Polycyclic aromatic hydrocarbons (PAHs) constitute a large family of organic pollutants emitted in the environment as complex mixtures, the compositions of which depend on origin. Among a wide range of physiological defects, PAHs are suspected to be involved in disruption of reproduction. In an aquatic environment, the trophic route is an important source of chronic exposure to PAHs. Here, we performed trophic exposure of zebrafish to three fractions of different origin, one pyrolytic and two petrogenic. Produced diets contained PAHs at environmental concentrations. Reproductive traits were analyzed at individual, tissue and molecular levels. Reproductive success and cumulative eggs number were disrupted after exposure to all three fractions, albeit to...

Effect of fasting on self-feeding activity in juvenile sea bass (Dicentrarchus labrax) ArchiMer
Benhaim, David; Begout, Marie-laure; Pean, Samuel; Brisset, Blandine; Leguay, Didier; Chatain, Beatrice.
In various experiments under self-feeding conditions, sea bass groups could be divided into three categories regarding feeder actuation: high, low and zero-triggering fish. In all cases few high-triggering fish were responsible for a high percentage of the feed delivery. A question was raised about the role played by feeding motivation in such high-triggering status acquisition. It was approached by applying a 3-week fasting period in order to induce similar negative specific growth rate (SGR) in two groups of fish of similar mean weight but with either a low or a high coefficient of variation for weight (CVw)(T-low: CVw similar to 11%, 3 tanks of 60 fish each; T-high: CVw similar to 20%, 3 tanks of 60 fish each). These groups were created to test the...

The release of polycyclic aromatic hydrocarbons (PAHs) into the environment has increased very substantially over the last decades leading to high concentrations in sediments of contaminated areas. To evaluate the consequences of long-term chronic exposure to PAHs, zebrafish were exposed, from their first meal at 5 days post fertilisation until they became reproducing adults, to diets spiked with three PAH fractions at three environmentally relevant concentrations with the medium concentration being in the range of 4.6–6.7 μg g−1 for total quantified PAHs including the 16 US-EPA indicator PAHs and alkylated derivatives. The fractions used were representative of PAHs of pyrolytic (PY) origin or of two different oils of differing compositions, a heavy fuel...

Sedentary behaviour establishment in 0-group common sole Solea solea: a laboratory video-tracking study ArchiMer
Durieux, Eric; Le Duigou, Mathieu; Millot, Sandie; Sasal, Pierre; Begout, Marie-laure.
Spontaneous swimming activity of 0-group common sole (Solea solea) was evaluated using a video-tracking system under laboratory conditions. An experiment was conducted during two consecutive days on individuals sampled in June, July, September and November (2004) in a coastal nursery ground (Pertuis Charentais, Bay of Biscay, France). The measured behavioural variables were: distance travelled and frequency of occurrence of burying, swimming and immobility. 0-group sole showed a relatively clear circadian activity in line with the artificial light conditions (day, twilight and night). Swimming activity of 0-group sole decreased drastically from June to July, thereafter remaining at a very low level in September and November. Such important changes reflect...

Consistency in European seabass coping styles: A life history approach ArchiMer
Ferrari, Sebastien; Millot, Sandie; Leguay, Didier; Chatain, Beatrice; Begout, Marie-laure.
Recent years have seen a growth of interest in the consistent differences in individual behaviour over time and contexts constituting so-called “individual coping styles”. An understanding of this inter-individual variation is essential to improve our knowledge of the adaptive value of behaviour. Coping styles may have implications in diverse fields, so the development of appropriate screening methods for each species appears to be the most effective way to extend our knowledge and to incorporate behavioural responses into selection-based breeding programmes, to improve the domestication and welfare of farmed fish. We tested 30 juvenile seabass (Dicentrarchus labrax) at least twice in individual-based tests (feeding recovery in isolation, aggressiveness,...

Temporal changes in lipid condition and parasitic infection by digenean metacercariae of young-of-year common sole Solea solea (L.) in an Atlantic nursery ground (Bay of Biscay, France) ArchiMer
Durieux, Eric; Galois, Robert; Begout, Marie-laure; Sasal, Pierre; Lagardere, Francoise.
Little attention has been paid to flatfish parasitism as a potential factor of variation in year-class strength. The aim of this study was to evaluate the relationship between parasitic infection and nutritional condition of young-of-year (YOY) common sole. Sole were collected monthly from May (early settlement) to November 2004 in a mussel pole culture area, using a push-net or a trawl, and dissected for parasite identification and counts. Total lipids were extracted from the whole body and lipid classes quantified. Triacylglycerols to sterols (free form) ratio (TAG/ST) was used as an index of nutritional status. Digenean metacercariae infection, characterized by two dominant genera, displayed clear seasonal dynamics and the highest records yet reported...

Genetic variability of environmental sensitivity revealed by phenotypic variation in body weight and (its) correlations to physiological and behavioral traits ArchiMer

Adaptive phenotypic plasticity is a key component of the ability of organisms to cope with changing environmental conditions. Fish have been shown to exhibit a substantial level of phenotypic plasticity in response to abiotic and biotic factors. In the present study, we investigate the link between environmental sensitivity assessed globally (revealed by phenotypic variation in body weight) and more targeted physiological and behavioral indicators that are generally used to assess the sensitivity of a fish to environmental stressors. We took advantage of original biological material, the rainbow trout isogenic lines, which allowed the disentangling of the genetic and environmental parts of the phenotypic variance. Ten lines were characterized for the...

Tagging juvenile seabass and sole with telemetry transmitters: medium-term effects on growth ArchiMer
Begout, Marie-laure; Coves, Denis; Dutto, Gilbert; Laffargue, Pascal; Lagardere, Francoise.
The effects of tagging with acoustic transmitters on the growth of juvenile seabass, Dicentrarchus labrax (L.) (initial mean mass +/- SD, 173 g +/- 23.4) in a 47 d tank experiment, and sole. Solea solea (L.) ( 103.2 g +/- 14.8) in a 72 d tank experiment and (104.0 g +/- 18.4) in a 58 d salt marsh mesocosm experiment were examined. Twenty externally tagged seabass grew more slowly than the 20 with surgically implanted tags, which reached the same mass as nine control fish. Movements of the external transmitter's harness caused abrasions of the skin and loss of the tag in 60% of the cases. We thus recommend implanting transmitters for telemetry studies of juvenile seabass weighing between 120 and 214 g and carrying a tag that represents 2.2-2.5% of body...

Early individual electronic identification of sea bass using RFID microtags: A first example of early phenotyping of sex-related growth ArchiMer
Ferrari, Sebastien; Chatain, Beatrice; Cousin, Xavier; Leguay, Didier; Vergnet, Alain; Vidal, Marie-odile; Vandeputte, Marc; Begout, Marie-laure.
Although individual electronic tagging using PIT (Passive Integrated Transponder) tags is well established, it is mainly used for fish > 60 mm in length. Since electronic tagging is an ideal identification method, we used RFID (Radio Frequency Identification) microtags (6 mm in length, 1 mm in diameter, 10 mg in mass) to characterize individual fish from the early stages of their development and throughout their lifecycles. We used sea bass, Dicentrarchus labrax, (105 day-old fish weighing between 100 and 1100 mg) and studied the effects of intra-coelomic tagging in half the population using different endpoints including survival and tag reading, growth over 6 months and swimming responses. Dead fish were counted daily, biometric data were collected at...

Systematic Screening of Behavioral Responses in Two Zebrafish Strains ArchiMer
Vignet, Caroline; Begout, Marie-laure; Pean, Samuel; Lyphout, Laura; Leguay, Didier; Cousin, Xavier.
Wild-type (WT) zebrafish are commonly used in behavioral tests, but the term WT is not a precise description, and corresponds to many different strains (e.g., AB, TU, WIK, and others). Previous studies compared the physiological, behavioral, or metabolic characteristics of different zebrafish strains (indigenous WT populations versus laboratory WT strains). AB and TU are widely used, but at least one study has demonstrated behavioral differences between them. To choose the most appropriate strain for our experiments, we systematically screened behavioral responses of AB and TU fish in several assays. We analyzed the locomotion activity and responses to a light/dark challenge in adults and larvae, and exploratory behavior and color conditioning in adults....

Activity patterns, home-range size, and habitat utilization of Sarpa salpa (Teleostei : Sparidae) in the Mediterranean Sea ArchiMer
Jadot, Catherine; Donnay, A; Acolas, M; Cornet, Y; Begout, Marie-laure.
Acoustic telemetry was used to record diel movement and habitat utilization of the salema (Sarpa salpa) (Teleostei: Sparidae) during three consecutive summers from 2000 to 2002 in the Calvi and Achiarina bays of Corsica in the Mediterranean Sea. A total of 18 fish was equipped with acoustic transmitters inserted in the body cavity, 13 were tracked in the Bay of Calvi (275 mm +/- 26.9 L-F), and 5 in Achiarina Bay (260 mm +/- 33.6 LF). Two different systems were used to track the fish. The one used in the Bay of Calvi was a manual receiver and a directional hydrophone. The second system, used in Achiarina Bay, was a radioacoustic-positioning (RAP) system that continuously monitored the movements of the fish. Fish positions were put in a geographic...

Long-term monitoring of individual fish triggering activity on a self-feeding system: An example using European sea bass (Dicentrarchus labrax) ArchiMer
Coves, Denis; Beauchaud, M; Attia, J; Dutto, Gilbert; Bouchut, C; Begout, Marie-laure.
In two experiments, a computerized on-demand feeding system coupled with a PIT tag monitoring device was used to continuously record the triggering activity by ca. 50 individual sea bass for 55 days (Exp. 1, initial average body weight and coefficient of variation, CV: 299 g, 15%) and 69 days (Exp. 2, 157 g, 13%). Each group was stocked in 1 m(3) tanks and exposed to a water temperature of 21 +/- 1 degrees C and a light regime of 16:8 LD. Only 5% of the triggering activity was not associated with simultaneous PIT tag detection. Although each individual was detected via PIT tag during the experiment, only 67% and 74% of the fish actuated the trigger at least once in Exp. 1 and Exp. 2, respectively. Moreover, only two fish in Exp. 1 and one fish in Exp. 2...

First links between self-feeding behaviour and personality traits in European seabass, Dicentrarchus labrax ArchiMer
Ferrari, Sebastien; Benhaim, David; Colchen, Tatiana; Chatain, Beatrice; Begout, Marie-laure.
Most studies carried out with seabass under self-feeding conditions report an intriguing social structure that is built around the device and the food dispenser with three coexisting triggering categories: high-triggering (HT), low-triggering (LT) and zero-triggering (ZT) fish. However, neither sex nor feeding motivation or hierarchy can explain the establishment of this specialization. We characterised the personality of seabass with the commonly used restraint and open field tests and assessed the link between personality traits and individual triggering activity towards the self-feeder apparatus. We found no differences between triggering categories during the restraint test but high triggering fish were characterised as shyer than low- and...

Innovative behaviour in fish: Atlantic cod can learn to use an external tag to manipulate a self-feeder ArchiMer
Millot, Sandie; Nilsson, Jonatan; Fosseidengen, Jan Erik; Begout, Marie-laure; Ferno, Anders; Braithwaite, Victoria A.; Kristiansen, Tore S..
This study describes how three individual fish, Atlantic cod (Gadus morhua L.), developed a novel behaviour and learnt to use a dorsally attached external tag to activate a self-feeder. This behaviour was repeated up to several hundred times, and over time these fish fine-tuned the behaviour and made a series of goal-directed coordinated movements needed to attach the feeder’s pull string to the tag and stretch the string until the feeder was activated. These observations demonstrate a capacity in cod to develop a novel behaviour utilizing an attached tag as a tool to achieve a goal. This may be seen as one of the very few observed examples of innovation and tool use in fish.

Self-feeding behaviour and personality traits in tilapia: a comparative study between Oreochromis niloticus and Sarotherodon melanotheron ArchiMer
Benhaim, David; Akian, Dieudonne; Ramos, Mathieu; Ferrari, Sebastien; Yao, Kouakou; Begout, Marie-laure.
Hybridization aims at combining valuable traits from two species into a single group. Nile tilapia Oreochromis niloticus (NT) and Black-chinned tilapia Sarotherodon melanotheron (BCT) are respectively characterized by fast growth and water salinity tolerance which attract the breeders who could take advantage of both species. The first step is to characterize both species behavior in different contexts. The aim of this study was to compare the self-feeding behaviour between NT and BCT with a design allowing to reveal individual and group feed demand behaviour and then to identify the individual specialization that builds around the device and the food dispenser. The second objective was to estimate the links between the individual specialization and...

Feeding responses of hatchery-reared gilthead sea bream (Sparus aurata L.) to a commercial diet and natural prey items ArchiMer
Andrew, J; Begout, Marie-laure; Kadri, S; Holm, J; Huntingford, F.
Many fish species have evolved feeding mechanisms and behaviours enabling them to feed on specific prey. However, such mechanisms may not be optimal for feeding on commercial-pelleted diets in aquaculture. Gilthead sea bream chew and occasionally eject pellets or parts of pellets from the mouth when feeding on commercial diets. This may result in an increase in nutritional waste from the intensive culture of this species. In this study we examined the prevalence of this food processing behaviour in two sizes of sea bream, feeding on three types of natural prey items in comparison to a commercial pellet, to give an insight into the circumstances in which excess chewing and ejection of food items from the mouth occurred. These included two hard-textured food...

Exploration behaviour and flight response toward a stimulus in three sea bass strains (Dicentrarchus labrax L.) ArchiMer
Millot, Sandie; Begout, Marie-laure; Chatain, Beatrice.
Domestication and selection may affect fish behaviour, sometime as soon as at the first generation of domestication. However, knowledge about how both processes impact on fish spatial exploration and swimming activity still is to be improved. The objective of this experiment was (i) to evaluate spatial exploration behaviour and swimming activities of three sea bass strains having different domestication and selection levels and (ii) to analyse their responses to an acute stress. Sea bass exploration and swimming activities were studied before, during and 40 min after a stimulation (standardized fall of an object). The experimental tank was divided in to four zones, and the time spent, the distance travelled in each zone and the swimming complexity were...

Effects of T-bar and DST Tagging on Survival and Growth of European Hake ArchiMer
Jolivet, Aurelie; De Pontual, Helene; Garren, Francois; Begout, Marie-laure.
Controlled experiments were conducted to assess the effects of T-bar and DST tagging on post-release survival and growth of European hake. In this study, two groups of each 30 hake were considered: small fish (SF, average total length: 29.9 cm +/- 2.2 cm) and large fish (LF, average total length: 36.4 cm +/- 2.5 cm). Within each size group, fish were randomly assigned to one of 3 treatment groups: control (C), T-bar tagging referred as conventional tagging (CT) and DST tagging (DST) with dummy tags. After 4 months, the overall survival rate was 35%. Smaller fish were less impacted by the stress induced by handling, anaesthesia and tagging and in the SF group, the survival rates were similar (30%) for CT or DST. Specific growth rates were highly variable...
