Disinfectants and antiseptics used in mollusc hatcheries and nurseries ArchiMer
Francois, Cyrille; Pepin, Jean-francois.
Increasing development of aquaculture business has thrown up new requirements in terms of prevention and control of mollusc diseases. In closed structures, one way to reduce infectious risk is to make use of anti-infectives . Specific drugs like antibiotics should be used only for therapeutic purpose against identified agents. Less specific anti-infectives like disinfectants could be employed for preventive and also for curative goals against causal pathogens and opportunist agents. A critical analysis synthesizes the bibliography available on disinfection in mollusc hatcheries and nurseries. Physical and chemical tools are reviewed then selected depending on four criteria: (1) efficiency against most important pathogens of molluscs, (2) safety for...

Space-time clustering of mortality notifications in Pacific oysters of Charente sluices, France, 2008-2010 ArchiMer
Lupo, Coralie; Mandard, Yann-vari; Arzul, Isabelle; Francois, Cyrille; Garcia, Celine; Renault, Tristan; Bareille, Nathalie.
Current monitoring of exceptional mortality events of oysters in France consists in passive surveillance, leading to delayed investigation of causation by scientists and management by policy makers. Early detection of clusters of mortality events in time and space would increase possibilities for their investigation and management. A space-time cluster analysis of existing mortality notifications in Pacific oysters was conducted in France, Charente sluices, from 2008 to 2010. As an example of potential demographic confounder of clustering, the analyses were adjusted on oyster age. Several clusters were identified in space and time each studied year. Patterns of mortality notifications were consistent with the introduction and spread of a contagious disease...

Du concept de santé à la détermination d’intervalles de référence chez l’huître creuse Crassostrea gigas ArchiMer
Francois, Cyrille.
Of interest of everyone, health is not an easy concept to figure out. Although there is no universal definition of health, its various uses and meanings allow us to specify its biological, psychic and social dimensions. In animal health, the biological dimension remains the most studied, there is therefore no consensual definition of international animal health. So how to understand the health of marine shellfish? A living being can be functionally described in physiopathology by monitoring biomarkers under laboratory conditions. In men and many animals, "normal" biomarker values have been proposed; these have been determined by carrying out measurements or observations in samples of healthy populations, the rigorous selection of these populations and...

Since its molecular characterisation, Ostreid herpesvirus 1 (OsHV-1) has been regularly detected in Crassostrea gigas in France. Although its pathogenicity was demonstrated on larval stages, its involvement during mortality outbreaks at the juvenile stage was highly suspected but not evidenced. To investigate mortality outbreaks, the French National Network for Surveillance and Monitoring of Mollusc Health (REPAMO) carried out two surveys in juvenile C. gigas. The first survey lasted from 1998 to 2006 and was an epidemiological inquiry occurring when oyster farmers reported mortality outbreaks. The second survey, a longitudinal one, was set up in 1998 to complete the network observations on OsHV-1. Data analysis showed a specific pattern of mortality...

Défis de la surveillance des maladies chez les coquillages marins en France ArchiMer
Lupo, Coralie; Francois, Cyrille; Arzul, Isabelle; Garcia, Celine; Joly, Jean-pierre; Renault, Tristan.
The basic concepts and objectives of animal disease surveillance are common to all types of animal production. Yet, the procedures for implementation have been mostly developed for terrestrial animal productions. Disease surveillance in marine shellfish calls for specific measures in view of the peculiarities of marine biology and rearing techniques. The main objective of disease surveillance in marine shellfish is to detect as early as possible exotic and newly emerging diseases. With this objective in mind, a national surveillance network for surveillance of mollusc diseases called Repamo has been developed. It combines passive reporting and risk-based surveillance.

High-throughput phenotyping of health biomarkers in Crassostrea gigas by magnetic resonance imaging ArchiMer
Francois, Cyrille; Quellec, Stéphane; Collewet, Guylaine; Peyramaure, Paul; Musse, Maja; Lamy, Jean-baptiste; Sauriau, Pierre-guy; Haure, Joel.
High-throughput phenotyping of shellfish’s traits constitutes a challenge due to the specificities of aquatic animals. Appropriate measuring tools have often to be developed or adapted, in order to continuously and automatically acquire data of biomarkers of interest. IMAGIGAS project aimed to explore a non-destructive and non-invasive approach by using magnetic resonance imaging (MRI) to monitor at high-throughput two biomarkers (body weight and temperature) of the pacific oyster, before and during infectious challenges by Ostreid Herpesvirus type 1 (OsHV-1). Pacific oysters and OsHV-1 suspension and contaminated seawater were produced at Ifremer facilities. The experiments were conducted in 2016-2017 by three staffs (Ifremer LGPMM, Irstea IRM Food,...

Repamo network : a surveillance tool for mollusc health ArchiMer
Garcia, Celine; Arzul, Isabelle; Chollet, Bruno; Francois, Cyrille; Joly, Jean-pierre; Miossec, Laurence; Robert, Maeva.
At the present time losses due to diseases are one of main factors limiting the aquaculture development. Unceasing increase of exchanges between different countries favours the risk of introduction and propagation of new pathogen agents. At the beginning of the 90's the European Union laid the surveillance of mollusc health inside each Member State in order to protect the safety of exchanges. Indeed few measures are available to protect molluscs from diseases. Only the prophylactic approach is possible; this approach is based on a zoosanitary surveillance of mollusc populations and/or a development of resistant populations against some diseases. Created in 1986, the French network for surveillance and monitoring of mollusc health (REPAMO) ensures the...

The search for novel compounds of marine origin has increased in the last decades for their application in various areas such as pharmaceutical, human or animal nutrition, cosmetics or bioenergy. In this context of blue technology development, microalgae are of particular interest due to their immense biodiversity and their relatively simple growth needs. In this review, we discuss about the promising use of microalgae and microalgal compounds as sources of natural antibiotics against human pathogens but also about their potential to limit microbial infections in aquaculture. An alternative to conventional antibiotics is needed as the microbial resistance to these drugs is increasing in humans and animals. Furthermore, using natural antibiotics for...

Molecular detection and quantification of the protozoan Bonamia ostreae in the flat oyster, Ostrea edulis ArchiMer
Robert, Maeva; Garcia, Celine; Chollet, Bruno; Lopez-flores, Inmaculada; Ferrand, Sylvie; Francois, Cyrille; Joly, Jean-pierre; Arzul, Isabelle.
Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. The assay had a 6 log- dynamic range, mean inter- and intra-assay variation coefficients of <1% and a minimum detection limit of 50 gene copies per reaction. Using...
