Several strains, one disease: experimental investigation of Vibrio aestuarianus infection parameters in the Pacific oyster, Crassostrea gigas ArchiMer
Travers, Marie-agnes; Tourbiez, Delphine; Parizadeh, Leila; Haffner, Philippe; Kozic-djellouli, Angelique; Aboubaker, Mohamed; Koken, Marcel; Degremont, Lionel; Lupo, Coralie.
This study investigated oyster infection dynamics by different strains of Vibrio aestuarianus isolated before and after the apparent re-emergence of this pathogen observed in France in 2011. We conducted experiments to compare minimal infective dose, lethal dose 50 and bacterial shedding for six V. aestuarianus strains. Whatever the strain used, mortality was induced in juvenile oysters by intramuscular injection and reached 90–100% of mortality within 5 days. Moreover, bacterial shedding was comparable among strains and reached its maximum after 20 h (≈10 EXP5 bacteria/mL/animal). Similarly, our first estimations of lethal dose 50 were comparable among strains (minimal infective dose around 0.4 × 10EXP5 bacteria/mL and LD50 around 10EXP5 bacteria/mL) by...

Défis de la surveillance des maladies chez les coquillages marins en France ArchiMer
Lupo, Coralie; Francois, Cyrille; Arzul, Isabelle; Garcia, Celine; Joly, Jean-pierre; Renault, Tristan.
The basic concepts and objectives of animal disease surveillance are common to all types of animal production. Yet, the procedures for implementation have been mostly developed for terrestrial animal productions. Disease surveillance in marine shellfish calls for specific measures in view of the peculiarities of marine biology and rearing techniques. The main objective of disease surveillance in marine shellfish is to detect as early as possible exotic and newly emerging diseases. With this objective in mind, a national surveillance network for surveillance of mollusc diseases called Repamo has been developed. It combines passive reporting and risk-based surveillance.

REPAMO : a French network for the surveillance of molluscs health ArchiMer
Guichard, Benjamin; Garcia, Celine; Joly, Jean-pierre; Lupo, Coralie; Chollet, Bruno; Robert, Maeva; Omnes, Emmanuelle; Arzul, Isabelle.
The REPAMO is a French Network for the surveillance of Mollusc diseases that notably meets the obligations of the European Directive 2006/088/EC. It is entirely funded by the French State. The coordination of this network entrusted to the Institut Français de Recherche pour l`Exploitation de la Mer since 1986 aims at monitoring the notifiable endemic diseases, detecting the emergence of exotic or new pathogens and ensuring a minimum level of surveillance of the general health condition of molluscs. Beside IFREMER, different partners are involved in the surveillance of mollusc health at the national level: the Departmental direction for territories and sea who are the local representatives of the competent authority and the producers who have the obligation...

Improving early detection of exotic or emergent oyster diseases in France: identifying factors associated with shellfish farmer reporting behaviour of oyster mortality ArchiMer
Lupo, Coralie; Osta-amigo, Axel; Mandard, Yann-vari; Peroz, Carole; Renault, Tristan.
Farmers’ vigilance is essential for the detection of epidemics, including potential emerging diseases, in marine shellfish. A field study was conducted to investigate oyster farmers’ reporting practices and behaviour, and to identify factors influencing the reporting process of oyster mortality, with the ultimate aim of improving early detection of unexplained oyster mortality outbreaks. A retrospective case-control study of oyster farmers from Charente-Maritime (France) was designed, based on interviews with 27 non-reporting and 89 reporting farmers, further split into 40 formerly-reporting and 49 currently-reporting farmers. Information about farmer and farm characteristics, farming practices, farm health history and related financial compensation on...

Infectious diseases in oyster aquaculture require a new integrated approach ArchiMer
Pernet, Fabrice; Lupo, Coralie; Bacher, Cedric; Whittington, Richard J..
Emerging diseases pose a recurrent threat to bivalve aquaculture. Recently, massive mortality events in the Pacific oyster Crassostrea gigas associated with the detection of a microvariant of the ostreid herpesvirus 1 (OsHV-1 µVar) have been reported in Europe, Australia and New Zealand. Although the spread of disease is often viewed as a governance failure, we suggest that the development of protective measures for bivalve farming is presently held back by the lack of key scientific knowledge. In this paper, we explore the case for an integrated approach to study the management of bivalve disease, using OsHV-1 as a case study. Reconsidering the key issues by incorporating multidisciplinary science could provide a holistic under-standing of OsHV-1 and...

Modeling the Transmission of Vibrio aestuarianus in Pacific Oysters Using Experimental Infection Data ArchiMer
Lupo, Coralie; Travers, Marie-agnes; Tourbiez, Delphine; Barthélémy, Clement; Beaunée, Gaël; Ezanno, Pauline.
Vibrio aestuarianus is a bacterium related to mortality outbreaks in Pacific oysters, Crassostrea gigas, in France, Ireland, and Scotland since 2011. Knowledge about its transmission dynamics is still lacking, impairing guidance to prevent and control the related disease spread. Mathematical modeling is a relevant approach to better understand the determinants of a disease and predict its dynamics in imperfectly observed pathosystems. We developed here the first marine epidemiological model to estimate the key parameters of V. aestuarianus infection at a local scale in a small and closed oyster population under controlled laboratory conditions. Using a compartmental model accounting for free-living bacteria in seawater, we predicted the infection dynamics...

Comment améliorer la précocité de l'alerte en conchyliculture ? Exemple des mortalités de moules en 2014 dans les Pertuis Charentais ArchiMer
Lupo, Coralie; Prou, Jean.
Surveillance of shellf ish health is based on clinical surveillance of shellfish mortality, including a detection phase and a mandatory notification phase. While control measures are limited, early detection of pathogens is a challenge which needs to be met. The relevance and feasibility of an enhanced clinical surveillance based on phone interviews were studied. The results made it possible to reconstruct the spatial and temporal evolution of the outbreaks of mussel mortality which occurred in Pertuis Charentais in 2014. Moreover, a qualitative analysis of the interviews revealed contextual elements that were used to enhance the previous description and to devise causal scenarios which could be used as research hypotheses. By incorporating the...

Spatial epidemiological modelling of infection by Vibrio aestuarianus shows that connectivity and temperature control oyster mortality ArchiMer
Lupo, Coralie; Dutta, Bhagat Lal; Petton, Sebastien; Ezanno, P; Tourbiez, Delphine; Travers, Marie-agnes; Pernet, Fabrice; Bacher, Cedric.
Vibrio aestuarianus infection in oyster populations causes massive mortality, resulting in losses for oyster farmers. Such dynamics result from host-pathogen interactions and contagion through water-borne transmission. To assess the spatiotemporal spread of V. aestuarianus infection and associated oyster mortality at a bay scale, we built a mathematical model informed by experimental infection data at 2 temperatures and spatially dependent marine connectivity of oyster farms. We applied the model to a real system and tested the importance of each factor using a number of modelling scenarios. Results suggest that introducing V. aestuarianus in a fully susceptible adult oyster population in the bay would lead to the mortality of all farmed oysters over 6 to...

Stakeholders' perceptions, attitudes and practices towards risk prevention in the food chain ArchiMer
Lupo, Coralie; Wilmart, O.; Van Huffel, X.; Dal Pozzo, F.; Saegerman, C..
An online survey was conducted to describe stakeholders' perceptions, attitudes and practices towards risk prevention in the food chain and to explore if common features could be extracted from different fields of competency or groups of stakeholders. Out of 80 participants, 60% believed that pathogenic microorganisms were the main hazard to prevent. Twenty-four percent perceived climate change as the main risk factor. Seventy-three percent believed that hazards in the food chain are preventable and they often showed a positive attitude towards risk prevention measures. The opinion of 75% of stakeholders was that prevention measures should be compulsory and under the shared responsibility of both food business operators and competent authority....

Preventing and mitigating farmed bivalve disease: a Northern Ireland case study ArchiMer
Fox, Michaela; Christley, Robert; Lupo, Coralie; Moore, Heather; Service, Matt; Campbell, Katrina.
Shellfish production forms a large proportion of marine aquaculture production in Northern Ireland (NI). Diseases represent a serious threat to the maintenance and growth of shellfish cultivation with severe consequences to production output and profitability. In Northern Ireland, production generally benefits from a good health status with the absence of notifiable diseases, except for localised cases of Bonamia ostreae, Marteilia refringens and ostreid herpes virus. In this paper, we qualitatively explore that the prevalence, risk, impact, mitigation and experience shellfish farmers in this region have in relation to disease. Sixteen semi-structured interviews were conducted with stakeholders within the sector. The interviews were transcribed verbatim,...

Bilan 2016 du dispositif national de surveillance de la santé des mollusques marins ArchiMer
Lupo, Coralie; Fleury, Elodie; Normand, Julien; Osta Amigo, Axel; Robert, Stephane; Canier, Lydie; Francois, Cyrille; Garcia, Celine.
