The fishing of pollack which concerns essentially, France, United Kingdom and Norway, and at a lower level Ireland and Denmark, is situated between 10 and 15000 tons a year. In 1996, France was the first producer of Pollack. Since 1980, a decrease of production is observed in Europe. In France, landing reached 3000t. in 1998 (Suquet, 2001). However, the demand remaining strong, aquaculture could be a solution to satisfy the deficit between the French production and the domestic demand.

La domestication des poissons : le cas du thon rouge ArchiMer

Bluefin tuna shows adaptative physiology features allowing life in a wide range of physical and climatic parameters like temperature, salinity and hydrostatic pressure, hence the species fulfil the main basic requirements for progressive domestication and rearing in captivity. The high mortalities recorded after handling and the losses observed after abrupt variations of environment in the cages, emphasised the extreme sensitivity to stress of tuna. As consequences, the progresses about the control of tuna biological cycle are very slow due to limited handling possibilities and tuna aquaculture is restricted to fattening of wild fish. At this time, bluefin tuna production progresses without domestication of the species.

Setting tools for the early assessment of the quality of thawed Pacific oyster (Crassostrea gigas) D-larvae ArchiMer

Parameters used to assess the survival of larvae after cryopreservation generally misestimate the damages that prevent larval development. The objectives of the present study were to 1) define the reliability of the survival rate, assessed at 2 and 7 days post fertilization, to estimate Pacific oyster larval quality after thawing, and 2) select complementary tools allowing an early and reliable estimation of their quality. Sperm motility in European hake, Merluccius merluccius, and characterization of its spermatozoa concentration and volume, spermatocrit, osmolality and pH ArchiMer

Due to stock declines there is a growing interest in farming of European hake Merluccius merluccius. Thus, knowledge of its sperm biology is of importance not only for purposes of broodstock management, but also for the development of sperm preservation techniques. Hake sperm were collected from mature males caught during the summer-early autumn waters off western Norway and during the winter-early spring in the Bay of Biscay (France). Sperm quality characteristics were assessed after storage at 4 degrees C for 25 +/- 14 h. Effect of age and environmental conditions on the movement characteristics of Pacific oyster (Crassostrea gigas) trochophores ArchiMer

The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. Egg production in the turbot (Scophthalmus maximus) in relation to broodstock management ArchiMer

Egg production data from individual hand stripped female turbot were collected and analysed over several consecutive spawning seasons, on natural or shifted conditions. Egg quantity, egg viability and number of spawns were examined from one spawning season to the other, and inside the spawning season. These parameters seemed to increase from a year to the next one, except for the oldest fishes. Spawns were spread over a three-month period and occurred at the same time in the period in successive seasons for most females. Egg production seemed related to the feeding level and the earliness of spawning season appeared depended on the growth rate. Cryopreservation of sperm in marine fish ArchiMer

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Effect of temperature, volume of ova batches, and addition of a diluent, an antibiotic, oxygen and a protein inhibitor on short-term storage capacities of turbot, Psetta maxima, ova ArchiMer

The effect of different parameters on shea-term storage capacity of turbot ova was assessed over a 45-h period after ova collection for fertilization rates and over a 9-h period after ova collection for hatching rates. Increasing the volume of ova sampling from 0.5 to 2.5 mt, as well as adding an antibiotic-antimicotic solution or oxygen did not significantly change the storage capacity of ova. Regarding the hatching rates, a higher storage ability was recorded at 8 and 13 degrees C, compared to 3 degrees C. The mean composition of the ovarian fluid was determined (n = 57 spawns). After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. Individual monitoring of gonad development in the European flat oyster Ostrea edulis by in vivo magnetic resonance imaging ArchiMer

Previous studies have shown that magnetic resonance imaging is a very appropriate non-invasive technique for quantifying the growth of somatic and gonadic tissues and sex determination in the Pacific oyster, Crassostrea gigas. Despite a thinner gonad, harder to distinguish than that of the Pacific oyster, we showed in this study that it was possible to distinguish and quantify the development of the gonad of the European flat oyster, Ostrea edulis, using a 3D MRI sequence with a suitable finer resolution. The quality of oyster spermatozoa has become an issue for modern aquaculture. However, reliable protocols used to assess gamete quality are lacking. The aim of this work is to define a standardised experimental fertilisation protocol for Pacific oyster oocytes. Six experiments have been carried out in this study. Sexual dimorphism of drumming muscles in European hake (Merluccius merluccius) ArchiMer

Dissections of mature and non-mature European hake males and females (N=142) collected in waters off the western coast of Norway and in the Bay of Biscay (France) in 2004-2006 demonstrate for the first time that this gadoid species contains drumming muscles. There were differences in drumming muscles weight with body length, sex and maturity stage. This study shows that the difference between females and males is primarily manifested during the spawning season, seen both in the French and Norwegian samples. For the mature females the drumming muscles dry weight increases only slightly, if at all, with increase in total length. For mature males there is a corresponding rapid increase. Slow-release GnRHa treatment prevented atresia during vitellogenesis and induced ovulation of captive wreckfish (Polyprion americanus) ArchiMer

The potential of wreckfish (Polyprion americanus) to breed in captivity was examined. Ovarian activity varied seasonally, with vitellogenesis beginning in November and ending in May. Slow-release implant with gonadotropin-releasing hormone agonist (GnRHa) prevented atresia, and both GnRHa injections or implants at the end of vitellogenesis triggered ovulation after 7 days. Spontaneous spawning was obtained only in one female, and the few eggs produced were of poor viability. Manual stripping and artificial fertilization produced viable gametes and embryos. Therefore, present captivity conditions inhibited final oocyte maturation (FOM) and spawning, but not vitellogenesis and spermatogenesis. GnRHa treatments stimulate FOM and ovulation of viable eggs.

Cryobanking of aquatic species ArchiMer

Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Spermatozoa were cryopreserved with CPA 0.7 M trehalose in 0.8 M Me2SO and a two-step freezing process was used: straws were first maintained in nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and stored for one week before use.
