Quantification of ruminal microbiota and production of methane and carbonic dioxide from diets with inclusion of glycerin Arq. Bras. Med. Vet. Zootec.
Santos,V.C.; Ezequiel,J.M.B.; Homem Junior,A.C.; Pinheiro,R.S.B..
The aim of this study was to quantify the liquid-associated bacteria and protozoa of the rumen and to evaluate the potential of in vitroproduction of gases CH4 and CO2 of diets with inclusion of glycerin for sheep. Two diets were formulated with a roughage:concentrate ratio of 20:80, according to the presence or absence of glycerin: G0 - control, without glycerin; and G10 - 10% glycerin as part of the concentrate. To quantify the liquid-associated bacteria and protozoa from the rumen and production of CH4 and CO2 gases, a completely randomized design was used. The diets had no effect (P>0.05) on production and composition of liquid-associated protozoa and bacteria from the rumen of lambs. Likewise, a (P>0.05) diet effect was not observed on...

Resistance to antibiotics in Gram-negative bacteria isolated from broiler carcasses Arq. Bras. Med. Vet. Zootec.
Moreira,M.A.S.; Moraes,C.A..
One hundred and ninety-seven isolates of Gram-negative bacteria, comprising 10 genera, were isolated from poultry carcasses at a processing plant in order to investigate resistance to low levels of antibiotics. The samples were taken just after evisceration and before inspection. Most of the isolates were of Samonella and Escherichia. Other genera present were Enterobacter, Serratia, Klebsiella, Kluyvera, Erwinia, Citrobacter, Pseudomonas and Aeromonas. Distinct profiles of antibiotic resistance were detected. Resistance to more than two antibiotics predominated and spanned several classes of antibiotics. Salmonellae and escherichiae were mainly resistant to the aminoglycosides, followed by tetracycline, nitrofuran, sulpha, macrolide, chloramphenicol,...

In vitro effect of pH on resistance of ruminal bacteria to intracellular potassium depletion, and effect of pH and ionophores on ammonia and microbial protein production Arq. Bras. Med. Vet. Zootec.
Leopoldino,W.M.; Lana,R.P.; Borges,A.C.; Mantovani,H.C.; Teixeira,R.M.A.; Oliveira,J.S.; Jaremtchuk,A.R.; Eifert,E.C.; Martins,R.G.R..
Ruminal fluid from steers fed on pasture was incubated with artificial media at pH 5.5 and 7.0 in two experiments. In the first, the effect of monensin level on resistance of ruminal bacteria to potassium depletion was evaluated; in the second, effects of the ionophores monensin and lasalocid on ammonia and protein production were quantified. In experiment 1, culture media affected potassium level. The monensin concentration needed to cause half maximal potassium depletion was 2.77µM at pH 5.5 but was 0.056µM at pH 7.0, showing that bacteria incubated at pH 5.5 were more tolerant to monensin than those incubated at pH 7.0. Both ionophores as well as increased acidity caused decreased ammonia production. Both ionophores inhibited ammonia production by 56%,...

Identification and antimicrobial susceptibility of bacteria isolated from corneal ulcers of dogs Arq. Bras. Med. Vet. Zootec.
Prado,M.R.; Brito,E.H.S.; Girão,M.D.; Sidrim,J.J.C.; Rocha,M.F.G..
A total of 22 clinical specimens were obtained from 19 dogs with corneal ulcer (16 unilateral and three bilateral) for isolation and antimicrobial susceptibility evaluation of the isolated bacteria. Bacterial growth was observed in 100% of the samples (n=22). Staphylococcus intermedius was the predominant species (35.5%), followed by Corynebacterium xerosis (19.3%). Gentamicin, ciprofloxacin, chloramphenicol and tobramycin had a high efficacy against all of the isolated bacteria. The results evidenced that 80.7% of the isolates were Gram positive cocci and Gram positive bacilli, and that those microorganisms were sensitive to gentamicin, ciprofloxacin, chloramphenicol and tobramycin.

Sponges in Peru and their potential as a source of antibacterial compounds Boletín de Investigaciones
Cóndor-Luján,Báslavi; Francia-Quiroz,Juan C..
ABSTRACT In the last decades, sponges have emerged as the largest source of active compounds of animal origin for pharmaceutical purposes. In Peru information on these animals is incipient and scattered, which limits taking the right measures for their adequate management, exploitation, and conservation. The aim of this study is to provide an update on the scope and perspectives of sponge research in Peru, with an emphasis on species richness and their antibacterial potential. Likewise, research initiatives carried out in other South American Pacific countries are reviewed. Research efforts in Peru have allowed the description of 46 species; however, the sponge richness is underestimated. On the other hand, in other latitudes, active principles with...

Effect of buctril super (Bromoxynil) herbicide on soil microbial biomass and bacterial population BABT
Abbas,Zafar; Akmal,Muhammad; Khan,Khalid Saifullah; Fayyaz-ul-Hassan,.
The present study aimed to evaluate the effect of bromoxynil herbicide on soil microorganisms, with the hypothesis that this herbicide caused suppression in microbial activity and biomass by exerting toxic effect on them. Nine sites of Punjab province (Pakistan) those had been exposed to bromoxynil herbicide for about last ten years designated as soil 'A' were surveyed in 2011 and samples were collected and analyzed for Microbial Biomass Carbon (MBC), Biomass Nitrogen (MBN), Biomass Phosphorus (MBP) and bacterial population. Simultaneously, soil samples from the same areas those were not exposed to herbicide designated as soil 'B' were taken. At all the sites MBC, MBN and MBP ranged from 131 to 457, 1.22 to 13.1 and 0.59 to 3.70 µg g-1 in the contaminated...

Inhibition of bacteria contaminating alcoholic fermentations by killer yeasts BABT
Meneghin,Maria Cristina; Reis,Vanda Renata; Ceccato-Antonini,Sandra Regina.
The aim of this work was to study the in vitro antibacterial activity possessed by killer yeast strains against bacteria contaminating alcoholic fermentation (Bacillus subtilis, Lactobacillus plantarum, Lactobacillus fermentum and Leuconostoc mesenteroides), in cell X cell and cell X crude toxin preparations. The bacteria were not inhibited by any S. cerevisiae killer strains (5 out of 11). The inhibition caused by two crude toxin preparations (Trichosporon figueirae and Candida sp) against L. plantarum was surprisingly high but not in the same extent for B. subtilis, especially with three killer strains (Candida glabrata, Pichia anomala and Candida sp). L. mesenteroides and L. fermentum strains were neither inhibited in cell X cell nor crude toxin X cell...

Microbiological analysis of critical points in the chicken industry BABT
Cansian,Rogério Luis; Floriani,Salete Teresa Radeski; Valduga,Eunice.
This work is focused on identifying microbial contamination in the scalding asepsis and cooling processes as well as in fresh sausages obtained. Salmonella was identified in two scald water samples but was absent in the water from chiller and in the final product, which might be explained in terms of chlorine addition and temperature reduction. The analysis revealed that MPN of Escherichia coli was in the range of <1-11/mL in the scald water and <1-64/mL in the chiller. Aeromonas sp. count showed 5 to 3.5x10¹ CFU/mL in the scald water and 9 to 3.7x10² CFU/mL in the chiller, probably due to the psycrophile characteristics of Aeromonas and evisceration process. The analysis of fresh sausages chicken revealed a sharp increase in the Aeromonas count...

Ross,Claudia; Menezes,Jose Roberto de; Svidzinski,Terezinha Inez Estivalet; Albino,Ulisses; Andrade,Galdino.
In tropical countries such as Brazil, there is not enough information about microbial contaminants in indoor environments with air conditioning systems. Microbial monitoring of such environments is important for the quality of human life. The aim of this work was to assess the fungal genera and bacterial morphotypes occurring in such environments. Air samples were taken indoors and outdoors from a public auditorium, a hospital, a company and a shopping center during the 2001 winter by using a six-stage impactor Millipore M air T® . Twenty-one fungal genera were identified. Bacterial morphological groups found were Gram positive and negative rods and Gram positive coccus.

Measurement of volatilized mercury by a mini-system: a simple, reliable and reproducible technique BABT
Cursino,Luciana; Mattos,Silvânia V. M.; Silva,Nilton O.; Chartone-Souza,Edmar; Nascimento,Andréa M. A..
A simple and easy new technique for volatilized mercury determination in biological systems was developed. This technique is fast and sensitive and can overcome the problems that arise due to the extremely low readings during the measurements and reproducibility in biological material (bacteria). It measures directly the volatilized metallic mercury of bacteria by means of a chemical adsorbent in a coupled mini-system, as a modified technique for mercury in air analysis. It is potentially of interest to the bioremediation and bacterial mercury resistance communities

Ardra profiles of bacteria and archaea in mangrove sediments with different levels of contamination in the estuarine complex of Paranaguá, Brazil BABT
Ribeiro,Catherine Gérikas; Steffens,Maria Berenice Reynaud; Etto,Rafael Mazer; Galvão,Carolina Weigert; Martins,César de Castro; Pedrosa,Fábio de Oliveira; Kolm,Hedda Elisabeth.
The mangrove's sediments from the coastal areas under human activities may contain significant contaminations by hydrocarbons, even when there are no visual evidences of it. The microorganisms are essential to these ecosystems, especially in the control of their chemical environment. Sediment samples were collected in two regions under different environment conditions (pristine and contaminated) of the Paranaguá Estuarine Complex (Paranaguá Bay and Laranjeiras Bay), Brazil. Aliphatic hydrocarbons were determined by the GC-FID to assess the status of contamination of the studied areas. The total DNA was extracted from these samples. The 16S rRNA gene was amplified by the PCR reactions with the pair of primers 21F and 958R for the archaeal domain, and 27F...

Reducing COD level on oily effluent by utilizing biosurfactant-producing bacteria BABT
Jacobucci,Daniela Franco Carvalho; Oriani,Maria Raquel de Godoy; Durrant,Lucia Regina.
Two bacteria isolated from crude oil contaminated soil, Pantoea agglomerans and Planococcus citreus, produced biosurfactants utilizing 1.5% of kerosene and olive oil as the sole carbon sources, respectively. The bacteria and the biosurfactants produced were introduced to oily effluent, arising from margarine and soap industry. Emulsification activities were determined by increases in the absorbance of the oil-in-water emulsions at 610 nm, whereas the water-in-oil emulsions were expressed as the height (cm) of the emulsion layers formed. The 72 h incubation experiment resulted in a COD (Chemical Oxygen Demand) reduction of 76% with Planococcus citreus strain and 70% with Pantoea agglomerans. The COD reduction with bacterial biosurfactants was over 50% in 24...

Direct RAPD evaluation of bacteria without conventional DNA extraction BABT
Araújo,Welington Luiz; Angellis,Derlene Attili de; Azevedo,João Lúcio.
The present work reports successful DNA amplification of Pantoea agglomerans and Bacillus pumilus through Random Amplified Polymorphic DNA (RAPD). For this, template DNA was obtained without conventional DNA extraction. The procedure was as follows: cultures grown for 20 hours in 5 mL LB medium were centrifuged and the resulting preparation was suspended in TE buffer. After boiling, the cell suspension was diluted and 2.0 µl were used in reactions of 15 µl. The results showed no significant differences among the RAPD profile of the PCR reactions derived from the boiling and phenol extraction methods, suggesting the utilization of this method for genetic population analysis.

Functional Genes of Microorganisms, Comprehending the Dynamics of Agricultural Ecosystems BABT
Reali,Catiusca; Pilz Júnior,Harry Luiz; Meile,Jean Christophe; Galindo,Sabine Shorr; Fiuza,Lidia Mariana.
ABSTRACT The microbial composition of different types,in ecosystems (including agro-ecosystems), has been investigated in a rapidly growing number of studies in the past few years. The importance of microorganisms, regarding the maintenance and stability of nutrients in agroecosystems, is a key to maintain the sustainability of a crop. Molecular tools to study microbial communities are possible through many methods such as RISA, DGGE, TGGE, clone libraries, T-RFLP, RAPD, SSCP and more recently NGS (Next-Generation Sequencing). DGGE is widely employed to characterize the diversity and the community dynamics of microorganisms in the environment, making possible to find out specific groups through functional genes, allowing access to data that cannot be...
