Because of its rapid growth, its good reputation and the promising aquaculture experiences conducted in Norway and Scotland, cod is considered as a good candidate for cold water aquaculture. The aim of this paper is to present the work carried out in France since 1999: the selection of cod as a promising candidate for aquaculture and the assessment of its rearing performances in this environment. |

Pathogenic bacteria are known to be one of the main factors affecting Pecten maximus larval survival in hatcheries. As a result, juvenile production often relies on the use of antibiotics during larval culture. However, limitations of the usage of chemicals such as chloramphenicol in aquaculture have been reinforced due to their negative environmental impact and alternatives are accordingly needed. Thus, the importance of bacterial transfer from oocytes to larvae has been studied here as well as procedures to limit larval mortality in P. maximus. In order to reduce bacterial contamination during larval development, we focused on two periods, broodstock conditioning and post-fertilization. The animals were conditioned for 2 months with two erythromycin... |

Vibrios are frequently associated with oyster mortality; however whether they are the primary causative agent or secondary opportunistic colonizers is not well understood. Here we combine analysis of natural infection dynamics, population genomics and molecular genetics to ask (i) to what extent oysters are passively colonized by Vibrio population present in the surrounding water, (ii) how populations turn over during pathogenicity events and (iii) what genetic factors are responsible for pathogenicity. We identified several populations of Vibrio preferentially associated with oyster tissues. Among these, Vibrio crassostreae is particularly abundant in diseased animals while nearly absent in the surrounding water, and its pathogenicity is correlated with... |

During larval rearing phase of turbot, a substantial variability in survival and growth is frequently observed. In this paper, we studied the effects of hydrodynamics in larvae rearing tanks on the survival and the growth of fed and starved larvae, using the following conditions: water flows of 600 and 1200 ml.mn(-1), constant and progressive ones, air flows of 10 and 30 ml.mn(-1), downward and upward water circulation, with direction change during the rearing phase. We also studied the interactions between water circulation direction and feeding conditions by (1) shifting the first feeding day (4 occasions, at 8 hours intervals, from 52 to 76 hours after 95% of hatching) and (2) using 2 amounts of preys (which differ by a factor 3). The results show that... |

The survival of turbot eggs and the rearing capacities of larvae stemmed from artificial fertilization practices using frozen-thawed spermatozoa were evaluated. Furthermore, the viability of sperm samples stored during a 9 month period in liquid nitrogen was assessed. No significant difference in the fertilization rate, hatching rate, survival and wet weight of 10-day old larvae were observed using fresh or frozen-thawed spermatozoa. The motility recorded at 10 s and 60 s post-activation and the fertilization capacity of frozen-thawed spermatozoa were not significantly decreased during a 9 month storage period in liquid nitrogen. These results confirm the high quality of the turbot spermatozoa stemmed from the cryopreservation process, allowing their use... |

The first aim of this study was to determine the stocking density limits for Pacific oyster Crassostrea gigas larvae reared in flow-through system (FTS) and recirculating aquaculture systems (RAS). The second aim was to examine biofilm formation on the larval tank wall and its interaction with larvae growth. Three larvae concentrations were tested: 50, 150, and 300 mL−1. Chemical parameters and larvae performance were measured. The biofilm was observed by scanning electron microscopy, and its bacterial composition was investigated by pyrosequencing analysis of part of the 16S rRNA gene. The highest growth (13 µm day–1), survival (87%) and metamorphosis (50%) rates were observed in FTS at 50 larvae mL–1, while lower and similar performances occurred at... |

Since 2008, mass mortalities of 1-yr-old Crassostrea gigas associated with the ostreid herpesvirus OsHV-1 μVar have occurred along all the coasts of France when seawater temperature reaches 16 to 17°C. The present study aimed to characterize the effect of temperature on oyster survival in combination with OsHV-1 DNA quantification by standard real-time PCR and total vibrio population levels in oyster tissues. To examine the effect of seawater temperature on disease transmission and related mortality of oysters, cohabitation experiments were conducted between healthy naïve oysters and oysters previously exposed to field conditions in areas where mortalities were occurring. Oysters initially maintained in controlled conditions (free of mortality and negative... |

Diseases pose an ongoing threat to aquaculture, fisheries and conservation of marine species, and determination of risk factors of disease is crucial for management. Our objective was to decipher the effects of host, pathogen and environmental factors on disease-induced mortality of Pacific oysters (Crassostrea gigas) across a latitudinal gradient. We deployed young and adult oysters at 13 sites in France and we monitored survival, pathogens and environmental parameters. The young oysters came from either the wild collection or the hatchery while the adults were from the wild only. We then used Cox regression models to investigate the effect of latitude, site, environmental factors and origin on mortality risk and to extrapolate this mortality risk to the... |

Background: As a major threat to the oyster industry, Pacific Oyster Mortality Syndrome (POMS) is a polymicrobial disease affecting the main oyster species farmed across the world. POMS affects oyster juveniles and became panzootic this last decade, but POMS resistance in some oyster genotypes has emerged. While we know some genetic loci associated with resistance, the underlying mechanisms remained uncharacterized. So, we developed a comparative transcriptomic approach using basal gene expression profiles between different oyster biparental families with contrasted phenotypes when confronted to POMS (resistant or susceptible). Results: We showed that POMS resistant oysters show differential expression of genes involved in stress responses, protein... |

Over the last decade, innate immune priming has been evidenced in many invertebrate phyla. If mechanistic models have been proposed, molecular studies aiming to substantiate these models have remained scarce. We reveal here the transcriptional signature associated with immune priming in the oyster Crassostrea gigas. Oysters were fully protected against Ostreid herpesvirus 1 (OsHV-1), a major oyster pathogen, after priming with poly(I·C), which mimics viral double-stranded RNA. Global analysis through RNA sequencing of oyster and viral genes after immune priming and viral infection revealed that poly(I·C) induces a strong antiviral response that impairs OsHV-1 replication. Protection is based on a sustained upregulation of immune genes, notably genes... |

Since 2008, mass mortalities of 1-yr-old oysters Crassostrea gigas associated with ostreid herpes virus OsHV-1 μVar have occurred along all coasts of France. These mortality events represent the most serious crisis for the French oyster industry since the introduction of C. gigas in the early 1970s. Temperature is commonly one of the major triggering factors of disease epizootics, notably for aquatic species. In France, OsHV-1 is generally detected in dying oysters when seawater temperature is higher than 16° C. The influence of temperature on OsHV-1 detection and expression was also demonstrated for C. gigas larvae and suspected for juveniles. The present study aims to characterize the effect of temperature on disease transmission and survival related... |

Pacific oyster mortality syndrome affects juveniles of Crassostrea gigas oysters and threatens the sustainability of commercial and natural stocks of this species. Vibrio crassostreae has been repeatedly isolated from diseased animals and the majority of the strains have been demonstrated to be virulent for oysters. In this study we showed that oyster farms exhibited a high prevalence of a virulence plasmid carried by V. crassostreae while oysters, at an adult stage, were reservoirs of this virulent population. The pathogenicity of V. crassostreae depends on a novel transcriptional regulator, which activates the bidirectional promoter of a Type 6 Secretion System (T6SS) genes cluster. Both the T6SS and a second chromosomal virulence factor, r5.7, are... |
