Identificación filogenética de Candidatus Liberibacter solanacearum y su efecto en la calidad fisiológica de la semilla de chile jalapeño. Colegio de Postgraduados
Olivares Mercado, Perla Xochitl.

Cre-loxP recombination vectors for promoter studies Electron. J. Biotechnol.
Pedersen,Nina; Tuxen Poulsen,Thomas; Skovgaard Poulsen,Hans.
For promoter studies the cloning, subcloning and transfer to different plasmid vectors usually requires use of restriction enzymes and ligation reactions. One obstacle is the nucleotide polymorphisms of eukaryotic genomic DNA, which has the consequence that a sequence often differs from published sequences. Therefore sequencing, rigorous restriction enzyme analysis or introduction of suitable sites has to be performed prior to cloning and subcloning. In addition, conventional methods using restriction enzymes, insert purifications and ligations is expensive and labour demanding. We have developed a fast, efficient and inexpensive Cre recombinase-loxP based method, which allows cloning of promoter regions and subcloning of these into a variety of vectors in...

Vaccines for finfish aquaculture: What do we need to know to make them work? Electron. J. Biotechnol.
Dixon,Brian.
Aquaculture still faces serious economic impacts due to the loss of animals to disease. A conservative estimate of 5% losses due to disease means that the finfish aquaculture industry loses over $1 billion annually on a global scale. One proven way to prevent costly disease outbreaks is to vaccinate fish against common or known pathogens. Current vaccination schemes still result in losses, however, and this may be due in part to vaccine design. Vaccines are currently designed using state of the art knowledge of immune responses, which is based primarily on mammalian studies. Just how applicable is this information to fish immunity and vaccine design, however? This review discusses what is currently known for teleost fish about two key processes that drive...

LaNe RAGE: a new tool for genomic DNA flanking sequence determination Electron. J. Biotechnol.
Park,Daniel J.
The determination of genomic DNA sequence flanking a known region is often problematic. Existing technologies depend on multiple, efficient enzyme-catalysed preparative processing steps and/or rely on relatively inefficient ‘one-sided' PCR mechanisms. I demonstrate the application of a simple ‘two-sided' PCR-based approach, lariat-dependent nested PCR for rapid amplification of genomic DNA ends (LaNe RAGE), applied to the mouse GAPDH and PGK1 gene flanking sequences. This demonstration offers great promise in applications such as genome walking, transposon mutagenesis mapping and DNA fingerprinting

PCR assembly of synthetic human erythropoietin gene Electron. J. Biotechnol.
Bustami,Yazmin; Yahya,Ahmad Ramli Mohd; Muhammad,Tengku Sifzizul Tengku; Shu-Chien,Alexander Chong; Abdullah,Amirul Al-Ashraf; Noor,Mohd Azizan Mohd; Arip,Yahya Mat.
Human erythropoietin (huEPO) is a glycoprotein with important physiological functions, such as erythropoiesis, angiogenesis, and wound healing. A therapeutic protein, huEPO is commonly used to treat patients suffering from renal and non-renal anemia. Recombinant human erythropoietin (rhuEPO) and endogenous huEPO are similar with respect to their biological and chemical properties. In this study, we describe the construction of synthetic huEPO gene to produce rhuEPO. The synthetic huEPO gene was constructed by overlapping oligonucleotides assembly and amplified by polymerase chain reaction (PCR). Twenty oligonucleotide sets, covering the huEPO gene sequence and two newly introduced restriction enzyme sites, were pulled together and amplified using Pfu DNA...

A legume genomics resource: The Chickpea Root Expressed Sequence Tag Database Electron. J. Biotechnol.
Jayashree,B; Buhariwalla,Hutokshi K; Shinde,Sanjeev; Crouch,Jonathan H.
Chickpea, a lesser-studied grain legume, is being investigated due to its taxonomic proximity with the model legume genome Medicago truncatula and its ability to endure and grow in relatively low soil water contents making it a model legume crop for the study of agronomic response to drought stress. Public databases currently contain very few sequences from chickpea associated with expression in root tissues. However, root traits are likely to be one of the most important components of drought tolerance in chickpea. Thus, we have generated a set of over 2800 chickpea expressed sequence tags (ESTs) from a library constructed after subtractive suppressive hybridization (SSH) of root tissue from two closely related chickpea genotypes possessing different...

Attitudes towards genomic: Research in four Latin American countries Electron. J. Biotechnol.
Rodríguez Yunta,Eduardo; Valdebenito Herrera,Carolina; Misseroni,Adelio; Fernández Milla,Lautaro; Outomuro,Delia; Schiattino Lemus,Irene; Ferrer Lues,Marcela; Lolas Stepke,Fernando.
The present reflection refers to data obtained about the social representations of genomic research and its applications through interviews with legislators and lawyers, biomedical researchers and civilians and the review of scientific and legal literature in four Latin American countries: Argentine, Chile, Mexico and Peru. Several issues are addressed: little access to prevention and therapeutic methods, lack of equity in health benefits, commercialization of gene sequences through patents which leads to commercial exploitation of underdeveloped countries, the possibility of physical or psychological damage or genetic discrimination, the possibility of genetic modifications or abortion for eugenic reasons, the necessity of safeguarding confidentiality,...

Taura syndrome of penaeid shrimp: cloning of viral genome fragments and development of specific gene probes ArchiMer
Mari, Jocelyne; Bonami, Jean-robert; Lightner, Donald V.
The ssRNA genome extracted from purified Taura Syndrome Virus (TSV) was transcribed into double-stranded, blunt-ended cDNA and was used to construct cDNA libraries either in pUC 18 or in pBluescript II KS-vectors. Twelve recombinant plasmids chosen after screening of the libraries were subjected to restriction enzyme digestions for determination of size inserts and restriction maps. Two of them, pP15 and pQ1, were selected for probe construction. The inserts, 1500 and 1300 base pairs (bp) respectively, were DIG-11dUTP-labelled and the corresponding probes were named P15 and Q1. On northern blots and dot blots, using different denaturation methods, the 2 probes hybridized specifically with extracted RNA-TSV genome, TSV and infected TS shrimp homogenates. No...

Diversite de populations microbiennes thermophiles d'une cheminée hydrothermale océanique profonde : cultures d'enrichissement en bioréacteur et isolement d'espèces nouvelles ArchiMer
Postec, Anne.
To explore the microbial diversity at deep sea vents, molecular techniques based on the analysis of the gene coding the 16S ribosomal RNA permitted to highlight a large archaeal and bacterial diversity. Nevertheless, the diversity of the hydrothermal microorganisms cultivated to date remains much lower than the diversity described with molecular tools. Innovative methods should therefore be used in order to cultivate new microorganisms, and also determine their metabolic properties and their potential role in the ecosystem. That was the purpose of this study: a bioreactor gas-lift was used to perform enrichment cultures in continuous, during 50 days, in thermophilic (60°C) and hyperthermophilic (90°C) conditions. A hydrothermal chimney sample (Rainbow...

Obtaiment of Pudu (Pudu pudu) deer Embryos by the Somatic Nuclear Transfer Technique International Journal of Morphology
Venegas,Felipe; Guillomot,Michel; Vignon,Xavier; Servely,Jean-Luc; Audouard,Chistophe; Montiel,Enrique; Le Bourhis,Daniel; Perón,Sandrine; Soto,Pilar; Rojas,Mariana.
The objective of this project is to produce pudu embryos, obtained by means of fibroblast transfer taken from the ear of one animal, to the oocytes of a domestic ruminant such as cattle. In further work, the pudu embryos will be transferred to the uterus of synchronized recipient females of another species. Biopsies 1mm in size were obtained from the outer rim of the ear of two male pudu deer belonging to the Buin Zoological park, Santiago, Chile. The cell lines have been established and kept according to protocols used in cattle. The oocytes are obtained through a puncture in the oocyte-cumulus complex (OCC) from cow ovaries recovered from the butcher. Each oocyte is enucleated and fused with an isolated fibroblast which is inserted beneath the pellucid...

Arthropod venom Hyaluronidases: biochemical properties and potential applications in medicine and biotechnology J. Venom. Anim. Toxins incl. Trop. Dis.
Bordon,Karla C F; Wiezel,Gisele A.; Amorim,Fernanda G.; Arantes,Eliane C..
AbstractHyaluronidases are enzymes that mainly degrade hyaluronan, the major glycosaminoglycan of the interstitial matrix. They are involved in several pathological and physiological activities including fertilization, wound healing, embryogenesis, angiogenesis, diffusion of toxins and drugs, metastasis, pneumonia, sepsis, bacteremia, meningitis, inflammation and allergy, among others. Hyaluronidases are widely distributed in nature and the enzymes from mammalian spermatozoa, lysosomes and animal venoms belong to the subclass EC 3.2.1.35. To date, only five three-dimensional structures for arthropod venom hyaluronidases (Apis mellifera and Vespula vulgaris) were determined. Additionally, there are four molecular models for hyaluronidases fromMesobuthus...

Wendling,Ivar; Sturion,José Alfredo; Stuepp,Carlos André; Reis,Cristiane Aparecida Fioravante; Ramalho,Magno Antonio Patto; Resende,Marcos Deon Vilela de.
Abstract: The objective of this work was to evaluate the feasibility of early selection of open-pollinated yerba mate (Ilex paraguariensis) progenies, and to classify the best parents and candidates for clones. The germplasm is composed of 140 progenies collected from areas within the natural distribution of the species in Southern Brazil and a commercial genotype (control). The experiment was established in Ivaí, in the state of Paraná, Brazil, in March 1997, in randomized complete blocks, with ten replicates, and linear plots of six plants. The commercial mass (kg per plant) of leaves and branches smaller than 7 mm was harvested at 2.5, 4.5, 6.5, and 18.7 years of age. The statistical evaluation was performed using the mixed model procedure with...
