Members of the archaeal order Methanomassiliicoccales are methanogens mainly associated with animal digestive tracts. However, environmental members remain poorly characterized as no representatives not associated with a host have been cultivated so far. In this study, metabarcoding screening combined with quantitative PCR analyses on a collection of diverse non-host-associated environmental samples revealed that Methanomassiliicoccales were very scarce in most terrestrial and aquatic ecosystems. Relative abundance of Methanomassiliicoccales and substrates/products of methanogenesis were monitored during incubation of environmental slurries. A sediment slurry enriched in Methanomassiliicoccales was obtained from a freshwater sample. It allowed the... |

The possibility of cultivation of the giant kelp Macrocystis pyrifera was successfully experienced in France on the north coast of Brittany. In order to evaluate the possibility of eventual acclimation on the French shore, study tour was undertaken on the Californian and Chilian coasts on main kelp beds. Effects of such beds on general ecology, fish resources, sportfishing, navigation, beach cleanliness, and alginates industry are analyzed. Factors controlling kelp distribution and hazards of wide distribution are discussed. Macrocystis seems to be able of developing and maintaining a large standing crop and to contribute to a large and stable associated fauna. The advantage of enhancing fishing greatly outweight the minor disadvantage of navigational... |

A study was conducted in the subtropical area of Southern Brazil to determine the survival of pathogens in soybean residues under conventional and no-tillage cultivation systems from March to September of 1998 and 1999. The pathogens most frequently isolated were Colletotrichum truncatum, Phomopsis spp., Cercospora kikuchii, Fusarium spp., Macrophomina phaseolina, and Rhizoctonia solani. Other fungi isolated were Myrothecium roridum, Penicillium sp., Chaetomium sp., Epicoccum sp., Corynespora cassiicola and Trichoderma sp. The percent of survival of each pathogen varied according to the month and the year. Survival of C. truncatum, Phomopsis spp. and C. kikuchii were significantly reduced (p<0.05) from the first to the last evaluation either on buried... |

ABSTRACT The hydroponic growth system is a convenient platform for studying whole plant physiology, especially for root morphological and nutritional analysis. However, we found that most hydroponic systems described in the literature are not suitable for citrus plants. In this study, a hydroponic system for citrus was designed, comprising three principal components: power and time switch, aeration and hydroponic culture. Herein, details of the protocol were described, including equipment setup, seed pregermination and cultivation, together with preparation and transfer of nutrient solution into hydroponics. In order to demonstrate the adaptability of the trifoliate orange plant to our hydroponic system, comparative tests between soil- and... |

The aim of this study was the study of the abiotic environment of the carnivorous pitcher plant Heliamphora nutans (Sarraceniaceae), including the microclimate and the geochemistry of the soil of the growing sites on Roraima Tepui and discuss their relevance within the recent model of carnivorous plant ecology. The soil was peaty and low in nutrients. The microclimate on the site was very balanced, with moderately cool temperatures, a constant high humidity and very low wind speed. Heliamphora was not exposed to any recognizable climatic stress. Previous macroclimatic measurements reflected the growth conditions of Heliamphora only incorrectly, since humidity decreased drastically with height. The apparent conflict with the common model of carnivorous... |

With the advent of recombinant DNA technology, recombinant protein expression has become an important tool in the study of the structure, function and identification of new proteins, especially those with therapeutic functions. Escherichia coli has been the predominant prokaryote used in genetic engineering studies due to the abundance of information about its metabolism. Despite significant advances in molecular biology and immunology of infections, there are as yet no prophylactic drugs capable of preventing visceral leishmaniasis. It is therefore important to identify specific antigens in order to develop vaccines and diagnostic kits against this disease. The objective of this study was to evaluate the influence of culture medium on the production of... |

The objective of this work was to study the effect of caffeine and tannins on Pleurotus sp. cultivation and to evaluate the feasibility of using coffee husks as substrate for mushroom cultivation. Eight strains of P. ostreatus and two strains of P. sajor-caju were screened on a medium prepared from agar extract of coffee husk. Based on best mycelial growth and biomass production, the strain P. ostreatus LPB 09 was selected for detailed studies. With the increase of caffeine concentration, the mycelial growth and the biomass production decreased, and no growth was observed when concentration of caffeine was 2500 mg/L. Furthermore, Pleurotus did not degrade the caffeine, but absorbed it. Tannin under 100 mg/L in the medium stimulated the growth of mycelia,... |

Cordyceps is a fastidious pathogenic fungus infecting insects, and recent years have witnessed rapid progress in its medical properties. In this study, a wild isolate, C. cicadae MP12, was characterized through in vitro cultivation and its nuclear small-subunit (SSU) ribosomal DNA (rDNA) data. In vitro culture of C. cicadae MP12 was established by growing its fruiting bodies in a solid matrix. C. cicadae MP12 was inoculated into Cryptotympana atrata cicada pupae for in vivo culture, where the fungi developed its fruiting body as well. The contents of adenosine and cordycepin in dried fruiting bodies after culture were 1421.45µg/g and 1398.12 µg/g, respectively. Therefore, the established cultures from this study could be used for the production of various... |
