Production and characterization of endo-polygalacturonase from Aspergillus niger in solid-state fermentation in double-surface bioreactor BABT

Endo-polygalacturonase (endo-PG) production by Aspergillus niger T0005/007-2 in solid medium with 170 mm of height was evaluated in a cylindrical double surface bioreactor in 96-h experiments. Cell concentration close to 92 mg.g -¹ dm (mg per g of dry medium) in the standard condition (static) was achieved, whereas in tests under forced aeration of 1.4 and 2.8 L.min-1. Kg-1 mm (L of air per minute per Kg of moist medium) and with the central shaft fungal biomass attained approximately 100 mg.g-1 dm. Superior endo-PG activity was obtained with the central-shaft system, 78 U.g-1 dm (units per g of dry medium). Effects of hydrolysis and digestion in vitro on the activity of bovine plasma hydrolysates as inhibitors of the angiotensin I converting enzyme BABT

The angiotensin I-converting enzyme (ACE) inhibiting activity of bovine plasma hydrolyzates obtained by Alcalase 2.4 L at different degrees of hydrolysis (DH) was evaluated. For the evaluation of ACE inhibition (ACEI), Hippuryl-His-Leu was used as substrate and the amount of hippuric acid liberated by non-inhibiting ACE was determined by spectrophotometry at 228 nm. The results showed that the enzymatic hydrolysis increased the ACEI activity as compared with the un-hydrolyzed plasma. The highest activity was onbtained with a DH of 6.7%. Concentration by membrane separation processes of a medicinal product obtained from pineapple pulp BABT

The concentration of pineapple juice is needed to retain the bromelain activity and to standardize the composition and proteolytic activity. Thus, this work aimed to obtain a pure bromelain extract from the Ananas comosus L. Merril juice by membrane separation process. A 2² experimental planning was used to study the influence of pH and transmembrane pressure on the activity recovery by micro-filtration using a plain membrane. In second step, this enzyme was purified by the ultra-filtration using a 10 kDa millipore kit. Some Biochemical properties of polyphenoloxidase from spearmint (Mentha arvensis) BABT

Polyphenoloxidase (PPO; EC 1.14.18.1) extracted from Mentha arvensis leaves was isolated by (NH4)2SO4 precipitation and extensive dialysis. Its optimum pH and temperature varied with the substrate. The PPO showed activity with various diphenols. Km values were found 0.825, 0.928 and 7.41mM for caffeic acid, 4-methylcatechol and catechol, respectively. On heat-inactivation, half of the activity was lost after 60 and 15 sec at 70 and 75ºC, respectively. Measuring of residual activity showed a stabilizing effect of sucrose at various temperatures with activation energy (Ea) for inactivation increasing with sucrose concentration from 0 to 40% (w/w). Performance and macrophage activity of broilers fed with a sorghum meal with different yeast wall levels BABT

The aim of this work was to evaluate the effect of broiler breeders' age and yeast wall (YW) levels on broilers' performance and macrophage activitys. The experiment consisted in a completely randomized design and a 2 X 5 factorial arrangement and two controls of two broiler breeders age (34 and 57 weeks of age) and five YW levels (zero, one, two, three or four kg of YW/ton of diet). They received sorghum diet compared to a control corn/soybean meal diet. The age of the broiler breeders influenced the performance, but did not affect macrophage activity. Saccharomyces uvarum mannoproteins stimulate a humoral immune response in mice BABT

Yeasts discarded in industrial processes can be used as a nutritional supplement and to extract cellular components with biotechnological aims. In this study, the humoral immune response of Swiss mice treated with mannoproteins (MP) from the yeast Saccharomyces uvarum was evaluated. The mice were treated with MPs at different doses and times and inoculated with 2% sheep red blood cells. An increase in total Ig in mice treated with 100 μg of MP at the time of immunization or 24 h before was observed in the primary immune response; in the secondary immune response, an increase was observed in total Ig values for all groups, and an increase of IgG was observed in the mice treated with MPs (100 μg) at the time of immunization or 24 h before. Application of an Enzymatic Extract from Aspergillus niger as Coagulant for Cheddar Cheese Manufacture BABT

Abstract The coagulation of milk by a serin protease from Aspergillus niger NRRL3 was studied by rheology. Cheddar-type cheese was manufactured using 3.5% (v/v) of fungal enzymatic extract and fermentation-produced chymosin was used as control coagulant. Full composition and ripening of both kinds of Cheddar cheese were studied. Differences in the proteolysis of caseins, not only during cheese manufacture but also during ripening, affected cheese composition, texture and peptide profile. Microbial development during ripening was not affected by the coagulant used.

A Mathematical Model for the Removal of Organic Mater in Stabilization Ponds BABT

This work presents an application to systemize the construction of ponds systems for treatment of domestic sewage. It consisted of two anaerobic ponds operated in parallel during May/97 to April/99. These were connected in series with a chicaned facultative pond. The treatment system was controlled with samples collected from the crude sewage (compound sample), in the affluents and effluents of the ponds and along the flux of the anaerobic and facultative ponds. The following parameters were analyzed: pH, Biochemical Oxygen Demand, Chemical Oxygen Demand, Total Solids, Sedimentable Solids, Total Coliforms, Oxygen Consumed in Acid Medium (OCAM) and temperature.

Physico-chemical and sensorial evaluation of sugarcane spirits produced using distillation residue BABT

The objective of the present study was to analyze the use of vinasse from cachaça as an ingredient of the fermentation medium for the spirit production. The fermentations were conducted out in three successive batches using a Saccharomyces cerevisiae isolate. In the first batch, the sugarcane broth was only diluted with distilled water. In the second and third batches, the fermentations were carried out using the cane broth diluted with vinasse from the distillation of the sugarcane wines of the first and second batches, respectively at a concentration of 10% (v/v). Phenolic compounds and antioxidant activity of rice BABT

The aim of this work was to study the phenolic compounds identified in rice, their antioxidant activity and their potential beneficial effects on health. In vitro and in vivo studies evaluating the rice grains with different pericarp colour (light brown, red and black) showed potential beneficial effects on health related to the polyphenol content of the grain, such as reduction of oxidative stress, aid in the prevention of cancer, cardiovascular diseases and complications of diabetes, among others.

Fetal mummification in silky shark (Carcharhinus falciformis) from the Gulf of California, Mexico BABT

The aim of this work was the description of fetal mummification in silky shark. During July of 2001 in Santa Maria fishing camp, Gulf of California Mexico, fourteen pregnant female of silky sharks were captured. In all of them the embryos were found to be dehydrated. However, total observations were possible in only two litters. Six embryos in each analyzed litters were found. All the embryos were mummified, complete and without maceration. Because the litters were totally mummificated in several females, it was suggested that fetal mummification was produced by the environmental condition.

The objectives of this work were to describe the development and growth rate of the pine cones of Araucaria angustifolia during and after the pollination period, to identify the period of maximum growth of the cones and the period of pollination and maturation of the pine nuts. The adult individuals were found at the Setor de Ciencias Agrarias of the UFPR, in Curitiba - PR. Collections and measurements of pine cones were made in 2003 and 2005. The diameter and the length of the pine cones were measured 15 different times in both the years. This paper proposes a new feature, the Global and Local-Oriented Gabor Texture Histogram (GLOGTH), for person re-identification. Gastric cancer (GC) is a multifactorial disease with a high mortality rate in Brazil and worldwide. This work aimed to evaluate single nucleotide polymorphisms (SNP) rs1695, in the Glutathione S-Transferase Pi (GSTP1) gene in GC samples by comparative analysis Specific PCR - ASP and Dideoxy Single Allele-Specific PCR - DSASP methods. The DSASP is the proposed new method for allelic discrimination. This work analyzed 60 GC samples, 26 diffuse and 34 intestinal types. The SNP rs1695 of the GSTP1 gene was significantly associated with GC analyzed by DSASP method (χ2 = 9.7, P < 0.05). Major soil classes of the metropolitan region of Curitiba (PR), Brazil: I - mineralogical characterization of the sand, silt and clay fractions BABT

The aim of this work was to evaluate the mineralogical and chemical characteristics of most representative soils of the Region of Curitiba, Paraná State. Samples were collected at different depths. The results showed: (a) the quartz was the only identified mineral at the silt and sand fractions. The dominant clay mineral was Kaolinite, with contents ranging from 676.7 to 820.8 g kg-1. Molecular Cloning and Sequencing of AlkalophilicCellulosimicrobium cellulans CKMX1 Xylanase Gene Isolated from Mushroom Compost and Characterization of the Gene Product BABT

ABSTRACT A xylanolytic bacterium was isolated from mushroom compost by using enrichment technique. Results from the metabolic fingerprinting, whole-cell fatty acids methyl ester analysis and 16S rDNA sequencing suggested the bacterium to be Cellulosimicrobium cellulans CKMX1. Due to the xylanolytic activity of this bacterium, isolation and characterization of the xylanase gene were attempted. A distinct fragment of about 1671 bp was successfully amplified using PCR and cloned into Escherichia coli DH5α. A BLAST search confirmed that the DNA sequence from the amplified fragment was endo-1, 4-beta-xylanase, which was a member of glycoside hydrolase family 11. An Alternative Inactivant for Rift Valley Fever Virus using Cobra Venom-derived L-Amino Oxidase, which is Related to its Immune Potential BABT

ABSTRACT Vaccine improvement depends on the formulation, adjuvant type and inactivant used. The type of formulation may interfere with immunogenicity. The present work aimed to evaluate the inactivation activity and related immune potential of the Cobra venom-derived LAO enzyme compared to the currently used inactivants (BPL and formalin) for both animal and human vaccines. The RVF virus was completely inactivated within 6 hrs, 4 hrs and 2 hrs after treatment with Formalin, LAO and BPL, respectively. Properties of a biosurfactant produced by Bacillus pumilus using vinasse and waste frying oil as alternative carbon sources BABT

Biosurfactants are chemical molecules produced by the microorganisms with potential for application in various industrial and environmental sectors. The production parameters and the physicochemical properties of a biosurfactant synthesized by Bacillus pumilus using different concentrations of vinasse and waste frying oil as alternative carbon sources were analyzed. The microorganism was able to grow and produce a biosurfactant using both the residues. The surface tension was reduced up to 45 mN/m and the maximum production of crude biosurfactant was 27.7 and 5.7 g/l for vinasse and waste frying oil, respectively, in concentration of 5%. Effect of formulation on the quality of frozen bread dough BABT

The main objective of this work was to determine the influence of formulation on the stability of bread dough during frozen storage. Bread doughs containing gluten and trehalose were submitted to mechanical freezing at -30° C and stored frozen for 45 days. Two types of instant yeast were tested: (A) for sweet doughs and (B) for savoury doughs. Specific volume was significantly affected by the yeast type, type A showing better effect than type B. Frozen storage of the doughs negatively affected the specific volume, crumb hardness and technological score of the bread. The addition of 10% trehalose had a beneficial effect on the cell survival rate for both the yeasts.
