Micrurus spixii (PERUVIAN CORAL SNAKE) VENOM - PRELIMINARY BIOCHEMICAL AND ENZYMATIC CHARACTERIZATION J. Venom. Anim. Toxins

Micrurus spixii venom was studied after fractionation by Sephadex G-100 SF gel filtration chromatography. Several enzymatic activities and biological effects were investigated in whole venom and fractions. The venom was resolved in four peaks in a range of about 73.2-10.7 kDa molecular weight. Alkaline phosphatase and acetylcholinesterase activities were found in peak I, and procoagulant activity was seen in peak II. Phospholipase A2, hemorrhagic, and proteolytic activities were detected in peak III. A second procoagulant factor and proteinase were present in peak IV. Thrombin-like enzyme and direct hemolytic activities were not found in any assayed samples.

Toads (order: Anura; family: Bufonidae; genus: Bufo) are distributed throughout the world, but more species are found in areas of tropical and humid temperate climates. Although toads do not have a venom inoculation system, they are venomous animals because the glands covering the whole surface of their bodies secrete a milk-like venom of which composition is not yet completely known. Some of these glands are the bilateral glands located in post-orbital position. These glands, which are somewhat diamond-shaped and can be seen by the naked eye, are known as parotids. The present paper aimed at testing the action of non-lyophilized venom of Africanized bees Apis mellifera through topical applications on Diatraea saccharalis egg masses. The CL50, DL50 and the most susceptible age of eggs to the venom topic application were also determined. Three-day-old eggs were the most susceptible to the venom action with CL50 equal to 8.6 mg/ml and DL50 equal to 0.173 mg/mass. The venom loses its action after being stored for 15 days.

The search for biological antitumor agents has been pursued for over half a century. Snake venom has been shown to possess a wide spectrum of biological activities. The objectives of the present review are to evaluate the existing controversies on this subject published in a number of papers and to propose probable explanations for the phenomena observed. We reported our results obtained in a study, in which we evaluated the action of the venoms of Crotalus durissus terrificus and Bothrops jararaca on Ehrlich ascites tumor cells. The aim of this study was to compare individual venom samples of Crotalus durissus terrificus recently captured in the wild to evaluate possible differences in venom protein composition. Protein levels were quantified by biochemical method (Biuret) and then submitted to electrophoresis. Electrophoresis studies of native protein were performed in vertical slabs of polyacrylamide gel (PAGE), in an alkaline discontinuous buffer system, with a concentration of 10% in the separation gel. Scorpion envenoming is a public health concern in northeastern Venezuela. Specimens of the genus Tityus are responsible for most of these. In experimental animals, Tityus venom produces histopathological changes in the skeletal muscle and pancreas, but its toxicity to the reproductive system has not been studied. The aim of this work is to describe the histopathological changes in testis and epididymis of albino mice induced by the administration of Tityus n. sp. venom. The present study demonstrates that Vipera ammodytes venom is a potent inhibitor of human complement activation. Crude venom was fractionated by ion-exchange chromatography and each fraction was then tested for proteolytic, hemolytic and anti-complement activities. Three peaks decreased the hemolytic activity of human serum when measured using assays for the classical and alternative pathways of activation. One of the anti-complement peaks contains a protease capable of cleaving the C3 component of complement.

Some properties of a Africanized honeybee venom proteases were determined by enzymatic assays in solution, electrophoresis in SDS-PAGE, and gel filtration. Bee venom extracts were obtained by reservoir disruption, selective dialysis (cut off 12 kDa) to eliminate small components, such as the protease inhibitor present in the venom, and then fractionation of the dialyzed extract by gel filtration on a Sephadex G-100 column. The optimal conditions for the caseinolytic assays were pH 9.5, 2-hour digestion at 37 °C, and 1% casein concentration. It is well documented that several pharmacological substances are released within the victim's body after snakebite. These substances are also believed to be endogenously present in animals, specifically levels of myoglobin and histamine that are reported to rise after envenomation. However, there is no published data regarding the presence of these substances in venoms per se. This research reports the detection of myoglobin and histamine in snake, scorpion, honeybee, and toad venoms by immunological test. Aurelia aurita is a scyphozoan, abundant in the Mexican Caribbean during summer. Although usually innocuous, there is evidence of it causing harm to humans. This work investigates the biological activities of crude and fractionated extracts of A. aurita. Live specimens were collected between July and September 1999 from the Mexican Caribbean. The tentacular margin was dissected immediately and frozen at -50ºC. A nematocyst suspension was prepared, discharged, and the supernatants lyophilized. A comparison among the profiles of molecular exclusion chromatography in Sephadex G 100 column of venoms from Apis mellifera adansonii and Africanized honeybees revealed unique peaks which might be used to identify these populations. The venoms from hybrid populations resulting from the reciprocal mating of Apis mellifera adansonii and Africanized honeybees presented unique peaks, probably resulting from a synergistic effect between the parental genomes. The occurrence of characteristic peaks in venoms of hybrid populations might be used to identify these populations as well as to distinguish them from their parents.

A sandwich-type ELISA technique for specific and sensitive detection of Crotalus durissus terrificus venom antigens, horse-antivenom, human IgG and IgM antibodies was set up. Sixteen patients, 13 males and 3 females aged between 13 to 63 years (mean 33 ± 15) bitten by Crotalus durissus terrificus snakes were studied. Of the 15 patients, 6 had previously received anti-Crotalus venom and no seric venom was detected. For the other 9 patients studied, the venom levels ranged from 2 to 108 ng/ml according to the severity of each case. Seric antivenom was detected up to 44 days after the bite. ABSTRACT Background: Accidents caused by spiders of the genus Loxosceles constitute an important public health problem in Brazil. The venom of Loxosceles sp induces dermonecrosis at the bite site and systemic disease in severe cases. Traditional medicine based on plant-derived products has been proven to reduce the local effects of envenomation. The present study verified the healing effects of copaiba oil on lesions induced by the venom of L. intermedia. This work presents the experience of the Laboratory of Arthropods at Instituto Butantan, which maintains scorpions in captivity in order to obtain the venom used in the production of anti-arachnid serum. Between 1993 and 2000, the laboratory received 24.781 specimens of Tityus serrulatus in order to obtain poison for the production of anti-scorpion serum. Euscorpius (Scorpiones, Euscorpiidae): three cases of stings in northwestern Italy J. Venom. Anim. Toxins incl. Trop. Dis.

In the period between June 2008 and August 2009, three cases of stings of Euscorpius scorpions indigenous to Italy were treated at two different emergency departments (ED) in hospitals of the Piedmont region, northwest Italy: Santa Croce e Carle General Hospital in Cuneo, and Santissima Annunziata Hospital in Savigliano. Scorpion stings in Italy are rare and not well documented in the literature; this situation may raise doubts among medical personnel as to how such lesions are best treated. Analysis of the incidents confirms that the venom of Euscorpius do not provokes systemic poisoning in humans and in these cases even dermatological reactions were not significant.

A comparative study on the sensitivity of erythrocytes from different vertebrate species (avian, mammalian and reptilian) to the hemolytic action caused by cardiotoxin isolated from Naja naja atra venom was carried out. Cardiotoxin was able to induce direct hemolysis in washed erythrocytes from several animals, except for llama. The EC50 values from hemolysis of the most sensitive (cat) and the most resistant (snake) animal varied approximately tenfold. Purification and partial characterization of phospholipases A2 from Bothrops asper (barba amarilla) snake venom from Chiriguaná (Cesar, Colombia) J. Venom. Anim. Toxins incl. Trop. Dis.

Components with phospholipase A2 activity were isolated by gel filtration and cationic exchange chromatography from the venom of Bothrops asper snakes from Chiriguaná, Colombia (9°22´N; 73°37´W). Five fractions were obtained by the gel filtration, and PLA2 activity was found in fraction 3 (F3). In the cationic exchange chromatography, F3 showed eight components with PLA2 activity. Six of these components appeared as one band in polyacrylamide gel electrophoresis (SDS-PAGE). Fractions II and VII exhibited an optimal activity at pH 9 and 52ºC.
