Effect of Agaricus brasiliensis and Lentinula edodes mushrooms on the infection of passionflower with Cowpea aphid-borne mosaic virus BABT

The objective of the present study was to evaluate the protection of passion fruit plants against CABMV by using preparations from Agaricus brasiliensis and Lentinula edodes mushrooms. In experiments carried out in the greenhouse, the fruiting body extracts from some of the isolates of both mushrooms significantly reduced CABMV incidence in passion fruit plants. This protective effect occurred when the plant leaves, pre-treated with extracts, were later inoculated mechanically with the virus. However, the extracts did not protect the plants in experiments involving CABMV transmission by aphid vectors. The study of stem cells has evolved rapidly in recent decades. The importance is given to the concept that these cells are potentially able to become any cell type and have the power of self-renewal throughout the life of the organism. Mesenchymal stem cells (MSCs) can be isolated from various organs of the body such as bone marrow, adipose tissue, synovium, muscle and dermis, deciduous teeth, umbilical cord, placenta, liver, spleen and thymus. Grazing Scar Characteristics Impact Degree of Fungal Facilitation in Spartina alterniflora Leaves in a South American Salt Marsh BABT

Grazing scars of burrowing crabs and Hemiptera insects were simulated on leaves of the salt marsh grass Spartina alterniflora. Simulations of crab feeding generated two-fold higher fungal (ergosterol) content in leaves in comparison to that generated by insect scar simulations (1.26 ±0.55 and 0.57 ±0.25 µg per cm², respectively). This study provided evidence that herbivory could facilitate microbial infection by fungi in dominant South American salt marsh plants and indicated that specific feeding mechanisms used by different herbivores might differentially impact the strength of this interaction.

Purification and Characterization of a Polygalacturonase Produced by Wickerhamomyces anomalus BABT

The aim of this work was to study the purification and physicochemical properties of an endo-polygalacturonase (PG) produced by Wickerhamomyces anomalus isolated from the citrus fruit peels. The enzyme was purified to homogeneity from the culture filtrate of W. anomalus grown on the yeast nitrogen base medium with glucose as carbon and energy source and citrus pectin as inductor. After anion-exchange chromatography and gel filtration chromatography, PG activity was eluted as a single peak, yielding 21% of the original activity. After dialysis and cation-exchange chromatography, only one fraction with PG activity was obtained, recovering 56% of initial enzyme activity and 1.3-fold increase in specific activity. Isolation and Characterisation of a Reserve Protein from the Seeds of Cereus jamacaru (Cactaceae) BABT

We describe here the isolation and characterisation of a major reserve protein from the seeds of Cereus jamacaru. (Cactaceae). This protein has a molecular mass of 5319 kDa and was isolated by a combination of gel filtration chromatography and reverse phase HPLC. The amino acid composition of the protein was determined and it was shown to have similarities with the amino acid composition of several proteins from the 2S albumin storage protein family. The usefulness of this protein as a molecular marker in the Cactaceae is also discussed.

Activity determination of Na+ K+ - ATPase and Mg++ - ATPase enzymes in the gill of Poecilia vivipara (Osteichthyes, Cyprinodontiformes) in different salinities BABT

This work aimed to know the tolerance mechanisms through the salinity variation by Na+ K+ - ATPase and Mg++ - ATPase and enzymes encountered in the gills of Poecilia vivipara. In field, the presence of this species was observed in salinities of 0 and 28‰. In laboratory, these fish were maintained in aquarium with mean salinity of 30‰ and positive responses were obtained. Some adult specimens, collected in a lagoon of the Coqueiros Beach, were utilized as matrixes. In the experiments the specimens used were those born in the test aquarium. Protective effect of sodium selenite against the genotoxicity of aflatoxin B1 in human whole blood cultures BABT

This study was designed to investigate the effects of selenium and aflatoxin on human whole blood cultures (WBC) in relation to induction of sister-chromatid exchange (SCE). The results showed that the frequency of SCEs in peripheral lymphocytes was significantly increased by the direct-acting mutagen AFB1 (at doses 5 and 10 µM except for 1µM) compared to controls. When sodium selenite (Na2SeO3) was added alone at a molar ratio of 5x10-7 and 1x10-6, cells did not show significant increase in SCE frequency. Whereas, SCE rates induced by the various AFB1 concentrations could be significantly reduced by the presence of Na2SeO3 in a clear dose-related manner. Characterization of immobilized biomass by amplified rDNA restriction analysis (ARDRA) in an anaerobic sequencing-batch biofilm reactor (ASBBR) for the treatment of industrial wastewater BABT

The performance of an anaerobic sequencing-batch biofilm reactor (ASBBR- laboratory scale- 14L )containing biomass immobilized on coal was evaluated for the removal of elevated concentrations of sulfate (between 200 and 3,000 mg SO4-2·L-1) from industrial wastewater effluents. The ASBBR was shown to be efficient for removal of organic material (between 90% and 45%) and sulfate (between 95% and 85%). The microbiota adhering to the support medium was analyzed by amplified ribosomal DNA restriction analysis (ARDRA). Cloning and expression of meta-cleavage enzyme (CarB) of carbazole degradation pathway from Pseudomonas stutzeri BABT

In this work, the 1082bp PCR product corresponding to carBaBb genes that encode the heterotetrameric enzyme 2'-aminobiphenyl-2,3-diol 1,2-dioxygenase (CarB), involved in the Pseudomonas stutzeri ATCC 31258 carbazole degradation pathway, was cloned using the site-specific recombination system. Recombinant clones were confirmed by PCR, restriction enzyme digestion and sequencing. CarB dioxygenase was expressed in high levels and in active form in Escherichia coli BL21-SI using the His-tagged expression vector pDEST TM17 and salt induction for 4h.

This work had the goal to evaluate the influence of temperature and dry storage on the postharvest longevity of Epidendrum ibaguense flowers previously treated with silver thiosulfate (STS). The flowers were harvested and pulsed for 30 minutes with 2 mM STS. Then both, the STS treated and untreated flowers were packed in perforated low density polyethylene bags, packed in cardboard boxes and stored at 5 and 10°C, where they remained for four and eight days. Measurements of leaf gas exchange at different photosynthetic photon flux density (PPFD) levels were conducted in order to compare the photosynthetic traits of five neotropical rainforest tree species, with a special emphasis on empirical mathematical models to estimate the light response curve parameters incorporating the effects of leaf-to-air vapour pressure deficit (D) on the saturated photosynthetic rate (Amax). Antimicrobial effect of a crude sulfated polysaccharide from the red seaweed Gracilaria ornata BABT

The aim of this study was to determine the yield, chemical composition, specific rotation (SR), infrared (IR) spectroscopy and the effect on bacterial growth of a crude sulfated polysaccharide (SP) from the red marine alga G. ornata (Go). Go-1 (25°C), Go-2 (80°C), and Go-3 (80°C) were sequentially extracted and yielded 9.2%. The contents of sulfate (5.88-10.3%) and proteins (0.1-3.7%) were small. The values of SR were [µ]D20°f -19.0, -51.0, and -56.5, respectively. Abstract Thymoquinone (TQ), the main constituent of the volatile oil derived from Nigella sativa has shown pharmacological benefits against various diseases while nicotine is an active component in cigarette that is known to be detrimental. This study was conducted to assess the ameliorating effects of TQ on sperm count, membrane, mitochondria and testosterone of nicotine-treated rats. Rats were randomized into four groups: control, nicotine, TQ, and nicotine with TQ. Enzymatic systems involved in D-limonene biooxidation BABT

The biooxidation of limonene by an Aspergillus strain resulted in the production of perillyl alcohol and short chain fatty acids. Addition of ketoconazole, a known inhibitor of cytochrome P450 oxydase, eliminated the production of free acids, but did not affect biotransformation to perillyl alcohol.

Response of Diabrotica speciosa (Coleoptera: Chrysomelidae) to 1,4-dimethoxybenzene and analogs in common bean crop BABT

Several synthetic and commercial analogs of 1,4-dimethoxybenzene, a kairomone of Diabrotica speciosa, along with other compounds already shown to be attractive to other species of Diabrotica, were tested as attractant to D. speciosa. Yellow cup traps were lured with the compounds and installed in a common bean field. Assessments were conducted 24 h later. 1,4-dimethoxybenzene lured traps caught significantly more beetles than the control traps. Captures of traps lured with 1,4-dimethoxybenzene analogs did not differ from the control traps. Results showed that position and nature of the substituents on the aromatic ring played a crucial role in the activity of the natural compound. The aromatic ring was also very important to the activity of the kairomone.

Purification and characterization of β-Fructosidase with inulinase activity from Aspergillus niger - 245 BABT

Aspergillus niger - 245, a strain isolated from soil samples showed good β-fructosidase activity when inoculated in medium formulated with dahlia extract tubers. The enzyme was purified by precipitation in ammonium sulphate and percolated in DEAE-Sephadex A-50 and CM-cellulose columns, witch showed a single peack in all the purification steps, maintaining the I/S ratio between 0.32 to, 0.39. Optimum pH for inulinase activity (I) was between 4.0 - 4.5 and for invertase activity (S) between 2.5 and 5.0. The optimum temperature was 60O.C for both activities and no loss in activity was observed when it was maintained at this temperature for 30 min.
