TOXIGENICITY CHARACTERIZATION OF Clostridium perfringens FROM BOVINE ISOLATES J. Venom. Anim. Toxins
BALDASSI,L.; BARBOSA,M. L.; BACH,E. E.; IARIA,S. T..
Clinical samples from 71 bovine from different Brazilian states were processed for the analysis of anaerobe organisms with emphasis on the isolation and characterization of Clostridium spp. From these, eighty-nine Clostridium perfringens strains were recovered: 32 from liver, 19 from intestinal contents, 14 from kidney, 6 from rumen, 5 from nervous system, 4 from bone marrow, 2 from udder tract, blood, spleen and lung, and one from muscle. Four reference Clostridium perfringens types A, B, C, and D were used as controls in this study. All isolates were cultivated in appropriate media, and after centrifugation the supernatant and sediment were separated. From pure supernatant post exopolysaccharide (EPS) extraction, mouse toxigenicity tests were performed,...

Esterase electrophoresis of Clostridium perfringens bovine strains J. Venom. Anim. Toxins incl. Trop. Dis.
Baldassi,L.; Barbosa,M.L.; Bach,E.E.; Iaria,S.T..
Eighty-nine of 144 isolates of Clostridium perfringens obtained from 187 samples of 71 bovine in several Brazilian states were submitted to esterase electrophoresis for typing. Mobilites electrophoresis, as parameter, were settled down by isolates from ATCC pattern of types A, B, C, and D. Of the 89 isolates, 43 (48.3%) were characterized as electrophoretic type A, 20 (22.5%) as D, 18 (20.2%) as C, and 3 (3.4%) as B. Five (5.6%) isolates did not identity with any type. Similarly, esterase electrophoresis enabled the typing of 94.4 % of the isolates, demonstrating to be an appropriate method for animal sample analyses.

Characterization of Clostridium perfringens isolated from mammals and birds from Guwahati city, India J. Venom. Anim. Toxins incl. Trop. Dis.
Rahman,Mafruza S; Sharma,RK; Borah,P; Chakraborty,A; Devi,Mandakini RK; Longjam,N.
Of the 102 samples collected from mammals and birds, both domestic and captive wild, 48 were found to be positive for Clostridium perfringens. Most of the mammal isolates (84.38%) appeared to have been collected from clinically affected animals, while 33.33% of the bird samples were from clinically affected and 21.43% from apparently healthy birds infected with C. perfringens. Isolates revealed high sensitivity to ciprofloxacin, enrofloxacin and norfloxacin. Among the isolated C. perfringens, 30 (62.50%) showed DNase production. Hemolytic activity was recorded in 14 (24.16%) of the isolates and 28 (58.33%) showed phospholipase C production. All the phospholipase C positive isolates revealed the presence of cpa gene encoding alpha (α) toxin. Of the 102...

Molecular detection and characterization of cpb2 gene in Clostridium perfringens isolates from healthy and diseased chickens J. Venom. Anim. Toxins incl. Trop. Dis.
Tolooe,A; Shojadoost,B; Peighambari,SM.
Clostridium perfringens is an important pathogen in both human and veterinary medicine. Necrotic enteritis (NE) is the most clinically dramatic bacterial enteric disease of poultry induced by C. perfringens. The pathogenicity of this bacterium is associated with the production of extracellular toxins produced by some of its strains, such as beta2 toxin. The exact role of beta2 toxin in NE pathogenesis is still controversial. In the present study, C. perfringens isolates from healthy and diseased poultry flocks from different parts of Iran were analyzed by PCR assay to determine the presence of all variants of the beta2 toxin gene (cpb2). The products of two positive cpb2 PCR reactions were sequenced, compared to each other and to the cpb2 sequences...

Virulence exaltation of Clostridium perfringens strains from bovines J. Venom. Anim. Toxins incl. Trop. Dis.
Baldassi,L.; Barbosa,M.L.; Bach,E.E.; Iaria,S.T..
Ten out of eighty-nine strains biochemically identified as Clostridium perfringens, isolated from bovine organs, were selected by their different results showed in toxigenicity test on mice. Those and the standard strains, ATCC types A, B, C, and D, had their virulence exalted through serial intramuscular inoculation into guinea pigs. Results showed that, for toxigenic strains (6), one or two passages were enough to cause exaltation, while for the atoxigenic (4), five or six inoculations were needed. Esterase electrophoresis of standard and isolated strains, with and without exaltation, was performed. Electrophoresis analysis permits the following conclusions: strains that do not show any clinical symptoms in mice, when exalted demonstrate decreased...

Detection of bovine Clostridium perfringens by polymerase chain reaction J. Venom. Anim. Toxins incl. Trop. Dis.
Piatti,R. M.; Ikuno,A. A.; Baldassi,L..
A polymerase chain reaction (PCR) assay to detect Clostridium perfringens alpha toxin gene (cpa) was used to identify eighty-nine C. perfringens strains obtained from bovine clinical material. The strains were biochemically characterized as C. perfringens. The isolated strains were cultured on plates containing brain heart infusion agar with 5% sheep blood under anaerobic conditions. DNA extraction was performed by boiling. The 324 bp amplification product of cpa was observed in all isolates. C. sordellii, C. botulinum, C. novyi, and C. septicum were also tested but did not produce any alpha toxin gene amplification. These findings suggest that PCR is a useful assay in identifying C. perfringens toxin types.

Genotyping of Clostridium perfringens associated with sudden death in cattle J. Venom. Anim. Toxins incl. Trop. Dis.
Miyashiro,S; Baldassi,L; Nassar,AFC.
Toxigenic types of Clostridium perfringens are significant causative agents of enteric disease in domestic animals, although type E is presumably rare, appearing as an uncommon cause of enterotoxemia of lambs, calves and rabbits. We report herein the typing of 23 C. perfringens strains, by the polymerase chain reaction (PCR) technique, isolated from small intestine samples of bovines that have died suddenly, after manifesting or not enteric or neurological disorders. Two strains (8.7%) were identified as type E, two (8.7%) as type D and the remainder as type A (82.6%). Commercial toxoids available in Brazil have no label claims for efficacy against type E-associated enteritis; however, the present study shows the occurrence of this infection. Furthermore,...

Determination of multidrug resistance mechanisms in Clostridium perfringens type A isolates using RNA sequencing and 2D-electrophoresis BJMBR
Ma,Yu-Hua; Ye,Gui-Sheng.
In this study, we screened differentially expressed genes in a multidrug-resistant isolate strain of Clostridium perfringens by RNA sequencing. We also separated and identified differentially expressed proteins (DEPs) in the isolate strain by two-dimensional electrophoresis (2-DE) and mass spectrometry (MS). The RNA sequencing results showed that, compared with the control strain, 1128 genes were differentially expressed in the isolate strain, and these included 227 up-regulated genes and 901 down-regulated genes. Bioinformatics analysis identified the following genes and gene categories that are potentially involved in multidrug resistance (MDR) in the isolate strain: drug transport, drug response, hydrolase activity, transmembrane transporter,...

Antimicrobial susceptibility of Clostridium perfringens isolated from piglets with or without diarrhea in Brazil BJM
Salvarani,Felipe Masiero; Silva,Rodrigo Otávio Silveira; Pires,Prhiscylla Sadanã; Cruz Júnior,Eduardo Coulaud da Costa; Albefaro,Isabella Silva; Guedes,Roberto Maurício de Carvalho; Lobato,Francisco Carlos Faria.
The minimum inhibitory concentration (MIC) was determined for 13 antibiotics against Clostridium perfringens isolated from Brazilian piglets. The collection of isolates was performed in June to October 2010. All isolates were susceptible to amoxicillin and ceftiofur, whereas most were resistant to tetracycline and lincomycin. Avilamycin and narasin were more effective against isolates from non-diarrheic than from diarrheic piglets. The other antimicrobials were less active in need of high concentrations to inhibit the growth of the C. perfringens type A. These results suggest the need for further studies evaluating molecular factors related to the antimicrobial resistance of C. perfringens.

Cross protection by heat and cold shock to lethal temperatures in Clostridium perfringens BJM
García,Santos; Limón,Julio C.; Heredia,Norma L..
A heat shock pre-treatment of Clostridium perfringens provides protection against lethality from subsequent exposure to cold shock and vice versa. Heat shocked cells were more cold tolerant (one log reduction in 65 min) than control cells (one log reduction in 34 min). On the other hand, cold-shocked cells were more thermotolerant (D55= 17 min) than the control (D55= 6.5 min). The addition of chloramphenicol (an inhibitor of protein synthesis) in the experiments indicates that synthesis of new proteins was necessary for cross protection between both treatments.

Use of single-enzyme amplified fragment length polymorphism for typing Clostridium perfringens isolated from diarrheic piglets BJM
Shinya,Luciane Tieko; Baccaro,Maria Regina; Moreno,Andrea Micke.
Clostridium perfringens is an important pathogen in human and veterinary medicine. In swine, the agent is responsible for necrotic enteritis and enterotoxemia characterized by diarrhea, weight loss, delayed development and, in some cases, death. In the present study amplified fragment length polymorphism analyses (AFLP) was used to characterize 54 C. perfringens strains isolated from swine presenting diarrhea. Analysis of the results showed 29 distinct profiles with discriminatory index equal to 0.97. Partial correlation between the origin of the isolates and groups was drawn, and correlation was possible in only 18.5% of the samples. Characterization of the strains in biotypes (A, B, C, D and E), production of beta-2 toxin and enterotoxin were performed...
