Chromosomal diversity in three species of electric fish (Apteronotidae, Gymnotiformes) from the Amazon Basin Genet. Mol. Biol.

Cytogenetic studies were carried out on samples of Parapteronotus hasemani, Sternarchogiton preto and Sternarchorhamphus muelleri (Apteronotidae, Gymnotiformes) from the Amazon basin. The first two species exhibited both a 2n = 52 karyotype, but differed in their karyotypic formulae, distribution of constitutive heterochromatin, and chromosomal location of the NOR. The third species, Sternarchorhamphus muelleri, was found to have a 2n = 32 karyotype. In all three species the DAPI and chromomycin A3 staining results were consistent with the C-banding results and nucleolar organizer region (NOR) localization. Allozyme variation among three populations of the armored catfish Hypostomus regani (Ihering, 1905) (Siluriformes, Loricariidae) from the Paraná and Paraguay river basins, Brazil Genet. Mol. Biol.

Three Brazilian populations of the armored catfish Hypostomus regani (Ihering, 1905) were sampled, one from the Corumbá Reservoir in Goiás state, another from the Itaipu Reservoir in Paraná state and a third from the Manso Reservoir in Mato Grosso state. Allozyme electrophoresis was used to establish the genetic structure of the species, with the analysis of liver, heart and muscles tissues allowing the scoring of 25 loci from 14 enzymatic systems. Since capture is stressful to these animals, non-invasive sampling methods such as the use of feces can provide reliable sources of DNA. Interchromosomal distribution of gamma ray-induced chromatid aberrations in Chinese hamster ovary cells Genet. Mol. Biol.

Interchromosomal distributions of breakpoints from chromatid-type aberrations induced by gamma rays in Chinese hamster ovary (CHO) cells were analyzed. In most chromosomes the distribution was as expected from chromosome lengths for simple breaks or the respective relative corrected length in case of exchanges. There were deviations from expectation in a few chromosomes for chromatid breaks, interchanges, intra-arm intrachanges and inter-arm intrachanges. Especially interesting are the results concerning chromosomes 2 and 8, which were more often involved in exchanges than expected. An "exchange phenotype" for these chromosomes is proposed and possible explanations for the nonrandom distribution of chromosome breakpoints are presented.

Premature ovarian failure (POF) in Brazilian fragile X carriers Genet. Mol. Biol.

The gynecological and reproductive histories of 193 women from fragile X families were surveyed. Among the 101 carriers of the premutation, 14 experienced premature menopause, contrarily to their 37 fully mutated and 55 noncarrier female relatives. Although premature menopause showed a tendency to cluster in certain fragile X families, as a group, the premutated women experienced menopause earlier than noncarriers. This suggests that premature menopause may be the extreme effect of a spectrum of ovarian anomalies associated with the fragile X premutation.

The genus Phanaeus is included in the tribe Phanaeini, one of the most diverse tribes within the subfamily Scarabaeinae in terms of chromosomal characteristics. The effect of the bovine major histocompatibility complex (BoLA) locus on animal health may be due to a direct action of its alleles on immune functions, whereas its indirect effect on production traits might be explained by the better general health conditions of more productive animals. In the present study, the BoLA-DRB3 gene was investigated in 1058 cows belonging to seven Brazilian Gyr Dairy herds (Bos indicus, Zebu cattle). Comparative transcriptome analysis of the hippocampus from sleep-deprived and Alzheimer’s disease mice Genet. Mol. Biol.

Abstract We did a comparative analysis of the gene expression profiles of the hippocampus from sleep deprivation and Alzheimer’s disease (AD) mice. Differentially expressed genes (DEGs) were identified by comparing the transcriptome profiles of the hippocampus of sleep deprivation or AD mouse models to matched controls. The common DEGs between sleep deprivation and AD were identified by the overlapping analysis, followed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. The results showed that a total of 16 common DEGs showed similar change patterns in both sleep deprivation mice and AD mice. The individual response to diet may be influenced by gene polymorphisms. This study hypothesized that ADRB2 (Gln27Glu, rs1042714 and Arg16Gly, rs1042713), ADRB3 (Trp64Arg, rs4994) and GHRL (Leu72Met, rs696217) polymorphisms moderate weight loss. The study was a seven weeks dietary weight loss intervention with Brazilian adult obese women (n = 109). Genetic causes involved in Leishmania Chagasi infection innortheastern: Brazil Genet. Mol. Biol.

A sample of 502 individuals from 94 families from Jacobina, State of Bahia, Brazil, was investigated to determine the causal mechanisms involved in Leishmania chagasi (the causal agent of visceral leishmaniasis in the American hemisphere) infection, as measured by the intradermic reaction to antigens derived from this parasite, using complex segregation analyses. The results showed evidence of a major genetic mechanism acting on infection, with a frequency of a recessive (or additive) susceptibility gene (q) of approximately 0.45. A small multifactorial component (H = 0.29) acting in conjunction with a major recessive gene (q = 0.37) is not ruled out as a concomitant causative factor.

Three novel polymorphic microsatellite markers for the glaucoma locus GLC1B by datamining tetranucleotide repeats on chromosome 2p12-q12 Genet. Mol. Biol.

In order to identify new markers around the glaucoma locus GLC1B as a tool to refine its critical region at 2p11.2-2q11.2, we searched the critical region sequence obtained from the UCSC database for tetranucleotide (GATA)n and (GTCT)n repeats of at least 10 units in length. Three out of four potential microsatellite loci were found to be polymorphic, heterozygosity ranging from 64.56% to 79.59%. The identified markers are useful not only for GLC1B locus but also for the study of other disease loci at 2p11.2-2q11.2, a region with scarcity of microsatellite markers.

Characterization of ribosomal DNA (rDNA) in Drosophila arizonae Genet. Mol. Biol.

Ribosomal DNA (rDNA) is a multigenic family composed of one or more clusters of repeating units (RU). Each unit consists of highly conserved sequences codifying 18S, 5.8S and 28S rRNA genes intercalated with poorly conserved regulatory sequences between species. In this work, we analyzed the rDNA of Drosophila arizonae, a member of the mulleri complex (Repleta group). Using genomic restriction patterns, cloning and mapping of some representative rDNA fragments, we were able to construct a representative restriction map. RU in this species are 13.5-14 kb long, restriction sites are completely conserved compared with other drosophilids and the rDNA has an R1 retrotransposable element in some RU. We were unable to detect R2 elements in this species.
