A simplified DNA extraction method for PCR analysis of Camarotella spp. BABT

This work aimed to optimize an efficient and simple protocol for DNA extraction of Camarotella species, an obligate plant pathogen that cause verrucosis or "lixa" on coconut tree and other palms, facilitating the molecular studies of these biotrophic microorganisms. The method proposed enabled a fast, reproducible and reliable DNA extraction from Camarotella species.

Associações entre macroinvertebrados e peixes demersais na armação do Itapocoroy, Penha, Sc, Brasil BABT

In the period of January and December, 1995, there were collected by means of overtrawl, 12977 samples of macroinvertebrates and demersal fishes, in the Armação do Itapocoroy, Penha (SC), between the depths of 6 to 10 meters, in the periods of morning, afternoon, and evening. There were registered the incidence of 57 families, 80 kinds and 92 species distributed between the macroinvertebrates and demersal fishes. The families of greater incidence were: Portunidae (30.7%), Strombidae (11.4%), Sciaenidae (11.1%) and Bothidae (10.3%) which all together, contributed with 63.5% of the total samples. Characteristics of Immobilized Urease on Grafted Alginate Bead Systems BABT

This study evaluated the biological importance of immobilized urease enzyme over the free urease. The support material used for urease immobilization was alginate. Generally, the immobilization of urease in alginate gel showed a marked increase in Km and Vmax. However, the immobilized urease showed higher thermal stability than that of free enzyme. The rate of thermal inactivation of the immobilized enzyme decreased due to entrapment in gel matrix. Also, the activity of the immobilized urease was more stable in retention than that of the free enzyme during the storage in solution, although the activity of the immobilized enzyme was lower in comparison with the free enzyme. Changes in hyphal morphology due to chitosan treatment in some fungal species BABT

In this work, changes in the hyphal morphology due to chitosan treatment in some fungal species were studied. Scanning electron microscope (SEM) observations revealed that chitosans with molar fraction of acetyl groups (F A 0.16 and 0.18) and degree of polymerization (DP 1,089 and 1,242) had a direct effect on the morphology of the chitosan-treated fungi, reflecting its potential for causing a delay in the growth of Alternaria alternata (500 µg × mL-1), Botrytis cinerea (1,000 µg × mL-1), Penicillium expansum (1,000 µg × mL-1) and Rhizopus stolonifer (500 µg × mL-1). Mycelial aggregation and structural changes such as excessive branching, swelling of the cell wall and hyphae size reduction were observed in the micrographs.

Efeito do etanol e do peróxido de hidrogênio intragástrico sobre a mucosa gástrica de rã (Rana catesbeiana, SHAW) BABT

This work investigated the frog gastric mucosa response to hydrogen peroxide and ethanol induced injury. Acid and mucus secretion were estimated "in vitro" in control animals with intragastric absolute ethanol (1ml/30min. or 2ml/60min.) and hydrogen peroxide. The gastric mucosa morphological conditions were assessed "in vivo", concerning lesion area, pH and mucus. Ethanol (1ml/30min.) was observed to cause hyperemia, cell damage, rupture, edema, erosions, necrosis in gastric mucosa and significant increase in acid secretion. Absolute ethanol (2ml/60min.) caused a decrease in acid secretion due to alcalinization and an increase of mucus and pH. Different concentrations of 6-benzyladenine (6-BA) were used in vitro to induce buds in three types of explants: leaf discs, root segments and the seedling itself obtained from in vitro seed germination of Passiflora cincinnata Mast. The 0.5 mg.L -1 6-BA concentration was most suitable for all the three explants, however, bud formation time and means (direct/indirect) were different for each type of explant. Absorção de metais pesados do lodo de esgoto pelo feijoeiro (Phaseolus vulgaris L.) BABT

Toxicity and uptake of heavy metals of sewage sludge by beans (Phaseolus vulgaris L.) were evaluated in green house experiments. Treatments consisted of 1,0 ; 2,0 and 5,0% (m/m) of dry sewage sludge, collected from Londrina (Bom Retiro and ETE-Sul) and Curitiba (ETE-Belém and RALF). Bean ( variety IAPAR 57) was sown three times at 0, 120 and 240 days after the treatments have been applied. Contents of Cd, Cr, Co, Cu, Ni, and Pb in bean tissues cultivated with 5,0% (m/m) of all sewage sludge were similar to the control and Ba contents were reduced by increasing the quantity of sewage sludge in the soil. ABSTRACT Solar energy as a form of electric power generation is gaining ground in Brazil, being the subject of worldwide discussions on sustainability. The electric energy, a right of all citizens, represents a consumption of natural resources on a large scale. State schools in Paraná invested more than R$ 46 million to guarantee electricity and thus provide essential running conditions to the system in 2016. Evaluation of the biodegradation of different types of lubricant oils in liquid medium BABT

The aim of this work was to study the biodegradation of different types of automotive lubricant oils adapted to the aqueous medium using a base inoculum and an aqueous inoculum. Four treatments were carried out in two consecutive and similar experiments: T1 (control); T2 (half-synthetic oil); T3 (mineral oil); T4 (used oil). The results showed the following decreasing order of CO2 production in the Bartha and Pramer respirometers: T4 > T2 > T3 > T1. Thus, the used lubricant oil showed with highest biodegradability, followed by the half-synthetic one and the mineral oil. It was also observed that the mineral lubricant presented a longer period of adaptation compared to the half-synthetic one.

Yeasts and filamentous fungi in bottled mineral water and tap water from municipal supplies BABT

The main objective of this study was to analyse the occurrence of yeasts and filamentous fungi in drinking water as well as to investigate their correlation with the indicator bacteria of faecal pollution. Yeasts were detected in 36.6% and 11.6% of the bottled mineral on water dispensers and tap water samples from municipal system, respectively. Twenty-one (35.0%) of bottled mineral water and two (3.3%) of tap water samples were positive for filamentous fungi. For bottled mineral water 12 (20.0%) of 60 samples were positive for total coliform, compared with 3(5.0%)out of 60 samples from tap water. Anatomy of the pericarp and seed-coat of Lithraea molleoides (Vell.) Engl. (Anacardiaceae) with taxonomic notes BABT

The aim of the present work was to record anatomical data for the fruit and seed of Lithraea molleoides (Vell.) Engl, and compare the results with those for L. brasiliensis and the genera Schinus and Rhus. The L. molleoides fruit was a drupe with a friable and lignified exocarp. The mesocarp was parenchymatous with large secretory canals associated with vascular bundles. The endocarp consisted of four layers: an outer layer of polyhedral cells with prismatic crystals of calcium oxalate, and three inner layers of sclereids in a palisade arrangement. To test the assumption that exposure of the host to parasitoid for long periods could provoke superparasitism, the aim of this work was to test the consequences on the immature development time, productivity of parasitoids per pupa, sex ratio and rate of parasitism of Nasonia vitripennis bred in Chrysomya megacephala pupae. Each individual pupa was placed in a glass tube with one parasitoid female for 24, 48, 72 and 96 h period of exposure, under controlled laboratory conditions. Analytical quality in environmental studies: uncertainty evaluation of chemical concentrations determined by INAA BABT

Instrumental neutron activation analysis (INAA) is a measurement technique of high metrological level for the determination of chemical elements. In the context of BIOTA/FAPESP Program, leaves of trees have been evaluated by INAA for biomonitoring purposes of the Atlantic Forest. To assure the comparability of results in environmental studies, a leaf sample of Marlierea tomentosa (Myrtaceae family) showing the lowest concentrations of chemical elements was selected for the evaluation of analytical quality of the determination under unfavorable conditions. Influence of pH on the growth, laccase activity and RBBR decolorization by tropical basidiomycetes BABT

The basidiomycete fungi Lentinus crinitus and Psilocybe castanella are being evaluated in a bioremediation process of soils contaminated with organochlorine industrial residues in the Baixada Santista, São Paulo. The aim of the present study was to determine the influence of pH on the fungal growth, in vitro decolorization of anthraquinonic dye Remazol Brilliant Blue R (RBBR) and laccase activity. The pH of the culture medium influenced the growth of L. crinitus and P. castanella, which presented less growth at pH 5.9 and pH 2.7, respectively. Grape quality mapping for vineyard differential harvesting BABT

An experiment was carried out from 2005 to 2008, to calibrated NIR-based instrumentation and explore within field grape quality variability and map potential grape quality descriptors along vineyards, as a subsidy for differential harvesting,. The quality indicators (anthocyanin content, pH, titratable acidity and soluble solids) were subject to geo-spatial modeling. Subsequently, the data set was utilized to delineate "within-field" grape quality zone and to determine the timing of the harvest. Differential machine harvesting was implemented and segregation of wine grapes was done "on-the-go". The approach for field prediction of grape quality parameters and zone delineation allowed for separated fermentation for at least two wine styles.

Evaluation of short wave therapy equipment based on brazilian standards BABT

Short Wave Diathermy (SWD) equipment is used in physiotherapy and applied therapeutically using radio frequency (RF) radiation of 27.12 MHz. This article presents the results of a survey related to SWD, showing data about the equipment, conditions of use, visual inspection and analysis of accompanying documents, having as reference the requirements of the Brazilian standards NBR IEC 601-1 and NBR IEC 601-2-3. Electrical safety tests were also carried out for the equipment and the installation. The results showed that manufacturers do not completely follow the standards and there is a lack of knowledge of these standards by the physiotherapists. Degradation and detoxification of three textile Azo dyes by mixed fungal cultures from semi-arid region of Brazilian Northeast BABT

The aim of this work was to study the degradation and detoxification of three textile azo dyes (Reactive Red 198, Reactive Red 141 and Reactive Blue 214) by mixed fungal cultures from semi-arid region of Brazilian Northeast. Sediment samples of twenty water reservoirs in the surroundings of Serra da Capivara National Park, area of environmental preservation in the caatinga in the State of Piauí, with semi-arid climate, were evaluated in order to select the consortia of fungi capable to degrade and detoxify these dyes. The mixed fungal culture from Caldeirão Escuridão (CE) reservoir was the most efficient in the degradation and detoxification of the dyes tested.

Population dynamics of Musca domestica (Diptera: Muscidae): experimental and theoretical studies at different temperatures BABT

In the present study, the population dynamics of M. domestica was evaluated at two different temperatures, 20 and 30ºC. The dynamics was modeled using a density-dependent model of population growth. The temperatures investigated in this study produced no qualitative change in terms of dynamic behaviour, i. e. the population dynamics of M. domestica was characterized by a stable equilibrium at both temperatures. However, the steady state was influenced by the results obtained at different temperatures. The difference between the eigenvalues obtained at the two temperatures was probably the cause of the difference between the distinct steady states. The implications of these results for the population dynamics of M. domestica are discussed.
