Purification of papain from fresh latex of Carica papaya BABT

In the present study we wish to report a method of crystallizing papain from fresh papaya latex which gave higher yields than previously reported. This method does not involve the use of sulphydryl reagents. The papain thus obtained is practically pure and shows a single band when submitted to electrophoresis on polyacrylamide gel, and is identical to the papain obtained by other methods. In routine enzymatic assays, specific activity was measured using Z-gly-pNP and BAEE as substrates. Papain crystallized by this method, without the use of high concentrations of salts or thiol-containing substances such as cysteine and dithiothreitol, is obtained in the form of a complex with natural inhibitors existent in latex which can be removed by dialysis.

Interaction of temperature and light on seed germination in Tecoma stans L. Juss. ex Kunth (Bignoniaceae) BABT

The effects of the temperature and light on the control of seeds germination in Tecoma stans was studied in the present work. The influence of constant temperatures from 10 to 45ºC, with 5ºC intervals, under the white light and darkness were tested. The optimum temperature for the germination of the seeds was between 25 and 30ºC, for both the light and the dark treatments. The maximal germination was reached in the range of 15 to 35ºC under the light and of 20 to 40ºC in the darkness. The seeds showed highest synchronization of the germination near the optimal temperature. The germination in the field was tested under the two light conditions. Longitudinal distribution of Hypostomus punctatus (Osteichthyes, Loricariidae) in a coastal stream from Rio de Janeiro, Southeastern Brazil BABT

Hypostomus punctatus was collected between June/1987 and July/1988, in three sampling stations with different features, along the Ubatiba stream (Maricá, Rio de Janeiro state). Sampling stations 2 and 3 were more similar (50% similarity) to each other. The species was frequent and persistent in all sampling stations. The size varied from 1.0 to 22.2 cm. The occurrence of different size classes along the stream was probably related to the kind of environment. Young forms prevailed in the total sample and were mainly collected next to the submersed riparian vegetation. Adults occurred just in sampling stations 1 and 2, and were found most often in rapids with rocky bottom, and in pools among submersed branches.

Evaluation of Caesalpinia pulcherrima endospermic gum as affinity matrices for galactose-binding lectins interaction BABT

Lectins are proteins or glycoproteins able to bind, specifically and reversibly carbohydrates and glycoconjugates. Considering this ability, the utilization of Caesalpinia pulcherrima seeds polysaccharides as an affinity matrix was tested. The endospermic gum were solubilized in distinct concentrations of NaOH and treated with different amounts of epichlorohydrin (ECH) forming affinity gels with variable capacity for interaction with galactose- binding lectins. Systems biology approach to study the high altitude adaptation in tibetans BABT

The aim of this work was to study an integrative systems biology research strategy to construct a network including the protein-protein interactions (PPIs) and microRNAs (miRNAs) and identify the functional biological processes and pathways for high-altitude adaptation in Tibetans. The pathway enrichment analysis revealed that the genes in the network were mainly involved in signling the pathways and the function of microRNAs was concentrated in the signling pathways, which suggested that miRNAs might contribute to the Tibetans high-altitude adaptation through the participation in signaling pathway. These results contribute to better understanding on the high-altitude adaptation of the Tibetans.

Thermal and spectroscopic analyses on the molecular interaction between eucalyptus kraft pulp components and offset printing inks BABT

Thermal and infrared spectroscopic analyzes were carried out in order to study the kind of interaction between the anatomical components of the bleached eucalyptus kraft pulp with offset inks. A Bauer-McNett fiber classifier was used in order to obtain the anatomical components of the pulp, separately. The determinations of the enthalpy of the processes occurring in the samples of inks and pulp, as well as the enthalpy of these components interactions were obtained by Differential Scanning Calorimetry. In the interaction between pulp and offset ink, the reduced enthalpy values of the endothermic peak were interpreted as due to the released energy for adhesion of the ink pigment to the substratum. Production of allyl phenyl carbinol (APC) by biotrans-formation using Rhizopus arrhizus BABT

The objective of the present work was to study the stereoselective synthesis of homo-allylic alcohol using stereoselective hydrolysis by Rhizopus arrhizus and also to study the stereoselective synthesis of allyl phenyl carbinol (APC) or 1-Phenyl-3-butene-1-o by combination of chemical synthesis and biotransformation from cheap raw materials such as benzaldehyde and allyl bromide. Stereospecific synthesis of allyl phenyl carbinol (APC) was achieved by acetate hydrolysis by R. arrhizus giving R (+) enantiomer. Maximum enantiomeric excess of APC was obtained at 16 h where as maximum yield of it was obtained at 48 h of biotransformation.

Protein enrichment of cactus pear (Opuntia ficus - indica Mill) using Saccharomyces cerevisiae in solid-state fermentation BABT

The microbial protein bioconversion of cactus pear by yeast in solid medium was studied. Three cultivation variables used were: inoculum's concentrations (5, 10 and 15 %), substrate layer thickness (2, 4 and 6 cm) and temperature (30, 34 and 38 ºC). The rate of dry matter production and total protein were determined. Results obtained were variance analysis, gross energy and in vitro dry matter digestibility. The maximum protein amount achieved for the conditions studied in the present work was higher than 26 %, which was compatible or greater than those of conventional concentrates of protein supplements used for animal feed. Effect of salt on the growth and metabolism of Glycine max BABT

Soybean plants cultivated with 50, 100 and 200 mM of NaCl, revealed that root growth was less affected by salinity than shoots. Salinity led to a reduction in leaf area and an increase in water content of the roots. These factors could contribute to the adaptation of the plant, improving its hydration. Although nitrate and free amino acid levels were reduced by salt treatment in roots, protein content of leaves was not altered. Salinity led to alterations in xylem amino acid composition, with increases in Ser, Ala, Gaba and Pro and a decrease in Asn. Similar changes were seen for Asn and Ser in roots together with a much stronger increase in Gaba. First record of partial albinism and scoliosis in Odontophrynus occidentalis tadpoles (Anura: Cycloramphidae) BABT

Influence of ultrasound on sorbitol release by Zymomonas mobilis grown on high sucrose concentration BABT

This work investigated the effect of applying low intensity ultrasound on sorbitol release by Z.mobilis cultures grown on 200 g/L sucrose medium up to 48 h. The best sorbitol production was 36.09 g/L in 36 h culture. Ultrasound irradiation did not alter the sorbitol values detected after disrupting the cells with 20 minutes treatment.

Influence of temperature and diet on the development of Ulomoides dermestoides (Fairmaire, 1893) (Coleoptera, Tenebrionidae, Diaperinae) BABT

Ulomoides dermestoides (Fairmaire, 1893) develops in stored food products (peanuts, maize, oats, rice, sorghum, etc.) and breeds successfully in the laboratory. To determine the best conditions for development, experiments were set up in different temperatures and diets, similar to storage conditions of peanuts (Arachis hypogaea L.). The higher viability of individuals and the shorter developmental time were observed in the diet composed of hulls and seeds of fruits at 21 and 24°C.

Comparison of proximate chemical composition and texture of cupim, Rhomboideus m. and lombo, Longissimus dorsi m. of Nelore (Bos indicus) BABT

The proximate chemical composition of hump, known in Brazil as cupim, Rhomboideus m. (RB), of Nelore (Bos indicus) aged 24 months revealed it to be a unique beef muscle. It presents a lipid fraction 14-fold as high as that of Longissimus dorsi m. (LD) taken from the same animal (p<0.05), the highest value reported so far. This was unequivocally observed by the histological evaluation. Proportionally more protein fraction and conversely less moisture were also observed in RB. Analysis of collagen and its crosslinking with hydroxylysylpyridinium (HP) showed there to be 22.9% more collagen and 14-fold as much HP in RB as in LD. Rhizosphere Bacterial Composition of the Sugar Beet Using SDS-PAGE Methodology BABT

ABSTRACT The rhizosphere zone has been defined as the volume of soil directly influenced by the presence of living plant roots or soil compartment influenced by the root. During the growing season of 2014, the rhizobacteria of 23 sugar beet plants sampled from 12 sites in the west and north west of Iran were inventoried. Using a cultivation-dependent approach, a total of 217 bacteria were isolated from the rhizosphere. The bacterial isolates were tentatively grouped and documented based on polyacrylamide gel electrophoresis of whole-cell proteins and were found to represent 43 different protein electrotypes. Effects of trace volatile organic compounds on methane oxidation BABT

The effects of volatile organic compounds (VOCs) on methane oxidation in landfill cover soils were examined. The batch experiments were conducted using single and mixed VOCs, such as, dichloromethane (DCM), trichloroethylene (TCE), tetrachloroethylene (PCE), and benzene. The results from all combinations showed a decrease in methane oxidation rate with increase in VOC concentrations. Moreover, inhibition effects of TCE and DCM were found higher than benzene and PCE. The reduction of methane oxidation by benzene and PCE could be attributed to the toxicity effect, whereas TCE and DCM were found to exhibit the competitive-inhibition effect. Effect of carrageenan addition on the yield and functional properties of charqui (Jerked Beef) BABT

The objective of this work was to evaluate the application of carrageenan (CAR) to improve the functional properties of the jerked beef (JF) and to increase its processing yield. JB produced from Vastus lateralis with CAR (1.0%) at 25ºC and NaCl (15.0%) had approximately 15.0% higher moisture and a 32.0% higher processing yield in comparison to the control samples.JB-CAR presented shear force approximately 5.0 and 20% lower in the samples uncooked salted and desalted cooked, respectively, and sensorial acceptance above 80%. Temporal expression pattern of the insulin-like growth factor II and fibroblast growth factor transcripts in avian embryogenesis BABT

In this study, the abundance of IGF-II and bFGF transcripts was estimated in the chicken embryos using the competitive RT-PCR analysis. Significant enhancements in the abundance of IGF-II mRNA were observed at stages HH1 and 5, and a new accumulation in these levels was observed at stage HH18 in comparison to the basal levels. The abundance of bFGF mRNA increased significantly at stages HH18 and 20, followed by an upregulation in the expression of these transcripts at stage HH26. These findings provided important information about the temporal expression pattern of IGF-II and bFGF transcripts in the whole chicken embryos during in ovo development.

Photosynthetic rate of the aquatic macrophyte Egeria densa Planch. (Hydrocharitaceae) in two rivers from the Itanhaém River Basin in São Paulo State, Brazil BABT

Egeria densa is a submerged aquatic macrophyte which, in Brazil, causes problems specially for electric energy generation. The purpose of this study was to evaluate the photosynthetic rate of E. densa in two rivers (Aguapeú and Mambu) with different limnological features, both located in the Itanhaém River Basin. This species is abundant in both rivers without causing damages to their multiple uses.
