Immunity to self co-generates regulatory T cells Nature Precedings

Immune responses to self are kept in check by tolerance mechanisms, including suppression by regulatory T cells (Tregs). The defective generation of Tregs specific for self-antigens may lead to autoimmune disease. We identified a novel population of human CD4^+^ Tregs, characterized by high surface expression of CD52, which is co-generated in response to autoantigen. Blood CD4^+^CD52^hi^ T cells were generated preferentially in response to low-dose autoantigen and suppressed proliferation and interferon-[gamma] production by other T cells. Depletion of resting CD4^+^CD52^hi^ T cells enhanced the T-cell response to autoantigen. Using microarray analysis, we showed up-regulation of Toll-like receptors 1,2,4,7,8, NF-[kappa][BETA], TNF-[alpha], p38-MAPK and MHC molecules in human peripheral blood mononuclear cells following infection with Plasmodium falciparum. We report herein further studies based on time-course microarray analysis with a focus on malaria-induced host immunity. The effect of metrifonate on the immunity of the exposed fish are evaluated through the determination of the antibody titer and mortality rate during the challenge trial. In the chronic toxic exposure to metrifonate by dose 0.17mg/liter for 8 weeks, the more clear signs were darkness of fish color with hemorrhagic patches & loosing of scale at end of the trial the color return to normal. Determination of some blood parameters revealed significant decrease of white blood cell count, as there were lymphopenia and nueutrophilia during the chronic exposure to metrifonate. The innate immune activity of the freshwater turtle Phrynops geoffroanus (Schweigger, 1812) was investigated, using a sheep-red-blood cell hemolysis assay. The time- and concentration-dependent hemolytic activity of the turtle plasma was low compared to that reported for other reptiles. However the plasma of P. geoffroanus exhibited higher activity at elevated temperatures, resulting in temperature-dependent hemolysis. The sensitivity of turtle plasma to temperature could be interpreted as a mechanism by which freshwater turtles use basking behavior to elevate body temperature, thus enhancing the innate immune response. This study evaluated the pathogenic characteristics of bacteria isolated from Litopenaeus vannamei during an outbreak at the Laboratory of Marine Shrimp, UFSC, Santa Catarina State, Brazil. Their virulence potential in larvae and juvenile shrimp and the effects on the total haemocyte count, phenoloxidase activity and serum agglutinate titre were examined after experimental infection. Bacterial strains were isolated from larvae and adult shrimps, identified by the AP120E biochemical system as: two strains of Vibrio alginolyticus, three of Aeromonas salmonicida and one of Pasteurella multocida sp. and Pasteurella sp. Then, the effects of crude propolis powder (CPP) on growth, hemato-immune parameters of the Nile tilapia, as well as its effects on resistance to A. hydrophila challenge were investigated. Abstract: This study investigated the effects of stocking density and dietary supplementation of probiotic (Lactobacillus) on growth, traits of carcass, lymphoid organs and intestinal microbial count of Japanese quail. Quail chicks (one week old) were randomly allotted to 6 groups in a 2×3 factorial experiment included 2 levels of stocking density (100, and 75 cm2/bird) and 3 levels of probiotic (0.00, 0.02 and 0.04 g/kg diet). Quail reared at 100 cm2/ bird showed significantly the highest body weight and daily gain. Quail reared at 75 cm2/ bird had significantly the lower average of feed consumption and the best ratio of feed conversion. The thymic morphometry analysis was used for determining apoptosis and atrophy of the thymus of eight puppies inoculated with canine distemper virus (CDV). Three healthy dogs were used as negative controls. Sections, 5µm thick, were stained by HE and Shorr, and the latter were evaluated by morphometry. CDV nucleoprotein was detected by immunohistochemistry. Morphometric results confirmed lymphoid hypotrophy in CDV inoculated dog thymuses, more stroma, less parenchyma and higher apoptotic index/field than negative control (not inoculated) puppies. Apoptosis plays a role in the mechanism of thymus atrophy that develops in canine distemper.

The aim of this study was to evaluate the immune response of hybrid surubim vaccinated by intraperitoneal injection and by immersion against Aeromonas hydrophila. Vaccine was prepared with formalin-inactivated A. hydrophila. Forty eight fishes (47±9g) were distributed in 12 tanks (4 fish / tank) in the following treatments (4 tanks / treatment): unvaccinated fish, vaccinated intraperitoneally and vaccinated by immersion. After 21 days, intraperitoneally vaccinated fishes showed the highest values of total protein and immunoglobulins, agglutination titer and serum antimicrobial activity. This study investigated the adjuvant potential of W/O/W multiple emulsions and microemulsions, comparing them with traditional aluminum hydroxide and oil-in-water emulsion adjuvants against bluetongue vaccine (BTV). Local inflammatory reactions were assessed in rabbits by measuring the temperature of the animals and the skin thickness at the site of application. Antibodies titers were determined by serum-neutralization test. Histological analyses of lesions at the site of adjuvants application were done. The hallmark of chronic Chagas' disease cardiomyopathy (CCC) is the finding of a T cell-rich inflammatory mononuclear cell infiltrate in the presence of extremely few parasites in the heart lesions. The scarcity of parasites in affected heart tissue casts doubt on the direct participation of Trypanosoma cruzi in CCC heart tissue lesions, and suggests the possible involvement of autoimmunity.
