Many therapeutic, diagnostic and prophylactic procedures used in hospitals are of potential genetic risk. An evaluation was made of genotoxic occupational risk in 42 workers from the Hospital de Clínicas de Porto Alegre, RS, Brazil, who had been occupationally exposed to lead (solder), ethylene oxide (sterilization area), antineoplastic drugs (nurses and pharmacists) or ionizing radiation. They were compared with 42 unexposed individuals. There was an increase in the frequency of binucleated cytochalasin-blocked lymphocytes with micronuclei, though it was not significant (P = 0.058). The effects of an animal RNase fused to the late cotton pollen-specific promoter G9 in a plant system were investigated. Expression of the chimeric genes G9-uidA and G9-RNase in tobacco plants showed that the 1.2-kb promoter fragment of the G9 gene was sufficient to maintain tissue and temporal specificity in a heterologous system. GUS (beta-glucuronidase) expression was detected only in pollen from anther stage 6 through anthesis, with maximal GUS activity in pollen from stage 10 anthers. The stingless bee species Melipona quadrifasciata includes two subspecies, Melipona quadrifasciata anthidioids and Melipona quadrifasciata quadrifasciata. The morphological difference between the two subspecies is the presence of three to five continuous yellow stripes on the terga on the 3rd to 6th segments in workers and males of M. q. quadrifasciata, and two to five interrupted bands in M. q. anthidioides. Maize (Zea mays L.) microsatellite loci are useful as genetic markers because they are numerous, occur in every chromosome, and have a high content of polymorphism information. Furthermore, they can be amplified by PCR and the resulting fragments resolved on agarose gels. Heterodera glycines Ichinohe, the soybean cyst nematode (SCN) was detected in Brazil in 1992 and since then it has been causing losses to Brazilian soybean crop yields. SCN populations have great genetic diversity which makes it difficult to manage this disease. The taxonomy of the genus Manihot is still uncertain and the genetic origin of cassava (M. esculenta Crantz) continues to be controversial. We studied the degree of genetic relatedness between cassava and two naturally occurring species (M. flabellifolia and M. peruviana) which are probably involved in the evolution of cassava, using RAPD and AFLP molecular markers. Maize breeders frequently wish to use exotic germplasm in their breeding programs without losing specific characteristics of their adapted material. The nanuzae group of lizards includes three species, Tropidurus nanuzae, T. divaricatus and T. amathites. The first species is found along Serra do Espinhaço, in eastern Brazil, and the other two in the northern region of the Brazilian State of Bahia, in continental dunes on both margins of the São Francisco River. Restriction fragment length polymorphisms (RFLP) of the mtDNA in these species were detected in 53 restriction sites. The C-band distribution patterns of 105 angiosperm species were compared to identify general patterns or preferential sites for heterochromatin. The base-specific fluorochrome reaction of heterochromatin for 58 of these species and the role played by the average chromosome size in band distribution were also considered. The results showed that heterochromatin was preferentially located in similar chromosome regions, regardless of the distance from the centromere. This trend results in generalized bands, with heterochromatin distribution being identical in most chromosomes of a karyotype. An Australian Merino flock was screened for low (resistant) and high (susceptible) yellow predictive colour (YPC) breeding values in order to compare extreme individuals using the differential display of mRNA technique. One differentially expressed cDNA band was visualised only in the resistant group. This band showed no identity with the DNA sequences of public databases; however, they showed short homologies with three database sequences related to transmembrane signalling functions. The use of these candidate genes as DNA markers needs to be confirmed against sheep with a wide range of susceptibility to wool yellowing to verify the results. |

Skin biopsies are frequently indicated for investigation and/or confirmation of genetic disorders. Although relatively simple and noninvasive, these procedures require care in order to increase probability of success and to avoid patient discomfort and unnecessary repeated analyses and associated laboratory fees. The present report highlights the importance of skin biopsies in genetic disorder diagnosis and presents general rules for collecting, storing, transporting and processing samples. We recommend its reading to professionals intending to use this important and sometimes fundamental diagnostic tool. |

Microsatellites or short tandem repeats (STRs), DNA markers relatively abundant in the genome, have a high degree of polymorphism and therefore great potential for characterizing populations. The present study estimates genetic variability in a set of four microsatellites (BMS3013, BMS3004, HEL10 and TGLA122) in a Brazilian hybrid bovine breed (5/8 Aberdeen Angus x 3/8 Nelore). The objectives were to determine the effect of crossbreeding and selection in these animals' genetic diversity as well as to discover the herd's genetic relationship with that of other breeds. The present study provides data on the nucleolar organizer regions (NORs) of seven Brycon species based on mapping of the 18S rRNA genes by fluorescence in situ hybridization (FISH). Fluorescent signals were observed on the telomere of the long arm of two large submetacentric chromosomes, thus confirming the number and location of NORs previously revealed by other classical cytogenetic techniques. Although there were no inter- or intra-individual variations in the number and location of the 18S loci, NOR size polymorphism was observed between homologous chromosomes. The clustering and conservation of NORs in a single chromosome pair indicates a high level of NOR stability among species of the genus Brycon. |

In addition to bone marrow and peripheral blood, stem cells also occur in human umbilical cord blood (HUCB), and there is an increasing interest in the use of this material as an alternative source for bone marrow transplantation and gene therapy. In vitro hematopoiesis has been maintained for up to 16 weeks in HUCB cultures, but the establishment of an adherent, stromal layer has consistently failed.
