Chromatin differentiation between Theobroma cacao L. and T. grandiflorum Schum Genet. Mol. Biol.

A comparative analysis of mitotic chromosomes of Theobroma cacao (cacao) and T. grandiflorum (cupuaçu) was performed aiming to identify cytological differences between the two most important species of this genus. Both species have symmetric karyotypes, with 2n = 20 metacentric chromosomes ranging in size from 2.00 to 1.19 µm (cacao) and from 2.21 to 1.15 mm (cupuaçu). The interphase nuclei of both species were of the arreticulate type, displaying up to 20 chromocentres, which were more regularly shaped in cacao than in cupuaçu. Abstract Pollutants have adverse effects on human health and on other organisms that inhabit or use water resources. The aim of the present study was to assess the environmental quality of three watercourses in Monte Carmelo, MG, Brazil, using the micronucleus test on Tradescantia. For each treatment, 15 plants were exposed to water samples for 24 h. The control group was exposed to formaldehyde (0.2%) and the negative control to Hoagland solution. Subsequently the plants were placed in Hoagland solution for 24 h to recover. Cells were stained with 2% acetic carmine and examined by light microscopy. Identification in silico of putative damage responsive elements (DRE) in promoter regions of the yeast genome Genet. Mol. Biol.

We report an in silico analysis to identify nucleotide sequence motifs in DNA repair genes that may define a binding site for regulatory proteins during the induction of those genes by mutagens. The damage responsive elements (DRE) weight matrix generated in this analysis was used to search for homologous sequences in the promoter region of all genes, including putative gene and hypothetical open reading frames (ORFs), in the Saccharomyces Genome Data Base (SGD). Abstract Carbonic anhydrases (CA) belong to the family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide to bicarbonate. In the present work, we characterized the cDNAs of four Paracoccidioides CAs (CA1, CA2, CA3, and CA4). In the presence of CO2, there was not a significant increase in fungal ca1, ca2 and ca4 gene expression. The ca1 transcript was induced during the mycelium-to-yeast transition, while ca2 and ca4 gene expression was much higher in yeast cells, when compared to mycelium and mycelium-to-yeast transition. Wild cassava, Manihot spp.: Biology and potentialities for genetic improvement Genet. Mol. Biol.

Wild species of Manihot are progenitors of cassava. They constitute valuable genetic reservoirs presenting genes that show new characters. Screening of these species showed some of them to have a notably high percentage of protein combined with a low percentage of hydrocyanic acid. Study of natural habitats revealed resistance to drought and excessive soil aluminum toxicity as well as adaptation to low temperature. Some of the hybrids obtained showed high root productivity and resistance to stem borers. Apomixis was discovered in the wild and transferred successfully to the cultivate species.

The expression of AtchitIV gene was analysed in Arabidopsis plants submitted to abiotic stresses. Transcript accumulation was detected in leaves in response to UV light exposure, exogenous salicylic acid administration and wounding. Transgenic Arabidopsis plants carrying AtchitIV promoter::gus fusion also showed differential expression of the reporter gene in response to these treatments. The AtchitIV expression was also analysed during Arabidopsis embryo development. GUS assay demonstrated AtchitIV promoter activation in zygotic embryos from torpedo stage up to full maturation. This study assessed the abundance of microsatellites, or simple sequence repeats (SSR), in 19 Eucalyptus EST libraries from FORESTs, containing cDNA sequences from five species: E. grandis, E. globulus, E. saligna, E. urophylla and E. camaldulensis. Overall, a total of 11,534 SSRs and 8,447 SSR-containing sequences (25.5% of total ESTs) were identified, with an average of 1 SSR/2.5 kb when considering all motifs and 1 SSR/3.1 kb when mononucleotides were not included. Dimeric repeats were the most abundant (41.03%), followed by trimerics (36.11%) and monomerics (19.59%). The scale number in lateral sets (SNS) of Mugil sp. (Mugilidae, Perciformes) collected in the lagoon-estuarine region of Cananéia, State of São Paulo ranges from 33 to 39. Electrokinetic, kinetic and thermostability properties of lactate dehydrogenase (LDH) were tested to determine if individuals with different SNS correspond to different species or populations of mullet. As in many other teleosts, LDH-A*, LDH-B*, and LDH-C* loci were detected. Through a two-fold serial dilution method applied to 10 different tissues of Mugil sp., a bidirectionally divergent expression of these loci was suggested. Abstract The plant genes encoding ABCGs that have been identified to date play a role in suberin formation in response to abiotic and biotic stress. In the present study, 80 ABCG genes were identified in ‘Dangshansuli’ Chinese white pear and designated as PbABCGs. Based on the structural characteristics and phylogenetic analysis, the PbABCG family genes could be classified into seven main groups: classes A-G. Segmental and dispersed duplications were the primary forces underlying the PbABCG gene family expansion in ‘Dangshansuli’ pear. Effect of gamma-radiation and sodium azide on quantitative characters in rice (Oryza sativa L.) Genet. Mol. Biol.

Seeds of rice cultivar IAC-1246 received single and combined treatments of 10 or 20 Krad gamma-rays and 0.5 mM sodium azide (SA). The experiments were carried out to assess the effect of treatments on the mean and variance in second generation plants of the following quantitative traits: number of days to flowering (NDF), culm length (CL) and tiller number (TN). In general, the mutagenic treatments increased variance, but did not change the mean for the characters NDF and CL in the M2 generation. There was no increase in the mean or variance of TN. To understand the phylogenetic position of Larimichthys polyactis within the family Sciaenidae and the phylogeny of this family, the organization of the mitochondrial genome of small yellow croaker was determined herein. The complete, 16,470 bp long, mitochondrial genome contains 37 mitochondrial genes (13 protein-coding, 2 ribosomal RNA and 22 transfer RNA genes), as well as a control region (CR), as in other bony fishes. Anteaters belong to the Order Xenarthra / Family Myrmecophagidae and are the only members without teeth. There are three genera with four living species in the family Myrmecophagidae: Myrmecophaga tridactyla (giant anteater), Tamandua tetradactyla (southern lesser anteater), Tamandua mexicana (northern lesser anteater), and Cyclopes didactylus (silky anteater). The karyotypes of M. tridactyla (2n = 60), T. tetradactyla (2n = 54) and C. didactylus (2n = 64) have already been described. In the present paper, three female and two male specimens of giant anteater and one lesser anteater male were analyzed. Isoenzymatic variation in the germplasm of Brazilian races of maize (Zea mays L.) Genet. Mol. Biol.

There are more than 200 races of maize (Zea mays L.) divided into three groups (ancient commercial races, the recent commercial races, and indigenous races). Although the indigenous races have no commercial value, they have many important characteristics which can be incorporated into maize breeding programs. Most Brazilian indigenous germplasm race stocks were collected at least 40 years ago, and nothing is known of the genetic variability present in this germplasm. The genetic variability was assayed in 15 populations from four indigenous races of maize (Caingang, Entrelaçado, Lenha and Moroti) and five indigenous cultivars, using five isoenzymatic systems encoded by 14 loci. Aberrant methylation of CpG islands located in promoter regions represents one of the major mechanisms for silencing cancer-related genes in tumor cells. We determined the frequency of aberrant CpG island methylation for several tumor-associated genes: DAPK, MGMT, p14ARF, p16INK4a, TP73, RB1 and TIMP-3 in 55 brain tumors, consisting of 26 neuroepithelial tumors, 6 peripheral nerve tumors, 13 meningeal tumors and 10 metastatic brain tumors. Aberrant methylation of at least one of the seven genes studied was detected in 83.6% of the cases. Structural organization of polygalacturonase-encoding genes from Penicillium griseoroseum Genet. Mol. Biol.

The pectinolytic system of Penicillium griseoroseum has been studied as a model to investigate aspects of gene organization in filamentous fungi. Here we show that the endopolygalacturonase-coding genes previously isolated exist as single copies in the fungus genome. DNA blot analysis revealed the presence of corresponding genes in other Penicillium species, although only one or two genes were found in opposition to the endoPG gene family reported for other filamentous fungi.
