Effects of salinity on the physiology of the red macroalga, Acanthophora spicifera (Rhodophyta, Ceramiales) Acta Botanica

ABSTRACT Salinity is an important abiotic factor since it is responsible for the local and/or regional distribution of algae. In coastal regions, salinity changes with prevailing winds, precipitation and tide, and particularly in extreme intertidal conditions. Acanthophora spicifera is a red seaweed that occurs in the supratidal region in which changes in abiotic conditions occur frequently. This study evaluated the effects of salinity on the metabolism and morphology of A. spicifera. Algae were acclimatized under culture conditions with sterilized seawater for seven days. P. expansa e P. unifilis. Todos os espécimes utilizados neste trabalho foram procedentes do rio Javaés, entorno do Parque Nacional do Araguaia, Estado do Tocantins, Brasil. Após estudo sob estereomicroscópio, foram coletadas as gônadas para o procedimento histológico de rotina. Foram analisadas 187 amostras de P. expansa, das quais 81,2% foram identificadas como fêmeas e 18,7% como machos; e 98 de P. unifilis, das quais 31,6% eram fêmeas e 68,4% machos. Light microscope observation of circulating human lymphocytes cultured in vitro BABT

The purpose of this work was to study the isolation and a light microscopy technique for cultured lymphocytes. Blood samples were obtained by venipuncture with an anticoagulant added and centrifuged in a Percoll density gradient to separate the leukocytes. Lymphocytes were placed in 25 cm ³ tissue culture flasks at 37ºC. After culturing, they were fixed and stained with the methods used for blood smears. Results showed that not all fixing solutions and stains were an equally good choice for cultured lymphocytes.

The objective of this work was to optimize the spray drying of pequi pulp using maltodextrin as carrier agent and Tween 80 as surfactant agent. A central composite rotatable design was used to evaluate the influence of inlet air temperature (140 to 200°C), maltodextrin (15 to 30%) and surfactant (0 to 5%) concentration on the process performance and physicochemical and nutritional properties of the dried powdered pulp. The segmental structure of the aortic wall at the thoracic and abdominal levels were studied in domestic chicken by light microscopy (LM) and scanning electron microscopy (SEM). Selected histological sections were submitted to histomorphometric studies using image analysis methods. The variables studied were the thickness of the aortic layers in the three segments investigated: ascendant thoracic, descendant thoracic and abdominal parts as well as the tubular diameters and the mean number of elastic lamellae in the medial layer of each analyzed segment. The events involved in the structural interaction between the diazotrophic endophytic bacterium Herbaspirillum seropedicae, strain RAM10, labeled with green fluorescent protein, and pineapple plantlets 'Vitória' were evaluated by means of bright-field and fluorescence microscopy, combined with scanning electron microscopy for 28 days after inoculation. After 6 hours of inoculation, H. seropedicae was already adhered to the roots, colonizing mainly root hair surface and bases, followed by epidermal cell wall junctions. Structural and ultrastructural changes caused by bioaccumulation of As and Cr in brake fern (Pteris vittata), a known arsenic hyperaccumulator, were investigated. Potted plants of brake fern were exposed to metal treatments of As and Cr for three weeks. The objectives of this study were to identify the structural changes caused by Zn and Cd accumulation in shoots and roots of barley (Hordeum vulgare) plants; and to correlate metal accumulation with anatomical, physiological and morphological changes. Potted plants were exposed to metal treatments of Zn and Cd for 19 and 16 d respectively. Leaves, stems and roots were harvested to identify structural changes and analyze metal accumulation. In this work, it was used starch obtained from green banana (Musa cavendishii) and commercial corn (Zea mays) starch in order to compare the granule morphology and the rheological behavior of these gel-starches. Images of starch granules morphology were obtained from scanning electron microscope (SEM). The banana starch granules presented an oval and ellipsoidal shape with irregular diameters. Neverthless, the granules of corn starch showed a poliedric shape, with different sizes. The rheological behavior of gel starch solutions showed a non-newtonian character with a pseudoplastic behavior. Fruit salad as a new vehicle for probiotic bacteria Ciênc. Tecnol. Aliment.

Abstract This work aimed to study the use of fruit salads as carriers for Lactobacillus rhamnosus HN001. We evaluated the viability of this probiotic in fruit salads and the phsyico-chemical, microbiological and sensory properties of this food. Scanning electron microscopy (SEM) was used to verify microorganism adhesion on the fruit tissues. The viability of L. rhamnosus in fruit salads was 8.49 log CFU.g-1 after 120 hours. SEM images showed that fruit tissue provided protection for probiotic. Adhesion sites were observed in higher quantity in banana, apple and guava. The addition of L. rhamnosus did not alter texture of fruits (p > 0.05). Fruit salads containing probiotic had different values of pH and acidity compared to the control (p < 0.05)....

Surimi, washed fish mince after grinding with salt, has a firm and elastic gel texture or “ashi” after thermal processing. The major objectives of this study on horse mackerel (Trachurus trachurus) surimi are to optimize the technological conditions in the aim to obtain the best ashi, to determine the influence of frozen storage and of egg white and potata starch addition, and to establish the preventive effect of incorporation into surimi of different combinations of cryoprotectants and NaC1. Gel texture is, by order importance, dependent on cooking temperature, length of setting and cooking and length of grinding of surimi with texture additives after thawing.
