Alarfaj,Abdullah A.; Arshad,Mohammed; Sholkamy,Essam N.; Munusamy,Murugan A.
ABSTRACTPolyhydroxybutyrate (PHB) is a renowned biodegradable plastic that do not release any toxins or residues in the environment like petroleum based plastics. In the present study, 50 bacteria isolated from mangrove niche, Saudi Arabia, were screened for maximum PHB production. All the 50 strains showed positive for PHB production, of which one strain showed maximum of 137 mgL-1. The most PHB accumulated bacterium was selected and identified asBacillus thuringiensis KSADL127, based on phenotypic characterization and 16S rRNA sequence analysis. Characterization of extracted PHB was carried out by FT-IR, NMR, UV spectroscopy, DSC, TGA, and LC-MS, which later confirmed the presence of intracellular accumulated polymer and substantiated as PHB.

Characterization of the mucosal and systemic immune response induced by Cry1Ac protein from Bacillus thuringiensis HD 73 in mice BJMBR
Vázquez-Padrón,R.I.; Moreno-Fierros,L.; Neri-Bazán,L.; Martínez-Gil,A.F.; de-la-Riva,G.A.; López-Revilla,R..
The present paper describes important features of the immune response induced by the Cry1Ac protein from Bacillus thuringiensis in mice. The kinetics of induction of serum and mucosal antibodies showed an immediate production of anti-Cry1Ac IgM and IgG antibodies in serum after the first immunization with the protoxin by either the intraperitoneal or intragastric route. The antibody fraction in serum and intestinal fluids consisted mainly of IgG1. In addition, plasma cells producing anti-Cry1Ac IgG antibodies in Peyer's patches were observed using the solid-phase enzyme-linked immunospot (ELISPOT). Cry1Ac toxin administration induced a strong immune response in serum but in the small intestinal fluids only anti-Cry1Ac IgA antibodies were detected. The data...

Detection of cry1 genes in Bacillus thuringiensis isolates from South of Brazil and activity against Aanticarsia gemmatalis (Lepidoptera:Noctuidae) BJM
Bobrowski,Vera Lucia; Pasquali,Giancarlo; Bodanese-Zanettini,Maria Helena; Fiuza,Lidia Mariana.
The bacterium Bacillus thuringiensis (Bt) is characterized by its ability to produce proteic crystalline inclusions during sporulation. Cry1 protein has insecticidal activity and is highly specific to certain insects and not toxic to unrelated insects, plants or vertebrates. In this work, the patogenicity of twelve Bt isolates was tested against Anticarsia gemmatalis, one of the most important insect pests of soybeans. Spore-crystal complex was applied to the surface of artificial diets and the mortality of A. gemmatalis larvae was assessed seven days after each treatment. When compared to a control Bt isolate known by its high toxicity to A. gemmatalis larvae, four novel Bt isolates exhibited even higher toxic activities against the insect, resulting in...

Methodology for fast evaluation of Bacillus thuringiensis crystal protein content BJM
Alves,Lúcia M. Carareto; Lemos,Manoel V. F..
The development of the production and use of Bacillus thuringiensis in Brazil at a commercial scale faces certain difficulties, among them the establishment of efficient methodologies for the quantitation of toxic products to be commercialized. Presently, the amount of toxin is given in percentage by analyzing the samples total protein content. Such methodology however, does not measure the actual amount of active protein present in the product, since most strains express different endotoxin genes and might even produce b-toxin. Since the various types of toxins exhibit different antigenic characteristics, this work has as objective the utilization of fast immunological techniques to quantify the level of crystal protein. Crystal protein produced by a...

Structural and Functional Analysis of Giant Strong Component of Bacillus thuringiensis Metabolic Network BJM
Ding,D.W.; Ding,Y.R.; Li,L.N.; Cai,Y.J.; Xu,W.B..
The purpose of this work was to study the giant strong component (GSC) of B. thuringiensis metabolic network by structural and functional analysis. Based on so-called "bow tie" structure, we extracted and studied GSC with its functional significance. Global structural properties such as degree distribution and average path length were computed and indicated that the GSC is also a small-world and scale-free network. Furthermore, the GSC was decomposed and functional significant for metabolism of these divisions were investigated by comparing to KEGG metabolic pathways.

Polanczyk,Ricardo Antonio; Silva,Rogério Fernando Pires da; Fiuza,Lidia Mariana.
The fall armyworm (Spodoptera frugiperda) is one of the most important pests of maize, causing up to 20% production losses when defoliation occurs near to flowering, or even complete destruction of plants. Among the alternatives to control this pest, the use of Bacillus thuringiensis (Bt) has gained attention due to its efficiency and low impact on natural enemies. Strains Bt dendrolimus HD 37, Bt aizawai HD 68, Bt kurstaki HD 73, Bt darmstadiensis HD 146, and Bt thuringiensis 4412 were tested against second instar larvae in in vivo assays. Suspensions of Bt aizawai HD 68 and Bt thuringiensis 4412, containing 3 x 10(8) cells/ml, induced mortality of 100% and 80.4%, respectively. To test virulence, cell concentrations of 8 x 10(5) to 3 x 10(8) cells/ml of...

Field persistence of Bacillus thuringiensis on maize leaves (Zea mays L.) BJM
Haddad,Marinéia de Lara; Polanczyk,Ricardo Antonio; Alves,Sergio Batista; Garcia,Marcelo de Oliveira.
The persistence slope of Bacillus thuringiensis (Bt) based products in the field is an important parameter to evaluate their efficacy. The half-life, estimated based on persistence slope parameters, is one of the most effective tools to select microbial pesticides. The aim of this research was to study the relationship between viability loss of Bt spores on maize leaves and their concentration, comparing it with field persistence. The experimental design was split-plot on time, composed by maize plants, in which three concentrations (half, normal and double doses) of a Dipel commercial formulation were applied. In each plot three leaves in the upper part of three plants where randomly selected. Samples of these leaves were collected 3 to 72 hours after...

Genes de Bacillus thuringiensis: uma estratégia para conferir resistência a insetos em plantas Ciência Rural
Bobrowski,Vera Lucia; Fiuza,Lidia Mariana; Pasquali,Giancarlo; Bodanese-Zanettini,Maria Helena.
