Antibodies against pseudorabies virus in feral swine in southeast Brazil Arq. Bras. Med. Vet. Zootec.

Serum samples collected from 358 wild boars (Sus scrofa) in breeding farms in São Paulo, southeast Brazil, from 1998 to 2001, were tested for antibodies against pseudorabies virus (PRV) by means of serum neutralization (SN) and enzyme-linked immunobsorbent assay (ELISA). Seropositive animals were detected in three of seven herds analyzed. Overall seroprevalence as assessed by SN was 30.7%, ranging from 25.2% to 100% for the herds that presented seropositive animals. Indirect ELISA detected lower seroprevalence (19.3%). Sensitivity and specificity of ELISA were equal to 57.3% and 97.6%, respectively. Agreement was equal to 85.2% (P<0.0001). Ocular alterations in dogs naturally infected by Leishmania (Leishmania) chagasi Arq. Bras. Med. Vet. Zootec.

Ocular conditions, anti-Leishmania antibodies and total protein of the aqueous humor were studied in dogs naturally infected by Leishmania (Leishmania) chagasi. Fifty dogs were analyzed and assigned into two groups of 25 animals each. All animals were submitted to routine ophthalmic exam. Results showed that 76% of the affected animals presented ocular signs, being uveitis the predominant. The mean of total protein in the aqueous humor of animals with uveitis was higher (P<0.05) when compared to the mean of animals with other ocular signs or no clinical sign. The anti-Leishmania chagasi antibody values in plasma were superior to those found in the aqueous humor (P<0.05).

O objetivo do presente estudo foi determinar a dinâmica da produção de anticorpos anti-Babesia equi e B. caballi em éguas gestantes em condições naturais de campo em Minas Gerais e avaliar a eficiência da transferência passiva de anticorpos via colostro. Foram analisadas, pela imunofluorescência indireta, amostras de soros colhidas de 34 fêmeas aos 290, 305 e 320 dias de gestação, e no momento do parto, e de seus respectivos potros até 36 horas após o nascimento. Aos 290 dias de gestação, todas as fêmeas estavam positivas para B. equi e apenas duas estavam negativas para B. caballi. Production of specific polyclonal antibodies anti-human factor IX BABT

In this work, polyclonal antibodies anti-human Factor IX were produced in New Zealand rabbits by immunization with commercial pure human FIX (hFIX) (Octanyne®, Octapharma, USA). The serum containing immunoglobulins anti-hFIX was useful to detect hFIX antigen in human plasma fractions submitted to anionic exchange chromatographic process and with a large yield. Immunoassays (ELISA) using bovine serum albumin, trypsin and peptides generated by cleavage assays with trypsin as digestion enzyme was performed and revealed adequate specificity of the polyclonal antibodies produced.

Production of rabbit antibodies against purified Glucose oxidase BABT

Glucose oxidase is an active oxygen species generating enzyme produced from Aspergillus niger grown in submerged fermentation. Disintegration of the mycelium resulted in high glucose oxidase activity that was subjected to ammonium sulfate precipitation at 60-85% saturation rates that resulted to 6.14 U mg -1 specific activity. Purification of enzyme by anion exchange column (DEAE-Cellulose) resulted into 22.53 U mg-1 specific activity and 10.27 fold purification. This was applied to sephadex G-200 column for gel filtration chromatography. It was observed that enzyme achieved 59.37 U mg-1of specific activity with 27.08 fold purity and 64.36% recovery. Assessment of the plan for pre-exposition vaccination with Fuenzalida-Palacios anti-rabies vaccine BABT

Studies ere carried out to monitor the titres of anti-rabies antibodies taken from individuals exposed to the risk of infection. Fifty-seven individuals aged between 20 and 40, were vaccinated with a minimum titre of 1.3 IU Fuenzalida-Palacios type anti-rabies vaccine. The vaccination plan consisted of the application of three doses at two day intervals and a booster applied 28 days after the final dose. The accompainment of the antibody titres was achieved by means of the seroneutralization test over a total period of 18 months. To investigate the exposure of the Newcastle disease virus (NDV), infectious bursal disease virus (IBDV) and avian poxvirus (APV) in Magellanic penguins found on the beaches in Southern regions of Brazil, the frequency of serum antibodies was estimated in 89 samples taken during 2005 and 2006. All the penguins were negative for the presence of antibodies against NDV by hemagglutination inhibition test and to APV by indirect ELISA. The reactivity was similar to the positives controls using ELISA kit for the IBDV made in the chickens in 50 samples. Seroprevalence of antibodies against group A rotavirus in cattle from a pioneer frontier in Brazilian Amazon Braz. J. Vet. Res. Anim. Sci.

Rotavirus is a worldwide etiologic agent of diarrhea, responsible for large economic losses. We studied the seroprevalence of antibodies to group A rotavirus in cattle in 67 smallholder farms from Uruará municipality, using counterimmunoelectroosmophoresis with the NCDV strain as a standard antigen. Prevalence of positive smallholder farms was 95.6-100%. Significant differences were seen between age groups when the seropositivity rose from the youngest to the oldest groups and between females and males older than 1 year, when the seropositivity was higher in the first group.

This study evaluated the vaccination response to Haemophilus influenzae type b (Hib) in malnourished pregnant women (MN), cord blood (CB) and in infants at two and six months of age for comparison with a control group (C). Twenty-eight malnourished pregnant women and 29 pregnant controls were immunized with conjugated Act-HIB® in the third trimester of pregnancy. Blood samples were collected from all before the immunization, during labor (post immunization), and from CB. Are anti-interferon antibodies the cause of failure in: chronic HCV hepatitis treatment? BJID

A follow-up study was made of 94 chronic hepatitis C patients at a hepatitis clinic in Brazil, after interferon alpha (IFN-alpha) therapy, to determine the influence of anti-interferon antibodies on treatment outcome. Patients diagnosed as having chronic hepatitis C, confirmed by PCR (HCV RNA) and liver biopsy, were treated with interferon alpha 2a or 2b for at least six months, and were followed up for 24 weeks after termination of treatment in order to assess biochemical, virological and clinical pathology responses. Epitope of dengue virus E protein detect human antibodies associated with mild disease: a potential peptide for vaccine development BJID

ABSTRACT The antigenic potential of seven immunogenic peptides of the dengue virus was evaluated in the sera of patients with dengue confirmed by IgM/IgG serology. Antibodies IgM and IgG against dengue virus peptides were analyzed by ELISA in 31 dengue sero-positive and 20 sero-negative patients. The P5 peptide showed significant IgG immunoreactivity mostly in the sera of patients with dengue without warning signs in comparison with patients with dengue with warning signs, correlating with mild disease. This finding suggests that the low antibody response against P5 epitope could be a risk factor for higher susceptibility to dengue virus infection with warning signs, and that P5 could be a potential antigen for vaccine development.

American cutaneous leishmaniasis (ACL) is an endemic disease caused by Leishmania parasites. The ACL diagnosis is commonly accomplished by parasitological and immunological methods. The objective of this work was to evaluate the enzyme immunoassay (EIA) for ACL laboratorial diagnosis. IgG antibodies against Leishmania (Viannia) braziliensis promastigotes were researched. For the method standardization 240 sera were used: 72 from patients with positive parasitological diagnosis, 38 from normal individuals and 113 from individuals with other pathologies. S. pneumoniae is a significant cause of community-acquired pneumonia in the elderly, and accounts for the majority of the pneumonia deaths among the elderly. We conducted this randomized double-blind study to evaluate the immune response to a 23-valent pneumococcal polysaccharide vaccine and the persistence of antibodies two years after the vaccination in an elderly population in Santiago, Chile. OBJECTIVES: Estimate the prevalence of immunity to poliomyelitis (anti-polio antibodies) in the city of São Paulo/Brazil through a population-based survey. METHODS: A quantitative and inductive method was used to draw a representative sample of the population. Randomization and stratification (based on sex, age and residence region) was done, and 1,059 individuals were studied on a home-visit basis (structured questionnaires and blood samples). Previous studies have demonstrated a stronger seroreactivity against some synthetic peptides responsible for inducing neutralizing antibodies in injecting drug users (IDU) compared to that of individuals sexually infected with HIV-1 (S), but the effectiveness in terms of the neutralizing ability of these antibodies has not been evaluated. Our objective was to study the humoral immune response of IDU by determining the specificity of their antibodies and the presence of neutralizing antibodies. The neutralization capacity against the HIV-1 isolate MN (genotype B), the primary HIV-1 isolate 95BRRJ021 (genotype F), and the seroreactivity with peptides known to induce neutralizing antibodies, from the V2 and V3 loops of different HIV-1 subtypes, were analyzed....
