Ethyl acetate fraction from Angelica sinensis inhibits IL-1β-induced rheumatoid synovial fibroblast proliferation and COX-2, PGE2, and MMPs production Biol. Res.
Lee,Won-Seok; Lim,Jin-Han; Sung,Myung-Soon; Lee,Eun-Gyeong; Oh,Yoo-Jeong; Yoo,Wan-Hee.
BACKGROUND: The root of Angelica sinensis (AS), also known as "Dang-gui," was a popular herbal medicine widely used in the treatment of gynecological diseases in China, Korea, and Japan for a long time. This study aimed to determine the effects of ethyl acetate fraction from Angelica sinensis (EAAS) on the interleukin-1β (IL-1β)-induced proliferation of rheumatoid arthritis synovial fibroblasts (RASFs), and production of matrix metalloproteinases (MMPs), cyclooxygenase (COX) 2, and prostaglandin E2 (PGE2), involved in articular bone and cartilage destruction, by RASFs. RESULTS: RASF proliferation was evaluated with cholecystokinin octapeptide (CCK-8) reagent in the presence of IL-1β with/without EAAS. Expression of MMPs, tissue...

TAPCells, the Chilean dendritic cell vaccine against melanoma and prostate cancer Biol. Res.
Salazar-Onfray,Flavio; Pereda,Cristián; Reyes,Diego; López,Mercedes N.
Here we summarize 10 years of effort in the development of a biomedical innovation with global projections. This innovation consists of a novel method for the production of therapeutic dendritic-like cells called Tumor Antigen Presenting Cells (TAPCells®). TAPCells-based immunotherapy was tested in more than 120 stage III and IV melanoma patients and 20 castration-resistant prostate cancer patients in a series of phase I and I/II clinical trials. TAPCells vaccines induced T cell-mediated memory immune responses that correlated with increased survival in melanoma patients and prolonged prostate-specific antigen doubling time in prostate cancer patients. Importantly, more than 60% of tested patients showed a Delayed Type Hypersensitivity (DTH) reaction...

The soybean phytoestrogen, genistein, is increasingly consumed as an alternative therapeutic for age-related diseases, namely cardiovascular conditions, cancer and osteoporosis. However, despite the beneficial effects on health, concern has been raised that this isoflavone also acts as an endocrine-disrupting chemical. The aim of this study was to examine the effects of genistein on immunohistomorphometric features of pituitary adrenocorticotropic cells (ACTH) and blood concentrations of ACTH and corticosterone in orchidectomized middle-aged male rats. Sixteen-month-old Wistar rats were divided into sham-operated (SO), orchidectomized (Orx) and genistein-treated orchidectomized (Orx+G) groups. Genistein (30mg/kg/day) was administered subcutaneously for...

Structurally conserved C-RFa revealed prolactin releasing activity in vitro and gene expression changes in pituitary of seasonally acclimatized carp Biol. Res.
Romero,Alex; Montefusco-Siegmund,Rodrigo; Valenzuela,Guillermo; Kausel,Gudrun; Muller,Marc; Brauchi,Sebastian; Figueroa,Jaime.
Here we show the cloning and characterization of a novel homolog of prepro C-RFa cDNA from Cyprinus carpió. The deduced preprohormone precursor of 115 amino acids leads to a mature bioactive peptide of 20 amino acids with identical sequence to other teleost C-RFa. Modeling of the mature C-RFa peptide highlighted significant similarity to homologous human PrRP20, specifically the conserved amphipathic system defined by the C-terminal alpha-helix. Clearly, the synthetic C-RFa peptide stimulated prolactin release from primary cultured fish pituitary cells. For the first time, significant variation was shown in C-RFa mRNA and protein levels in the hypothalamus and pituitary between summer- and winter-acclimatized carp. Furthermore, C-RFa protein distribution...

Relationship between DAPI-fluorescence fading and nuclear DNA content: An alternative method to DNA quantification? Biol. Res.
GALLARDO-ESCÁRATE,CRISTIAN; ÁLVAREZ-BORREGO,JOSUÉ; VON BRAND,ELISABETH; DUPRÉ,ENRIQUE; DEL RÍO-PORTILLA,MIGUEL ÁNGEL.
In observations by confocal or conventional fluorescence microscopy, important factors should be considered in order to obtain accurate images. One of them, such as the fluorescence bleaching from highest intensity to lowest signal of fluorescence is a common problem with several DNA fluorochromes and especially for DAPI stain. The fluorescence of DAPI fades rapidly when it is exposed to UV light, under optimal conditions of observation. Although the fading process can be retarded using a mounting medium with antifading reagents, the photochemical process underlying the fluorescence decay has not yet been fully explained. In addition, no relationship between fluorescence fading and nuclear DNA content has been tested. In order to test this relationship, we...

Sepúlveda,Soledad J; Portella,Jimmy R; Noriega,Luis P; Escudero,Ernesto L; Noriega,Luis H.
The aim of this study was to review the experience and outcomes of assisted reproduction cycles with embryos grown up to day 5 of development, comparing different parameters according to the ages of the patients. We retrospectively studied 1,874 assisted reproduction cycles where embryo culture was extended up to the fifth or sixth day of development. All IVF and ICSI cycles were included, comparing, according to patient age, the following rates: blastocyst formation, pregnancy, implantation and abortion. As control, we analyzed cycles with donated oocytes from young donors (OD). The number of embryos reaching the blastocyst stage is similar in all groups of patients. Only the OD group was different in terms of blastocyst formation, pregnancy and...

The development of Alternaria alternata is prevented by chitinases and ß-1,3- glucanases from Citrus limon seedlings Biol. Res.
FANTA,NORA; ORTEGA,XIMENA; PÉREZ,LUZ M.
In addition to phytoalexin synthesis, the defense response of intact Citrus limon seedlings against Alternaria alternata involves both constitutive and induced enzyme activities such as chitinases (Ch) and ß-1,3-glucanases (Glu). A. alternata conidial germination was prevented by protein extracts from inoculated lemon seedlings, but also by extracts from mock-inoculated specimens. On the other hand, degradation of mycelia was accomplished only by protein extracts from inoculated seedlings. The presence of six Ch isoenzymes and of four Glu isoenzymes was detected in protein extracts from mock-inoculated seedlings. As a result of fungal inoculation, the isoenzyme pattern of Ch and Glu changed, making possible the detection of a new Ch isoenzyme and of three...

Chloroplast localization of Cry1Ac and Cry2A protein- an alternative way of insect control in cotton Biol. Res.
Muzaffar,Adnan; Kiani,Sarfraz; Khan,Muhammad Azmat Ullah; Rao,Abdul Qayyum; Ali,Arfan; Awan,Mudassar Fareed; Iqbal,Adnan; Nasir,Idrees Ahmad; Shahid,Ahmad Ali; Husnain,Tayyab.
BACKGROUND: Insects have developed resistance against Bt-transgenic plants. A multi-barrier defense system to weaken their resistance development is now necessary. One such approach is to use fusion protein genes to increase resistance in plants by introducing more Bt genes in combination. The locating the target protein at the point of insect attack will be more effective. It will not mean that the non-green parts of the plants are free of toxic proteins, but it will inflict more damage on the insects because they are at maximum activity in the green parts of plants. RESULTS: Successful cloning was achieved by the amplification of Cry2A, Cry1Ac, and a transit peptide. The appropriate polymerase chain reaction amplification and digested products confirmed...

Upregulation of lncRNA BANCR associated with the lymph node metastasis and poor prognosis in colorectal cancer Biol. Res.
Shen,Xiaogang; Bai,Yifeng; Luo,Bin; Zhou,Xiaogang.
Abstract Background Growing evidence has supported that long non-coding RNAs (lncRNAs) could play vital roles in the development, progression, and prognosis of colorectal cancer (CRC). However, little is known about the clinical significance of BRAF-activated non-coding RNA (BANCR) in CRC. The aim of this study is to explore the clinical value of lncRNA BANCR in CRC patients. Methods The expression of lncRNA BANCR was measured in 106 CRC tissues and 65 adjacent normal tissues using the quantitative real-time PCR. Results The study showed that lncRNA BANCR was highly expressed in CRC tissues compared with adjacent normal tissues (P < 0.001). In addition, high expression of lncRNA BANCR was positively correlated with the lymph node metastasis (P...

Growth differentiation factor 9 and bone morphogenetic protein 15 expression in previtellogenic oocytes and during early embryonic development of Yellow-tail Kingfish Seriola lalandi Biol. Res.
Palomino,Jaime; Herrera,Giannina; Dettleff,Phillip; Martínez,Víctor.
BACKGROUND: During fish oocyte maturation, specific molecules are expressed and accumulated within oocyte until fertilization and embryo development. Special attention have been paid in members of the transforming growth factor (TGF-β) superfamily; growth differentiation factor 9 (GDF9/gdf9) and bone morphogenetic protein 15 (BMP15/bmp15), which exert regulatory functions during oocyte maturation and follicle development. However, little attention has been paid to the involvement of these molecules during embryogenesis considering its importance for the formation of a good quality egg and subsequent embryo survival. The purpose of this study was to analyze the expression of gdf9 andbmp15 in previtellogenic oocytes and during early embryonic...

Nitric oxide is not involved in Neisseria gonorrhoeae-induced cellular damage of human Fallopian tubes in vitro Biol. Res.
GARCÍA,KATHERINE P; RUBILAR,PAULINA S; VARGAS,MACARENA F; CÁRDENAS,HUGO; RIOS,MIGUEL A; ORIHUELA,PEDRO A; VARGAS,RENATO H ; FUHRER,JUAN; HECKELS,JOHN E; CHRISTODOULIDES,MYRON; VELÁSQUEZ,LUIS A.
In the present study, we investigated whether cellular damage, as demonstrated by lactate dehydrogenase (LDH) release in the human fallopian tube (FT) infected by Neisseria gonorrhoeae (Ngo), correlated with high levels of nitric oxide synthase (NOS) mRNA and enzyme activity. Infection with Ngo induced a significant increase (~35-fold) in mRNA transcripts of the inducible isoform of NOS. Paradoxically, a reduction in NOS enzyme activity was observed in infected cultures, suggesting that gonococcal infection possibly influences translation of iNOS mRNA to the enzyme. In addition, treatment with the NOS inhibitor TRIM did not prevent gonococcal-induced cellular damage. In contrast, the addition of the inhibitor L-NAME induced a 40% reduction in LDH release,...

Analysis of 5382insC (BRCA1) and 6174delT (BRCA2) mutations in 382 healthy Chilean women with a family history of breast cancer Biol. Res.
JARA,LILIAN; AMPUERO,SANDRA; SECCIA,LORENA; BUSTAMANTE,MARIO; BLANCO,RAFAEL; OJEDA,JOSÉ MANUEL.
Breast cancer is the most common malignancy among women. Chilean studies reveal that this cancer presents the third highest mortality rate. A family history of breast cancer is one of the major risk factors for the development of this disease. BRCA1 and BRCA2 are the two main hereditary breast cancer susceptibility genes, and mutations in these genes are related to inherited breast cancer. In specific populations only some mutations have been found to be associated with susceptibility. The purpose of this study was to establish the frequency of 5382insC (BRCA1) and 6174delT (BRCA2) germline mutations in 382 healthy Chilean women with at least two relatives affected with breast cancer and in probands and their relatives from 8 high risk families for breast...

On cognitive ecology and the environmental factors that promote Alzheimer disease: lessons from Octodon degus (Rodentia: Octodontidae) Biol. Res.
Rivera,Daniela S.; Inestrosa,Nibaldo C.; Bozinovic,Francisco.
Cognitive ecologist posits that the more efficiently an animal uses information from the biotic and abiotic environment, the more adaptive are its cognitive abilities. Nevertheless, this approach does not test for natural neurodegenerative processes under field or experimental conditions, which may recover animals information processing and decision making and may explain, mechanistically, maladaptive behaviors. Here, we call for integrative approaches to explain the relationship between ultimate and proximate mechanisms behind social behavior. We highlight the importance of using the endemic caviomorph rodent Octodon degus as a valuable natural model for mechanistic studies of social behavior and to explain how physical environments can shape social...

Quantitation of diarrhetic shellfish poisoning toxins in Chilean Mussel using pyrenyldiazomethane as fluorescent labeling reagent Biol. Res.
GARCÍA,CARLOS; PEREIRA,PAULO; VALLE,LUIS; LAGOS,NÉSTOR.
Diarrhetic shellfish poisoning (DSP) is a gastrointestinal disease caused by lipid soluble polyether toxins produced by dinoflagellates and accumulated in shellfish. Diarrhetic shellfish poisoning is a worldwide threat to public health and the shellfish industry. To date, only four lipid soluble polyethers have been known as diarrhetic shellfish toxins. Among them, Okadaic acid (OA), Dinophysistoxin 1 (DTX-1, 35-methyl OA), Dinophysistoxin 2 (DTX-2, OA isomers) and Dinophysistoxin 3 (DTX-3, 7-O-acyl-35-methyl OA), all of which have free carboxilic groups. To perform quantitative analysis of DSP toxins in shellfish samples is a requirement, because DSP toxins are endemic in the Chilean mollusks of the southern regions, and although human symptoms of DSP...

Apical Maxi-chloride channel from human placenta: 12 years after the first electrophysiological recordings Biol. Res.
RIQUELME,GLORIA.
The Maxi-chloride channel was the first ion channel described by electrophysiological methods in placenta. Because it is difficult to access a complex epithelium such as the placenta for electrophysiological procedures, the studies of ion channels from placental membranes have been performed only very recently. It was only in 1993 that a direct demonstration of a high-conductance chloride channel in apical membranes of intact trophoblastic epithelium was mentioned, and two years later, the description of this channel was reported from purified placental apical membranes reconstituted into artificial lipid membranes suitable for patch-clamp recordings. This brief review comments on the work done with regard to the electrophysiological characterization and...

Abnormal Electrophysiological Activation in Schizophrenics during a Personal Traits Attribution Task Biol. Res.
SILVA,JAIME R; TORRES,WALTER M; ORTIZ,MANUEL S.
Changes in personal identity have been described as a major component in the expression of schizophrenia. Considering current evidence that relates the cortical structures of the medial line with the self, we hypothesize that there are differences in the frontomedial electrophysiological activation in schizophrenics when compared to normal subjects during tasks that require this function. In an experimental design, 9 schizophrenic and 7 normal subjects participated in a personal traits attribution task regarding the attribution of the self, another person and a simple reading. Electric brain activity was recorded with a dense array of 128 electrodes, with evoked brain potentials being obtained for the three conditions. Considering the evoked potential...
