPS profiles, Western blots, and serological reactions. On the basis of the results, it is suggested that the seven new O-serogroups are to be included in the existing serotyping system as serotypes O17-O23. However, the existence of further V. anguillarum strains that were not typable with any of the 23 O-antisera suggested the existence of additional...

Experimentally induced infections of European eel Anguilla anguilla with Anguillicola crassus (Nematoda, Dracunculoidea) and subsequent migration of larvae OMA
Haenen, O.L.M.; Grisez, L.; De Charleroy, D.; Belpaire, C.; Ollevier, F.P..
Migration patterns of third-stage Anguillicola crassus larvae, and pathogenesis of the lesions induced by third-stage larvae, was investigated in European eel Anguilla anguilla L. Young elvers (1g) were fed infected Paracyclops fimbriatus (Copepoda). Eel samples were collected and examined histologically at varying intervals during 6 mo post-infection period. Third-stage larvae (L-III) migrated directly through the intestinal wall and body cavity to the swimbladder within 17h post-infection. L-IV larvae were detected 3 mo post-infection, and immature adults were detected within 4 mo post-infection. The parasites occasionally showed aberrant migration paths. Pathological effects caused by the parasite...

Use of computer assisted sperm analysis (CASA) for monitoring the effects of pollution on sperm quality of fish: application to the effects of heavy metals OMA
Kime, D.E.; Ebrahimi, M.; Nysten, K.; Roelants, I.; Rurangwa, E.; Moore, H.D.M.; Ollevier, F.P..
Computer assisted sperm analysis (CASA) has been used to analyse the effects of cadmium and zinc on the motility of fish sperm treated either for 24h after partial dilution in an extender, or during the final dilution step on the microscope stage during video recording. The progressive motility of catfish sperm, which was not affected by storage for 24h, was decreased after exposure to 100 ppm cadmium or 2000 ppm zinc in extender for 24h. Only 1000 ppm cadmium or 2000 ppm zinc had any effect on motility when added to sperm at the final dilution stage. These results suggest that concentrations of cadmium which have been found to be present in fish gonads as a result of bioaccumulation from the water or diet may decrease the motility of sperm during...
