Blood glucose regulation during fasting in rats under food restriction since birth BABT

The effect of severe food restriction since birth on regulation of fasting glycemia in male Wistar rats was investigated. The control group (CG) had free supply of chow, while the restriction group (RG) received 50% of the amount ingested by the CG. The experiments were done in adult (60 days) overnight fasted rats in which glycemia, liver free glucose levels and hepatic glycogen concentration were measured. In part of the experiments in situ liver perfusion was done. The results showed that livers from the RG had higher glycogenolysis rates but lower gluconeogenesis rates from L-alanine (10 mM). Evaluation of gelling agents on anther culture: response of two soybean cultivars BABT

Anthers of two soybean cultivars were cultured in B5 long basal culture media gelled with agarose or PhytagelTM. Cytological examinations of the anthers were carried out during the first 45 days of culture to assay the viability and developmental stage of microspores. Frequency of callus formation was recorded at 45 days of culture. The analysis of variance of the microspore viability assay showed significant Cultivar X Gelling Agent X Day of Culture interactions. ABSTRACT In view of the traditional use of Tabebuia aurea for treating pain and inflammation, the antinociceptive pharmacological potential of T. aurea ethanolic extracts (TAEE) was investigated through in vivo experimental models. First, the MTT assay was conducted to determine the potential cytotoxicity of the TAEEs. Afterwards, the acetic acid-induced writhing test and the formalin-, and glutamate-induced nociception tests were performed on Swiss adult mice treated with TAEEs (100 and 200 mg/kg doses, p.o.), or saline solution (control groups, 10mL/kg, p.o.), or standard drugs: dipyrone 40 mg/kg (p.o.), and morphine 5,7 mg/kg (i.p). Preliminary assessment of the potential for mangrove oyster cultivation in Piraquê-açu river estuary (Aracruz, ES) BABT

At Piraquê-açu river estuary, Aracruz, ES, the technical viability of Crassostrea rhizophorae cultivation was determined through monthly measures in shell length and weight. Seeds of C. rhizophorae were put in cages and suspended in rafts. Increase in height and weight (flesh and dry) of the oysters was measured. During ten months (July/98 to May/99), oyster shell reached an average of 37.6 mm in shell height and 3.0 g in flesh weight (the whole animal). High mortality rates were registered and could be related to the high salinity water and to high predation observed, especially by flatworms Stylochophana divae and snails Cymatium parthenopeum, as well as fouling organisms such as barnacles, Serpulidae polychaetes and seed of the same species.

Age-related Changes in DNA Methylation Status of hTERT Gene Promoter of Oral Epithelial Cells BABT

The purpose of this study was to investigate the effect of aging on the DNA methylation status of two genes involved in tumorigenesis (telomerase gene hTERT and DNA repair gene- MLH1) and one in metabolism (methylenetetrahydrofolate reductase gene- MTHFR) in oral epithelial cells. DNA methylation analysis was performed by Methylation Sensitive Restriction Enzymes (MSRE) of healthy oral epithelial cells of child (6-10 years, n=21), young (20-25 years, n=19) and elderly (over 60 years, n=25). Studies were carried out on the localization and mapping of gas stations in the urban area of São Carlos city, for an environmental diagnosis related to soil, surface water and underground water contamination resulting from likely leaking in fuel underground storage tanks. Considering that a provision of legal restriction makes it possible that most of public areas are not located in gas station areas of potential risk, these information are useful for the development of programs designed to reduce risks to public safety and health and of the soil, surface water and underground water contamination.

Influence of an extract of Juglans regia on the growth of Escherichia coli, on the electrophoretic profile of plasmid DNA and on the radiolabeling of blood constituents BABT

The aim of this work was to study the influence of a walnut (Juglans regia) extract on the growth of Escherichia coli (E. coli) AB1157, on the plasmid DNA topology and on the labeling of blood constituents with technetium-99m (99mTc). An E. coli AB1157 culture, in stationary phase, was incubated with walnut and the growth of the culture was evaluated by optical density at 600 nm for 7 hours. Plasmid DNA samples were incubated with SnCl2 in presence or absence of walnut for 40 minutes, 0.8% agarose gel electrophoresis was performed, the gel was stained and the plasmid topological forms were visualized. Soil erosion determination in a watershed from northern Paraná (Brazil) using 137Cs BABT

The objective of this work was study the soil losses or gains in a watershed in the municipal district of Cambé, Paraná using 137Cs as marker for the determination of soil redistributions. A transect sampling was used to evaluate the influence of different tillage on soil erosion. One point, located in a forest area was sampled and analyzed to determine the reference inventory of cesium-137 deposited by fallout. The average value of the reference inventory was 292 Bq m-2. The cesium-137 inventory of the transect samples varied from 80 Bq m-2 to 403 Bq m-2. The sampling points in pasture presented soil losses. The sampling points in coffee plantation did not present losses or gains. The sampling points in soybean cultivated areas presented soil losses.

Search for alkaloids on callus culture of Passiflora alata BABT

Preliminary work on Passiflora alata leaves failed to detect harmane alkaloids using LC. The aim of this work was to investigate the production of harmane alkaloids through the cell culture of P. alata, inducing its precursor (L-tryptophan). The leaf explants presented satisfactory results after disinfection, and the callus formation was initiated in MS media with adequate quantities of phytohormones. Sixty days after inoculation, calli were inoculated in the optimized semi-solid MS media, with and without the addition of L-tryptophan (50, 100, 200 mg/L) and kept in standard conditions for 90 days. Occurrence of coliforms in water samples of the Perequê and Penedo Rivers in Paraná, Brazil BABT

The Perequê and Penedo rivers flow through the plains of Praia de Leste and fall into the sea between the municipalities of Pontal do Sul and Ponta do Poço. Both of these rivers belong to the sub-basin of Paranaguá Bay, and are constituent parts of the hydrographic basin of the Atlantic Ocean (State of Paraná). Four sites along the Perequê River (Stations 1-4) and one along the Penedo River (Station 5) were monitorated for water pollution over a 12-month period. During the period of December 1995 to November 1996, three water samples were collected from each site, placed in sterilized bottles and transported to the laboratory in ice-boxes. ABSTRACT The aim of this study was to assess the sperm quality and testicular histomorphometry of Wistar rats supplemented with extract and fractions of fruits of Tribulus terrestris L. The ethanolic extract was obtained by dynamic maceration of spray-dried fruit. This extract was fractionated by liquid-liquid partition, using increasing polarity solvents. Twenty male rats were separated in four groups, with five rats in each group. Population dynamics of Pseudevadne tergestina (Branchiopoda: Onychopoda) in Guanabara Bay, Brazil BABT

Populations of Pseudevadne tergestina were studied in Guanabara Bay, southeastern Brazil, to assess temporal variations in density and population dynamics. Data on temperature, salinity and zooplankton samples were taken from the superficial water of a fixed station, every 3 - 4 days, from February 2 through August 1, 2000. The highest abundance of this species was observed in March, when densities varied widely, from 20 to 600 ind. m-3. Abstract Ginger and white yam starches were investigated and compared with maize starch. Proximal composition, thermogravimetry, differential scanning calorimetry, microscopy, colourimetry, X-ray powder diffractometry and pasting profile were analysed. The unconventional starches presented higher protein and ash contents than the maize starch, that had the highest thermal stability. Higher gelatinisation temperatures were reported for ginger starch, and the enthalpy of the unconventional starches were similar. The maize starch presented the lowest gelatinisation values. Rate of water loss and sugar uptake during the osmotic dehydration of pineapple BABT

Water loss, sucrose gain and the variation in concentration of other natural fruit sugars (glucose and fructose) were studied during osmotic dehydration of pineapple slices (0.6 mm thick) in sucrose solution (60 % w/w) at three temperatures (30, 40 and 50ºC). As temperature increased from 30 to 50ºC, the apparent moisture and sucrose diffusivities (Dw and Ds) increased 3.8 and 2.8 times, respectively; therefore, the dehydration efficiency index (Dw/Ds) increased with temperature. The loss of glucose and fructose increased with temperature too. It was found that the solute content was a linear function of the moisture content and this relation was independent of the temperature during the first 600 minutes of dehydration.

Action of a pancreatin and an Aspergillus oryzae protease on whey proteins: correlation among the methods of analysis of the enzymatic hydrolysates BABT

The objectives of this study were to hydrolyze whey proteins using a pancreatin and an Aspergillus oryzae protease; to evaluate the degree of hydrolysis (DH) and the peptide profile; and to establish the correlations among the analytical methods. Ten hydrolysates were prepared at different reaction times and the highest DH was obtained by the protein content method. Good correlations (r > 0.87) between the methods of formaldehyde and orthophthalaldehyde (OPA), formaldehyde and osmometry as well as osmometry and OPA were observed using pancreatin. Similar results were obtained between OPA and soluble protein content for the A. oryzae protease.
