Carbon sources and polyethylene glycol on soybean somatic embryo conversion PAB

Somatic embryogenesis is an efficient method for the production of target cells for soybean genetic transformation. However, this method still offers low percentages of plant regeneration, and perhaps is related to the maturation process and high morphological abnormalities of the matured embryos. This study aimed to identify a maturation medium that could contribute to the outcome of more efficient plant regeneration results. Embryogenic clusters, derived from cotyledons of immature seeds of the soybean cultivars Bragg and IAS5, were used as starting material for embryos development. Plant architecture of Paspalum vaginatum schwartz modified by nitrate and ammonium nutrition PAB

Paspalum vaginatum Schwartz plants were grown under greenhouse conditions in a continuous-flow hydroponic culture, containing NO3- or NH4+or NH4NO3 as nitrogen source. After 30 days, the size of aerial biomass and root system decreased significantly when plants were supplied with NH4+as exclusive nitrogen source. Compared to NO3- treatment, reducing and non-reducing sugars were decreasing together with a significant increase in amino acids content. NH4+-nutrition caused tillers to grow toward an orthogravitropic position (average angle of 68° with respect to the horizontal), and with NO3--nutrition, tillers tended to become diagravitropic (average angle of 23°). Assimilate partitioning in leaves of the raffinose-storing herb Lamium album L.: photosynthesis and carbon partitioning throughout the photoperiod Rev. Bras. Bot.

Lamium album accumulates starch, sucrose and raffinose-family oligosaccharides (RFO) as the major products of photosynthesis. These products were measured in leaves throughout a sixteen-hour photoperiod and under various irradiance conditions. There was continuous accumulation of sucrose and starch. The rate of gas exchange was higher at 500 µEm² s-1 and 900 µEm²s-1 than at 300 µEm² s-1. The rate of photosynthesis did not decline over the sixteen-hour photoperiod, which suggested that there was no short-term feed back inhibition due to sucrose accumulation in this plant. In Lamium album, sucrose and raffinose-family oligosaccharides are the major products of photosynthesis that are stored in leaves. Using gas analysis and 14CO2 feeding, we compared photosynthesis and the partitioning of recently-fixed carbon in plants where sink activity was lowered by excision of flowers and chilling of roots with those where sink activity was not modified. Reduction in sink activity led to a reduction in the maximum rate of photosynthesis, to retention of fixed carbon in source leaves and to the progressive accumulation of raffinose-family oligosaccharides. Effects of preculture with sucrose and aba on cell suspensions water status and its relation with vitrification resistance Rev. Bras. Fisiol. Veg.

Changes in cell water relations during precultures were followed in an attempt to understand the mechanism of cell hardening for cryopreservation by vitrification. Medium containing 0.4 M sucrose (psiw=-1.45 MPa) and containing 5 mg L-1 of ABA (MT psiw=-0.73 MPa and MS psiw=-0.48 MPa) were used to harden cell suspensions of orange and carrot. Preculture in these medium did not cause a significant decrease of cell viability, however, it improved the cell survival to PVS2 and liquid nitrogen expositions. Cell suspension cultures of Bauhinia forficata Link, Curcuma zedoaria Roscoe and Phaseolus vulgaris L. were used to test plant ability to utilize an alternative to sucrose as carbon source and energy for growth. Glycerol, sorbitol and galactose were the alternative carbon sources tested. Cell suspension cultures established on liquid medium containing sucrose were transferred to culture medium supplemented with sucrose or glycerol, or sorbitol, or galactose as the sole carbon source. Fresh and dry weight increasing and protein content showed marked differences among the different carbon sources used. Sucrose was the best carbon source for all the three plant species tested. The pre-acclimatization stage can be used to improve micropropagation protocols and increase the yield of produced plants. The influence of sucrose and photon flux density (PFD) levels on the acclimatization of in vitro-grown 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstocks was evaluated. In order to increase our knowledge of sucrose metabolism in coffee species, sucrose and reducing sugar content were investigated in the fast fruit-ripening coffee species Coffea racemosa. Fruits were harvested regularly from anthesis, until the point of complete fruit maturation and tissue development, followed by measurement of fruit tissue weight. A metodologia de camada fina de células foi utilizada com sucesso para indução de embriogênese somática em região do meristema apical de plantas de pupunha mantidas em casa-de-vegetação. O efeito de três fontes de carbono: sacarose, glicose e manitol na indução de embriogênese somática foi avaliado. Embriões somáticos foram observados após a indução apenas nos meios contendo sacarose ou glicose em igual proporção.

Ilex paraguariensis (yerba mate) is a native species from South America and is a rich source of bioactive compounds. There is a lack of research efforts on the phytochemical investigation of callus culture from this species. In the present study, an effort was made to optimize callus culture conditions and to identify secondary compounds. Calli were induced from 10 genotypes using leaf explants and the best genotype was selected to evaluate the effects of cytokinin types and concentrations on callus induction and biomass accumulation. This study aimed to assess the biochemical responses of Parkia gigantocarpa saplings to waterlogging. Waterlogging was imposed by flooding the saplings in 14 kg pots to approximately 5 cm above the soil surface. The experimental design was completely randomized and involved two water conditions (control and waterlogging) over five assessment periods (after 0, 4, 8, 12 and 16 days of waterlogging), with five replications per treatment. Starch, sucrose, total soluble carbohydrates and total soluble proteins were measured. The saplings exposed to waterlogging showed significant reductions in starch concentrations (62.50%) and total soluble proteins (40.98%), especially in the leaves. Abstract: Plant resistance inducers are cost-effective and environmentally pleasing strategies of plant protection to mitigate biotic and abiotic agents threatening food safety and energy security. We, accordingly, present jasmonic acid and k-phosphite as low-cost strategies to enhance productive yield and technological quality of sugarcane crop. Exogenously treatment of the sugarcane variety SP81-3250 consisted of carrying out foliar application of jasmonic acid at 1, 1.5 and 2 ml L-1 and K-phosphite at 2, 4 and 6 ml L-1 before crop flowering. This study aimed to evaluate the efficacy of adding sucrose in vitrification solution of ovine embryos produced in vivo. Forty Dorper ewes were selected and superovulated. Immediately prior to the embryo collection by laparotomy, a laparoscopy was erformed to verify the superovulatory response. Virola surinamensis is a forest species widely distributed in the estuaries of the Amazon. These ecosystems are susceptible to contamination by Cadmium (Cd), indicating that the plant has strategies for tolerating this metal. The aim of this study was to assess the nitrogen and carbon metabolism of young plants of Ucuúba (Virola surinamensis) in the presence of cadmium with the perspective of the phytoremediation of contaminated environments.
