Phylogeny and phylogeography of Atlantic oyster species: evolutionary history, limited genetic connectivity and isolation by distance ArchiMer

The phylogenetic relationships between naturally occurring Atlantic Crassostrea oyster species were inferred through analyses of mitochondrial (cytochrome oxidase subunit I and 16S) and nuclear (second internal transcribed spacer) sequences. We also scored 15 allozyme loci on 422 oysters to study population structuring of C. rhizophorae and C. brasiliana along 9000 km of the Western Atlantic coastline. Despite morphological similarities, C. virginica was genetically more closely related to C. rhizophorae than to C. brasiliana. La production française d' huître creuse Crassostrea gigas représente, avec 144000 tonnes de production, la 4ème' place au ni veau mondial après la Chine (2 285 000 t), le Japon (225 000t.) et la Corée (1 85 000t.). Complex interactions between oyster, environment and pathogens were observed during the MOREST (2001-2005) project on summer mortality events of Crassostrea gigas oysters in France. The talk will show how nutrition level increasing reproductive intensity generates a risk for pathogen infection under synergistic effect of temperature and stress. Moreover, a genetic component is associated to specific reproductive performances and spawning behaviour. Expression of candidate genes related to metabolism, immunity and cellular stress during massive mortality in the American oyster Crassostrea virginica larvae in relation to biochemical and physiological parameters ArchiMer

Quantification of mRNA of genes related to metabolism, immunity and cellular stress was examined in relation to a massive mortality event during the culture of American oyster larvae, Crassostrea virginica which was probably, in regard to previous microbiological analysis, induced by vibrio infection. To document molecular changes associated with the mortality event, mRNA levels were compared to biochemical and physiological data, previously described in a companion paper. Mitochondrial and microsatellite genetic differentiation of the flat oyster Ostrea edulis along the european coasts, and comparison with allozyme data ArchiMer

Natural populations of the European flat oyster, Ostrea edulis, have been harvested since Roman times. This species is distributed along the European Atlantic coasts, from Norway to Morocco, as well as the Mediterranean Sea and Black sea coasts. Previous studies, based on allozymes, have concluded that isolation by distance is taking place in the Mediterranean sea and the Atlantic ocean. Because of the presumed non neutrality of these markers, another approach was made to examine the genetic differentiation of this species using nuclear microsatellite and mitochondrial markers, both considered as neutral, and to compare the results to those observed with allozymes.

Molecular identification and expression of the phosphoglucomutase (PGM) gene from the Pacific oyster Crassostrea gigas ArchiMer

Phosphoglucomutase is a key enzyme in glycolysis and has been widely studied in vertebrates and some invertebrates but no molecular information is available in marine invertebrates despite the importance of this marker in ecological and genetical studies. In this work, we isolated a cDNA and the corresponding genomic sequence that encode PGM-2 locus in the Pacific oyster Crassostrea gigas. We used sequences drawn from the database to construct an evolutionary framework for examining the position of mollusc PGM sequences among prokaryotic and eukaryotic homologues and showed that oyster PGM gene organization was closer to vertebrates PGM genes than other invertebrates as previously found in other Lophotrochozoa species. Phylogéographie des huîtres de mangrove de l'Océan Atlantique Sud : C. gasar et C. rhisophorae ArchiMer

Mangrove oysters include various species that are also present on different coasts. In the southern part of the Atlantic Ocean, Crassostrea rhizophorae has been observed along the southern coast of America and Crassostrea gasar along the western coast of Africa. Fifteen populations of these oysters were sampled from these two geographic areas. Their polymorphism was studied on the 16S mitochondrial fragment both by par sequencing and RFLP analysis. Some samples' caryotype were also analysed. Two haplotypes were identified: C. gasar was found in Africa, but also for the first time in South America, whereas C. rhizophorae was only found on the coasts of South America.

The European flat oyster, Ostrea edulis, occurs naturally from Norway to Morocco in the North-Eastern Atlantic and throughout the Mediterranean. The oyster has been a harvested species for at least 6000 years. However, it successively suffered from over harvesting and then from two protozoan diseases during the 1960s and its production has drastically decreased in Europe from that time. Evidence for the presence of the Portuguese oyster, Crassostrea angulata, in northern China ArchiMer

Although these two taxa were both introduced from Asia into Europe. one (C. gigas) was voluntarily introduced in the early 1970s, whereas the other (C. angulata) was presumed to be present in Europe for at least four centuries, but nearly disappeared because of disease. An amylase gene polymorphism is associated with growth differences in the Pacific cupped oyster Crassostrea gigas ArchiMer

This study investigated the non-neutrality of genetic polymorphism in two alpha-amylase genes (AMYA and AMYB) in the oyster Crassostrea gigas. Bi-parental oyster families, bred to be polymorphic for markers in these genes, were monitored for growth and survival for 1 year under standard culture conditions in two French production sites. Within-family genotype frequencies indicated that the two amylase genes were closely linked (c. 1.7 cM). Ils permettent désormais l'analyse approfondie et la discrimination fine de populations d'huîtres creuses Crassostrea gigas et de rechercher si certaines d'entre-elles n'auraient pas des caractéristiques intéressantes pour l'ostréiculture locale (croissance, résistance estivale, affinage ... ). Association among growth, food consumption-related traits and amylase gene polymorphism in the Pacific oyster Crassostrea gigas ArchiMer

To examine further a previously reported association between amylase gene polymorphism and growth in the Pacific oyster Crassostrea gigas, ecophysiological parameters and biochemical and molecular expression levels of alpha-amylase were studied in Pacific oysters of different amylase genotypes. Genotypes that previously displayed significantly different growth were found to be significantly different for ingestion and absorption efficiency. These estimated parameters, used in a dynamic energy budget model, showed that observed ingestion rates (unlike absorption efficiencies) allowed an accurate prediction of growth potential in these genotypes. Reproductive effort and growth in Crassostrea gigas: comparison of young diploid and triploid oysters issued from natural crosses or chemical induction ArchiMer
