Background Asian soybean rust (SBR) caused by Phakopsora pachyrhizi Syd. & Syd., is one of the main diseases affecting soybean and has been reported as one of the most economically important fungal pathogens worldwide. Knowledge of the genetic diversity of this fungus should be considered when developing resistance breeding strategies. We aimed to analyze the genetic diversity of P. pachyrhizi combining simple sampling with a powerful and reproducible molecular technique. Results We employed Amplified Fragment Length Polymorphism (AFLP) technique for the amplification of P. pachyrhizi DNA extracted from naturally SBR-infected plants from 23 production fields. From a total of 1919 markers obtained, 77% were polymorphic. The high percentage of...

ABSTRACT Sugarcane orange rust caused by Puccinia kuehnii has recently become an important disease in sugarcane crops and its spread is causing great concern to growers. In this study, we analyzed spores from symptomatic orange rust sugarcane leaves collected in multiple locations in Cuba in a 4-year-period in order to characterize morphological traits of P. kuehnii, establish an adequate molecular technique to characterize it, and determine its infection court in sugarcane. Orange rust caused by P. kuehnii was confirmed by polymerase chain reaction (PCR) and morphological characterization. AFLP markers detected high diversity in P. kuenhnii samples. Sequencing of rDNA regions, as expected, did not reveal differences and SSR markers designed for P....

An overview of sugarcane brown rust in Cuba Scientia Agricola
La O,María; Perera,María Francisca; Bertani,Romina Priscila; Acevedo,Ricardo; Arias,Marta Eugenia; Casas,Mario Alberto; Pérez,Juana; Puchades,Yaquelín; Rodríguez,Eida; Alfonso,Isabel; Castagnaro,Atilio Pedro.
ABSTRACT: Multiple pathogens affect sugarcane, among them Puccinia melanocephala, the causal agent of brown rust. This disease was first reported in Cuba in 1979 when it was responsible for a severe attack on the main sugarcane variety B4362. The aims of the present study were to give an overview of sugarcane brown rust in Cuba and show the current disease situation in the country. A retrospective analysis regard to sugarcane cultivar composition resistant to brown rust in Cuba was carried out. In addition 154 genotypes, including the most used progenitors in the breeding program and commercial varieties were evaluated under natural infection conditions. The identity of P. melanocephala was verified by Polymerase Chain Reaction (PCR) and by sequencing the...
