A Leptospirose é uma doença bacteriana de caráter zoonótico que afeta os animais domésticos, silvestres e o homem. Levantamentos sorológicos têm demonstrado o envolvimento de diferentes espécies sinantrópicas e silvestres na epidemiologia da doença. Com o objetivo de conhecer melhor a epidemiologia da Leptospirose dentro da Fundação Parque Zoológico de São Paulo, foi realizado um estudo sorológico nos animais silvestres mantidos em cativeiro, no período de 1996 a 1999. It was performed the comparison of the intensity and duration of agglutinating and neutralizing antibodies to serovar Hardjo in swines vaccinated with two commercial anti-leptospira bacterins. Thin layer microcolony culture associated with PCR for early identification of Mycobacterium bovis BJM

The initial growth of mycobacteria from 49 samples of cattle and buffalo organs collected in commercial slaughterhouses was compared between modified Middlebrook 7H11 thin layer microcolony culture and Stonebrink medium used in the isolation of Mycobacterium bovis. Aliquots were decontaminated by Petroff's method, processed and cultured in both media. The identity of the acid-fast bacilli stained by Ziehl-Neelsen was confirmed by PCR. Optical microscopy showed that results of the early observation of Mycobacterium bovis colonies in thin layer culture were similar to those obtained in macroscopic observation of the colonies in Stonebrink medium. Leptospires detection in kidney, liver and uterus of cows slaughtered in Paraná State, Brazil BJM

The objective of this trial was to detect leptospires in fragments of kidney, liver and uterus of 96 cows with unknown sanitary status, randomly chosen at slaughter in Paraná, Brazil. All 96 urine samples were submitted to direct examination using dark field microscopy. Positive samples in the direct examination and all kidney, liver and uterus fragments were cultured in EMJH modified medium and Tween 80/40/LH. Cross neutralizing antibodies in hamsters vaccinated with leptospiral bacterins produced with three serovars of serogroup Sejroe BJM

Three leptospiral bacterins, produced with different serovars of Serogroup Sejroe, namely the hardjo (bacterin A), wolffi (bacterin B) and guaricura (bacterin C), were evaluated in male hamsters (Mesocricetus auratus) by comparing the agglutinating and neutralizing antibodies titers using microscopic agglutination (MAT) and in vitro growth inhibition (GIT) tests. EMJH medium with 5-fluorouracil and nalidixic acid associated with serial dilution technique used to recover Leptospira spp from experimentally contaminated bovine semen BJM

Bovine semen experimentally contaminated with Leptospira santarosai serovar Guaricura was submitted to the modified EMJH medium with 5-fluorouracil (300mg/L) and nalidixic acid (20mg/L), named as "selective medium" and using the serial dilution technique, in order to evaluate the percentage of recovery of the added microorganism. The selective EMJH medium was found with higher percentage of recovery of leptospiras and minor losses of samples due to contamination with opportunistic microorganisms than the non-selective EMJH medium: 151/376 (40.0%) of positive growth; and 38/376 (10.0%) contamination and 58/376 (15%) and 129/376 (34.0%), respectively. Evaluation of the counterimmunoelectrophoresis reaction as a serodiagnosis test for swine leptospirosis BJM

The Counterimmunoelectrophoresis reaction (CIE) was tested as a genus specific test for swine leptospirosis diagnosis using three soluble extracts obtained from Leptospira sp, serovars pomona, icterohaemorrhagiae and patoc. The extracts were treated with hot Triton X-100 and applied to serum samples of animals divided in three groups: Group 1, 10 swines experimentally infected with the Pomona strain; Group 2, 50 naturally infected swines and Group 3, 10 swines control. Isolation of leptospira Serovars Canicola and Copenhageni from cattle urine in the state of Paraná, Brazil BJM

In 2001, 698 urine samples were randomly collected from cattle at a slaughterhouse in the State of Paraná, Brazil. Direct examination using dark field microscopy was carried out immediately after collection. Five putative positive samples were cultured in modified EMJH medium, yielding two positive cultures (LO-14 and LO-10). Typing with monoclonal antibodies revealed that the two isolates were similar to Canicola (LO-14) and Copenhageni (LO-10). Microscopic agglutination test results show that Hardjo is the most common serovar in cattle in Brazil. Rats and dogs are the common maintenance hosts of serovars Copenhageni and Canicola. With the aim of isolating Leptospira spp., blood serum, kidney, liver and genital tract of 137 female swine (40 sows and 97 gilts) and also urine samples from 22 sows were collected in a slaughterhouse in the State of São Paulo, from April 2003 to August 2004. We investigated the existence of cross-protection between two anti-leptospirosis monovalent experimental bacterins produced with two strains of Leptospira serogroup Pomona: Fromm strain of serovar Kennewicky, isolated from pigs in the United States, and strain GR6 of serovar Pomona isolated from pigs in Brazil. Both were added of aluminum hydroxide as an adjuvant. Inactivation of leptospires in pools of semen from three Holstein Friesian bulls, collected in an artificial vagina, was investigated. Spermatic concentration was adjusted in egg yolk citrate extender, submitted to the following treatments: A (control; without antibiotics); B (penicillin, 1,000 UI/mL - streptomycin, 1,000 µg/mL); C (amoxicillin, 1,000 µg/mL); D (ceptiofur sodium, 1,000 µg/mL); E (amoxicillin 1,000 µg/mL - ceptiofur sodium 1,000 µg/mL). Sorovares de leptospiras predominantes em exames sorológicos de bubalinos, ovinos, caprinos, eqüinos, suínos e cães de diversos estados brasileiros Ciência Rural

Em estudo retrospectivo abrangendo os anos de 1984 a 1997, foram realizados 15.558 exames sorológicos para leptospirose (SAM, com coleção de 24 sorovares), que incluíram: 284 ovinos, 879 bubalinos, 983 cães, 1.941 caprinos, 2.903 eqüinos e 8.568 suínos, distribuídos percentualmente por estado da seguinte forma: ovinos - SP (100%); bubalinos - SP (100%); cães - SP (80,7%), RS (0,10%), SC (0,10%) e PI (19,0%); caprinos - SP (33,1%), PB (63,7%) e CE (3,2%); eqüinos - SP (79,3%), RS (9,98%), SC (0,62%), PR (2,5%), RJ (0,17%), MG (1,96%), MT (3,99%), PB (1,3%) e PI (0,03%); suínos - SP (61,91%), RS (0,3%), SC (5,95%), PR (3,67%), RJ (0,88%), MG (24,38%), GO (1,12%), SE (0,2%), PE (0,90%), CE (0,34%) e MA (0,1%). Foram avaliados 304 cães de ambiente rural e urbano do município de Monte Negro, Rondônia, através do Antígeno Acidificado Tamponado (AAT), Soroaglutinação Lenta em Tubos (SAL) e 2-Mercaptoetanol (2-ME) para a pesquisa de anticorpos anti-Brucella abortus e da Imunodifusão em gel de ágar (IDGA) e Imunodifusão em gel de ágar com soro tratado com 2-Mercaptoetanol (IDGA-ME) para Brucella canis. Foram consideradas positivas as amostras reagentes nas provas confirmatórias do 2-ME e IDGA-ME. Verificaram-se 56 (18,4%) animais reagentes ao AAT e 12 (4,0%) reagentes a SAL. Foram estabelecidos critérios de interpretação do teste de tuberculinização aplicado ao diagnóstico da tuberculose em grupos de caprinos experimentalmente sensibilizados. Dos 30 animais utilizados, dez foram sensibilizados com Mycobacterium avium - amostra D4 (grupo A) e dez com Mycobacterium bovis - amostra AN5 (grupo B). Dez caprinos não foram sensibilizados e constituíram o grupo controle (grupo C).
