Replicating animal mitochondrial DNA Genet. Mol. Biol.
McKinney,Emily A.; Oliveira,Marcos T..
The field of mitochondrial DNA (mtDNA) replication has been experiencing incredible progress in recent years, and yet little is certain about the mechanism(s) used by animal cells to replicate this plasmid-like genome. The long-standing strand-displacement model of mammalian mtDNA replication (for which single-stranded DNA intermediates are a hallmark) has been intensively challenged by a new set of data, which suggests that replication proceeds via coupled leading-and lagging-strand synthesis (resembling bacterial genome replication) and/or via long stretches of RNA intermediates laid on the mtDNA lagging-strand (the so called RITOLS). The set of proteins required for mtDNA replication is small and includes the catalytic and accessory subunits of DNA...

Electroacupuncture inhibits apoptosis in annulus fibrosis cells through suppression of the mitochondria-dependent pathway in a rat model of cervical intervertebral disc degradation Genet. Mol. Biol.
Liao,Jun; Ke,Meigui; Xu,Teng; Lin,Lili.
The purpose of this study was to investigate whether treatment with electroacupuncture (EA) inhibited mitochondria-dependent apoptosis in annulus fibrosis (AF) cells in a rat model of cervical intervertebral disc degradation induced by unbalanced dynamic and static forces. Forty Sprague-Dawley rats were used in this study, of which 30 underwent surgery to induce cervical intervertebral disc degradation, 10 rats received EA at acupoints Dazhui (DU 14) and Shousanli (LI 10). TUNEL staining was measured to assess apoptosis in AF cells, immunohistochemistry was used to examine Bcl-2 and Bax expression, colorimetric assays were used to determine caspase 9 and caspase 3 activities and RT-PCR and western blotting were used to assess the mRNA and protein...

Mitochondrial DNA single nucleotide polymorphism associated with weight estimated breeding values in Nelore cattle (Bos indicus) Genet. Mol. Biol.
Biase,Fernando Henrique; Meirelles,Flávio Vieira; Gunski,Ricardo; Vozzi,Pedro Alejandro; Bezerra,Luiz A.F.; Vila,Reginaldo A.; Rosa,Artur J.M.; Lôbo,Raysildo B.; Martelli,Lúcia.
We sampled 119 Nelore cattle (Bos indicus), 69 harboring B. indicus mtDNA plus 50 carrying Bos taurus mtDNA, to estimate the frequencies of putative mtDNA single nucleotide polymorphisms (SNPs) and investigate their association with Nelore weight and scrotal circumference estimated breeding values (EBVs). The PCR restriction fragment length polymorphism (PCR-RFLP) method was used to detect polymorphisms in the mitochondrial asparagine, cysteine, glycine, leucine and proline transporter RNA (tRNA) genes (tRNAasn, tRNAcys, tRNAgly, tRNAleu and tRNApro). The 50 cattle carrying B. taurus mtDNA were monomorphic for all the tRNA gene SNPs analyzed, suggesting that they are specific to mtDNA from B. indicus cattle. No tRNAcys or tRNAgly polymorphisms were...

Role of the MAPK pathway in human lung epithelial-like A549 cells apoptosis induced by paraquat Genet. Mol. Biol.
Sun,Da-Zhuang; Song,Chun-Qing; Xu,Yong-Min; Dong,Xue-Song.
Abstract This study aims to investigate the value of mitogen-activated protein kinases (MAPKs) for paraquat (PQ)-induced apoptosis in human lung epithelial-like A549 cells and the specific mechanism. A549 cell apoptosis were induced by PQ. These cells were divided into six groups: control group (cells were cultured in RPMI-1640 medium); SP600125 group (cells were preconditioned with SP600125); SB203580 group (cells were preconditioned with SB203580); PQ group (cells were treated with PQ); SP600125+PQ group (cells were preconditioned with SP600125 following PQ); SB203580+PQ group (cells were preconditioned with SB203580 following PQ). The cell survival rate, apoptosis rate, and activities of caspase-3 and -9 were detected. When compared with the control...

Roles of the mitochondrial replisome in mitochondrial DNA deletion formation Genet. Mol. Biol.
Oliveira,Marcos T.; Pontes,Carolina de Bovi; Ciesielski,Grzegorz L..
Abstract Mitochondrial DNA (mtDNA) deletions are a common cause of human mitochondrial diseases. Mutations in the genes encoding components of the mitochondrial replisome, such as DNA polymerase gamma (Pol γ) and the mtDNA helicase Twinkle, have been associated with the accumulation of such deletions and the development of pathological conditions in humans. Recently, we demonstrated that changes in the level of wild-type Twinkle promote mtDNA deletions, which implies that not only mutations in, but also dysregulation of the stoichiometry between the replisome components is potentially pathogenic. The mechanism(s) by which alterations to the replisome function generate mtDNA deletions is(are) currently under debate. It is commonly accepted that stalling of...

Maternal oxidative stress and reproduction: testing the constraint, cost and shielding hypotheses in a wild mammal ArchiMer
Viblanc, Vincent A.; Schull, Quentin; Roth, Jeffrey D.; Rabdeau, Juliette; Saraux, Claire; Uhlrich, Pierre; Criscuolo, François; Dobson, F. Stephen.
1.Oxidative stress has been proposed as a central causal mechanism underlying the life history trade-off between current and future reproduction and survival in wild animals. 2.Whereas mixed evidence suggests that maternal oxidative stress may act both as a constraint and a cost to reproduction, some studies have reported a lack of association between reproduction and maternal oxidative stress. 3.The oxidative shielding hypothesis offers an alternative explanation, suggesting that mothers may pre-emptively mitigate the oxidative costs of reproduction by increasing antioxidant defences prior to reproduction. 4.We tested the oxidative constraint, cost, and shielding hypotheses using a longitudinal field study of oxidative stress levels in a species that...

Effect of reproduction on escape responses, metabolic rates and muscle mitochondrial properties in the scallop Placopecten magellanicus ArchiMer
Kraffe, E; Tremblay, R; Belvin, S; Le Coz, Jean-rene; Marty, Y; Guderley, H.
In scallops, gametogenesis and spawning can diminish the metabolic capacities of the adductor muscle and reduce escape response performance. To evaluate potential mechanisms underlying this compromise between reproductive investment and escape response, we examined the impact of reproductive stage (pre-spawned, spawned and reproductive quiescent) of the giant scallop, Placopecten magellanicus, on behavioural (i.e., escape responses), physiological (i.e., standard metabolic rates and metabolic rates after complete fatigue) and mitochondrial capacities (i.e., oxidative rates) and composition. Escape responses changed markedly with reproductive investment, with spawned scallops making fewer claps and having shorter responses than pre-spawned or...

Functional capacities of gill mitochondria in oyster Crassostrea gigas during an emersion/immersion tidal cycle ArchiMer
Dudognon, Tony; Soudant, Philippe; Seguineau, Catherine; Quere, Claudie; Auffret, Michel; Kraffe, Edouard.
Sessile animals that live on the foreshore undergo tidal cycles, and have to face variations in physical and chemical parameters such as oxygen concentration. During emersion, availability of dissolved oxygen can be lowered for bivalves, which have only a small reserve of seawater inside their closed shell. Differences in oxygen concentration are thus expected to lead to modifications of the metabolism, including changes in mitochondrial activity. Previous studies investigated air exposure under extreme conditions, which do not always reflect environmental conditions these invertebrates have to cope with. In this study, oxidative capacities of gill mitochondria of the oyster Crassostrea gigas were studied during a tidal cycle period, by comparing oysters...

Spatial compartmentalization of free radical formation and mitochondrial heterogeneity in bivalve gills revealed by live-imaging techniques ArchiMer
Rivera-ingraham, Georgina A.; Rocchetta, Iara; Bickmeyer, Ulf; Meyer, Stefanie; Abele, Doris.
Background: Reactive oxygen (ROS) and nitrogen (RNS) species are produced during normal unstressed metabolic activity in aerobic tissues. Most analytical work uses tissue homogenates, and lacks spatial information on the tissue specific sites of actual ROS formation. Live-imaging techniques (LIT) utilize target-specific fluorescent dyes to visualize biochemical processes at cellular level. Results: Together with oxidative stress measurements, here we report application of LIT to bivalve gills for ex-vivo analysis of gill physiology and mapping of ROS and RNS formation in the living tissue. Our results indicate that a) mitochondria located in the basal parts of the epithelial cells close to the blood vessels are hyperpolarized with high Delta Psi m, whereas...

Osmoregulation, bioenergetics and oxidative stress in coastal marine invertebrates: raising the questions for future research ArchiMer
Rivera-ingraham, Georgina A.; Lignot, Jehan-herve.
Osmoregulation is by no means an energetically cheap process, and its costs have been extensively quantified in terms of respiration and aerobic metabolism. Common products of mitochondrial activity are reactive oxygen and nitrogen species, which may cause oxidative stress by degrading key cell components, while playing essential roles in cell homeostasis. Given the delicate equilibrium between pro- and antioxidants in fueling acclimation responses, the need for a thorough understanding of the relationship between salinity-induced oxidative stress and osmoregulation arises as an important issue, especially in the context of global changes and anthropogenic impacts on coastal habitats. This is especially urgent for intertidal/estuarine organisms, which may...

The Cytoplasm Affects the Epigenome in Drosophila melanogaster ArchiMer
Grunau, Christoph; Voigt, Susanne; Dobler, Ralph; Dowling, Damian K.; Reinhardt, Klaus.
Cytoplasmic components and their interactions with the nuclear genome may mediate patterns of phenotypic expression to form a joint inheritance system. However, proximate mechanisms underpinning these interactions remain elusive. To independently assess nuclear genetic and epigenetic cytoplasmic effects, we created a full-factorial design in which representative cytoplasms and nuclear backgrounds from each of two geographically disjunct populations of Drosophila melanogaster were matched together in all four possible combinations. To capture slowly-accumulating epimutations in addition to immediately occurring ones, these constructed populations were examined one year later. We found the K4 methylation of histone H3, H3K4me3, an epigenetic marker...

HIGH RESOLUTION SEM OF INTRINSIC MUSCLE FIBERS OF ANTERIOR THIRD RET'S TONGUE International Journal of Morphology
Motoyama,Aracy A.; Watanabe,Ii-sei; Ogawa,Koichi.
The characteristics of intrinsic muscle fibers in the anterior third of the adult rat tongue were studied employing light microscopy, SEM and high-resolution SEM (HRSEM) methods. For light microscopy, specimens were fixed in Bouin's solution and embedded in paraffin. In order to identify the muscle fibers and bundles of collagen fibers, frontal sections were stained with hematoxylin-eosin, azo-carmin and picro-sirius. The results showed that muscle fibers near the lamina propria are fixed with connective tissue, constituting several groups. The bundles of muscle fibers are arranged in longitudinal, vertical and transversal directions. For SEM, specimens were fixed in modified Karnovsky solution and freeze-fractured in liquid nitrogen revealed the collagen...

Effect of Cigarette Smoking on the Structure of Hepatocytes: TEM Study International Journal of Morphology
Battah,Khairat A; Badran,Darwish H; Shraideh,Ziad A.
The liver is one of the major organs that is indirectly affected by cigarette smoke. The aim of this project is to define the histologic and ultrastructural changes in normal liver cells after exposing animals to cigarette smoke. Thirty albino rats were exposed to cigarette smoke for 90 days, followed by morphologic examination of their livers under light microscope and electron microscope. The liver cells of cigarette smoke exposed rats showed mild swelling with increased eosinophilia. Ultrastructural examination of these cells demonstrated cytoplasm with highly proliferated and crowded mitochondria. There were many electron dense mitochondria. These mitochondria were pleomorphic in shape compared to mitochondrias of control rats. Also, loss of...

Identification and functional analysis of a novel mitochondria-localized 2-Cys peroxiredoxin, BbTPx-2, from Babesia bovis OAK
Masatani, Tatsunori; Asada, Masahito; Hakimi, Hassan; Hayashi, Kei; Yamagishi, Junya; Kawazu, Shin-ichiro; Xuan, Xuenan.
Cysteine-based peroxidases, known as peroxiredoxins (Prx) or thioredoxin peroxidases (TPx), are important antioxidant enzymes that prevent oxidative damage caused by reactive oxygen species (ROS). In this study, we identified a novel mitochondrial 2-Cys Prx, BbTPx-2, from a bovine Babesia parasite, B. bovis. BbTPx-2 complementary DNA (cDNA) encodes a polypeptide of 254 amino acid residues. This protein has a mitochondrial targeting peptide at the N-terminus and two conserved cysteine residues of the typical 2-Cys Prx. By using a thiol mixed-function oxidation assay, the antioxidant activity of recombinant BbTPx-2 was revealed, and its antioxidant activity was comparable to that of a cytosolic 2-Cys Prx from B. bovis, BbTPx-1. Notably, we confirmed that...

Deformation of Mitochondrial Cristae in Human Neural Progenitor Cells Exposed to Valproic Acid Anais da ABC (AABC)
COSTA,RODRIGO M. DA; KARMIRIAN,KARINA; REHEN,STEVENS K..
ABSTRACT Neural development represents a dynamic process where mitochondrial integrity is decisive for neuronal activity. Structural changes in these organelles may be related to neurological disorders. Valproic acid (VPA) is an anticonvulsive drug commonly used for epilepsy treatment and its use is associated to increased risk of neuropsychiatric disorders. Recently we showed changes in shape and membrane potential in mitochondria from human neural progenitor cells (NPCs) exposed to VPA (da Costa et al. 2015). Here, we applied transmission electron microscopy and electron tomography to evaluate mitochondrial damage caused by VPA in NPCs. Results showed mitochondrial cristae disorganization in a dose dependent manner. Disturbance in mitochondrial...

Mitochondrial DNA damage associated with lipid peroxidation of the mitochondrial membrane induced by Fe2+-citrate Anais da ABC (AABC)
Almeida,Andréa M.; Bertoncini,Clélia R.A.; Borecký,Jiri; Souza-Pinto,Nadja C.; Vercesi,Aníbal E..
Iron imbalance/accumulation has been implicated in oxidative injury associated with many degenerative diseases such as hereditary hemochromatosis, beta-thalassemia, and Friedreich's ataxia. Mitochondria are particularly sensitive to iron-induced oxidative stress - high loads of iron cause extensive lipid peroxidation and membrane permeabilization in isolated mitochondria. Here we detected and characterized mitochondrial DNA damage in isolated rat liver mitochondria exposed to a Fe2+-citrate complex, a small molecular weight complex. Intense DNA fragmentation was induced after the incubation of mitochondria with the iron complex. The detection of 3' phosphoglycolate ends at the mtDNA strand breaks by a 32P-postlabeling assay, suggested the involvement of...

An overview of chagasic cardiomyopathy: pathogenic importance of oxidative stress Anais da ABC (AABC)
Zacks,Michele A.; Wen,Jian-Jun; Vyatkina,Galina; Bhatia,Vandanajay; Garg,Nisha.
There is growing evidence to suggest that chagasic myocardia are exposed to sustained oxidative stress-induced injuries that may contribute to disease progression. Pathogen invasion- and replication-mediated cellular injuries and immune-mediated cytotoxic reactions are the common source of reactive oxygen species (ROS) in infectious etiologies. However, our understanding of the source and role of oxidative stress in chagasic cardiomyopathy (CCM) remains incomplete. In this review, we discuss the evidence for increased oxidative stress in chagasic disease, with emphasis on mitochondrial abnormalities, electron transport chain dysfunction and its role in sustaining oxidative stress in myocardium. We discuss the literature reporting the consequences of...

Calcium and cell death signaling in neurodegeneration and aging Anais da ABC (AABC)

Transient increase in cytosolic (Cac2+) and mitochondrial Ca2+ (Ca m2+) are essential elements in the control of many physiological processes. However, sustained increases in Ca c2+ and Ca m2+ may contribute to oxidative stress and cell death. Several events are related to the increase in Ca m2+, including regulation and activation of a number of Ca2+ dependent enzymes, such as phospholipases, proteases and nucleases. Mitochondria and endoplasmic reticulum (ER) play pivotal roles in the maintenance of intracellular Ca2+ homeostasis and regulation of cell death. Several lines of evidence have shown that, in the presence of some apoptotic stimuli, the activation of mitochondrial processes maylead to the release of cytochrome c followed by the activation of...

Knockdown of apoptosis-inducing factor disrupts function of respiratory complex I Biocell
Varecha,Miroslav; Páclová,Daniela; Procházková,Jirina; Matula,Pavel; Cmarko,Dusan; Kozubek,Michal.
Recent findings suggest that apoptotic protein apoptosis-inducing factor (AIF) may also play an important non-apoptotic function inside mitochondria. AIF was proposed to be an important component of respiratory chain complex I that is the major producer of superoxide radical. The possible role of AIF is still controversial. Superoxide production could be used as a valuable measure of complex I function, because the majority of superoxide is produced there. Therefore, we employed superoxide-specific mitochondrial fluorescence dye for detection of superoxide production. We studied an impact of AIF knockdown on function of mitochondrial complex I by analyzing superoxide production in selected cell lines. Our results show that tumoral telomerase-positive (TP)...
