E2F1 plays a key role in cell-cycle regulation in mammals, since its transcription factor activity controls genes required for DNA synthesis and apoptosis. E2F1 deregulation is a common feature among different tumor types and can be a major cause of cell proliferation. Thus, blocking E2F1 expression by RNA interference represents a promising therapeutic approach. In this study, the introduction of specific short hairpin RNAs (shRNAs) reduced E2f1 expression by up to 77%, and impaired rat glioma cell proliferation by approximately 70%, as compared to control cells. Furthermore, we investigated the expression of E2f1 target genes, Cyclin A and Cyclin E. Cyclin A was found to be down-regulated, whereas Cyclin E had similar expression to control cells,... |

The effect of 11 marine-derived cryptides was investigated on proliferation, differentiation and maturation of human white pre-adipocytes (HWP). They were all formerly identified as potent Angiotensin-Converting-Enzyme inhibitors.Val-Trp (VW),Val-Tyr (VY), Lys-Tyr (KY), Lys-Trp (KW), Ile-Tyr (IY), Ala-Pro (AP),Val-Ile-Tyr (VIY), Leu-Lys-Pro (LKP), Gly-Pro-Leu (GPL), Ala-Lys-Lys (AKK) and Val-Ala-Pro (VAP) were previously found in fish products and coproducts as well as other marine resources like wakame. Treatment with AP, VAP and AKK greatly affected viability of HWP during the proliferation period while KW and VW treatment reduced the number of viable cells during the differentiation stage. A GPL and IY incubation during the differentiation stage allowed... |

The 1998 survey of green tides on the Brittany coast was performed in the same way as that of 1997 in order to allow comparison. 11 includes an inventory ofail the concerned sites, and a precise assessment ofthirteen ofthem, the most loaded. On these ones, nitrogen fluxes were surveyed during the critical period, and the green seaweed stock on the beach and in the first waves was quantified. The climatic conditions were opposite between the two years, very dry in spring and summer 1997, and wet in 1998. The irradiance contrast was inverse, and less pronounced. The nitrate contents and mostly the flows were more important in 1998. They generated nitrogen fluxes about 3.3 times those of 1997. The total number of sites concerned in 1998 is clearly higher than... |

On tens of sandy beaches of Brittany, a green seaweed (Ulva) blooms during spring and summer. In the sites where the proliferation is the most important, the total biomass amounted to 20 000 t of wet weight in 1997, at the time of maximum. These "green tides" occur every year in nearly the same locations. They have a very harmful impact on local tourism. In many communes, beached and rotting algae are collected and dumped (43 000 m 3 as a whole in 1997). The main development of biomass takes place between the surf zone and the bare sand. The outbreak requires three coexisting conditions : 1) a sandy beach gently sloping, 2) a weak residual current (i.e. once the tidal back and forth movement has been filtered), 3) a nitrogen loading, even moderate (ammonium... |

Introduction Massive proliferations of algae and, in particular green macroalgae, have been known all over the continents for several decades. They affect various littoral sites and belong to diverse species (Ulva, Enteromorpha and especially Cladophora). In 1990, on the North Brittany shore, more than 80 000 m3 ulvas have been mechanically removed by affected communities. Up until recently, the Arcachon basin escaped from such phenomenon, even if the diversity and quantity of algae found there had already undergone increasing periods in the past. However, the first algae proliferations only started in the 80's, which scale was large enough to be a nuisance to all activities taking place in the basin. At first (starting in1982), those proliferations were... |

The yearly study of Ulva in Brittany aims at building a time series of its proliferations and corresponding nutrient loadings. The goal is to detect results of wilful loading reductions, and to distinguish them from natural variations. This first actually quantified study is the starting bench-mark of the process. Forty-five concerned sites were listed in 1997. They are nearly the same as in 1988, 1991 and 1994 (previous rough census). The Côtes d'Armor and then the Finistère are the most affected departments. The Morbihan is less and file et Vilaine not at all. Ulva biomass was precisely quantified in about fifteen locations during the growing season, along with nitrogen loadings of local streams (mostly nitrate). The fluxes of nitrogen - the limiting... |

Antimetastatic properties on both murine and human osteosarcoma cell lines (POS-1 and KHOS) have been evidenced using exopolysaccharide (EPS) derivatives, produced by Alteromonas infernus bacterium. These derivatives had no significant effect on the cell cycle neither a pro-apoptotic effect on osteosarcoma cells. Based on this observation, these EPSs could be employed as new drug delivery systems for therapeutic uses. A theranostic approach, i.e., combination of a predictive biomarker with a therapeutic agent, has been developed notably by combining with true pair of theranostic radionuclides, such as scandium 47Sc/44Sc. However, it is crucial to ensure that, once complexation is done, the biological properties of the vector remain intact, allowing the... |

Tumorhead (TH) regulates neural plate cell proliferation during Xenopus early development, and gain or loss of function prevents neural differentiation. TH shuttles between the nuclear and cytoplasmic/cortical cell compartments in embryonic cells. In this study, we show that subcellular distribution of TH is important for its functions. Targeting TH to the cell cortex/membrane potentiates a TH gain of function phenotype and results in neural plate expansion and inhibition of neuronal differentiation. We have found that TH subcellular localization is regulated, and that its shuttling between the nucleus and the cell cortex/cytoplasm is controlled by the catalytic activity of p21-activated kinase, X-PAK1. The phenotypes of embryos that lack, or have excess,... |

The ultimate goal of periodontal therapy is to repair the damaged periodontal supporting tissues, permitting regeneration of the periodontal ligament. However, the cell response, the supportive matrix and the bioactive molecules use have not yet been well established. Bone marrow mononuclear cells were extracted from rat femurs and tibiae and cultured on a cross-linked collagen membrane, bone graft, or molar tooth to compare cell attachment and early proliferation on these materials. Cell attachment was quantified by light microscopy at 24, 48 and 72h, and cell proliferation was observed under a SEM after 72h. After 24h, the number of cells on bone graft was similar to that of the control and more than twice compared to collagen membrane (q=7.473... |

Organophosphorous pesticides are a healh problem worldwide, mostly for agroworkers, which are around 2600 millions of person in the year 2001. Little is known about male reproductive damage elicited by these chemicals. The acute effects of malathion (single intraperitoneal injection), 1/12 the LD50 to mice were analyzed at 1, 8, 16, 35 and 40 days after injection, in terms of spermatogenic cell proliferation and apoptosis and of Sertoli cell compromise as revealed by immunocytochemical detection of CK-18 (cytokeratin). The micronuclei test was done to assess for genotoxic activity of the agropesticide. The results showed decreased germ cell number followed by compensatory spermatogonial proliferation by 16 days (as shown by thymidine-H3 testicular uptake),... |

Overexpression of Short and Raji variants of Cellular FLICE-like inhibitory protein (c-FLIP) is capable of inhibiting apoptosis, while the function of the Long isoform depends of c-FLIPL concentration in cells. The aim of this study was to determine the effects of c-FLIPL knockdown in cervical cell lines. SiHa, C-4I and C-33A cervical cancer cell lines were analyzed. c-FLIPL level expression was determined by quantitative real-time PCR and western blotting. c-FLIPL was transiently downregulated by siRNA. The effects of knockdown of c-FLIPL on cell viability, proliferation and apoptosis were assessed by comparing with scrambled siRNA-transfected cells. SiHa and C-4I c-FLIPL knockdown cells showed increased viability compared with scrambled siRNA-transfected... |

The restriction of the mechanisms of cell proliferation in murine seminiferous epithelium, in terms of induction of programmed cell death until recently has not been fully analyzed. The aim of this work was to assess the effect of Malathion (MP) on testicular morphology and function in mouse spermatogenesis. For the experiments, male albino mice of strain NMRI-IVIC, weighing between 30-40 g were used, and divided into control and experimental groups of 5 each. The animals of the experimental groups were injected with a single dose of MP: 241mg/kg weight (1/12 LD 50 ) resuspended in 0.9% saline, intraperitoneally. Animals were sacrificed at 8.3, 16.6 and 33.2 days post-injection (first, second and third spermatogenic cycles). Testicular samples were... |

In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. At proliferation stage, the maximum number of new leaves (9.6) were produced on the medium containing 4 µM BAP + 0 µM GA3, whereas the maximum number of axillary shoots (4.1) were observed in the medium containing 4 µM BAP + 3 µM GA3. The results for rooting experiments suggested that the highest increase in stem height (49.5 mm) at the acclimatization stage was observed in plantlets treated with 1 µM... |

Abstract Background Pathologic vascular smooth muscle cell (VSMC) proliferation and migration after vascular injury promotes the development of occlusive vascular disease. Therefore, an effective chemical agent to suppress aberrant proliferation and migration of VSMCs can be a potential therapeutic modality for occlusive vascular disease such as atherosclerosis and restenosis. To find an anti-proliferative chemical agent for VSMCs, we screened an in-house small molecule library, and the selected small molecule was further validated for its anti-proliferative effect on VSMCs using multiple approaches, such as cell proliferation assays, wound healing assays, transwell migration assays, and ex vivo aortic ring assay. Results Among 43 initially screened... |

Abstract Background Ubiquitin specific peptidase 39 (USP39), an essential factor in the assembly of the mature spliceosome complex, has an aberrant expression in several cancer. However, its function and the corresponding mechanism on human osteosarcoma has not been fully explored yet. Methods The mRNA and DNA copies of USP39 were increased in osteosarcoma cancer tissues compared with the one in human normal tissues according to datasets from the publicly available Oncomine database. A further western blot analysis also demonstrated an aberrant endogenous expression of USP39 in three different osteosarcoma cells. Then lentivirus-mediated short hairpin RNA (shRNA) was designed to silence USP39 in human osteosarcoma cell line U2OS, which is used to test... |

Skeletal muscles have the potential to regenerate by activation of quiescent satellite cells, however, the molecular signature that governs satellite cells during muscle regeneration is not well defined. Myosin light chains (Myls) are sarcomere-related proteins as traditional regulator of muscle contraction. In this report, we studied the possible role of Myl in the proliferation of skeletal muscle-derived myoblasts. Compared to diaphragm-derived myoblasts, the extraocular muscle-derived myoblasts with lower levels of Myl proliferated faster, maintained a longer proliferation phase, and formed more final myotubes. It was found that blockading Myl with anti-Myl antibody or knockdown of Myll by siRNA targeted against Myll could enhance the myoblast... |
