Optimisation de la méthode de quantification de l'infection à Bonamia ostreae chez l'huître plate Ostrea edulis par P.C.R. quantitative ArchiMer

Le Laboratoire de Génétique et de Pathologie a développé un programme de recherche concernant la bonamiose, maladie due au protiste Bonamia ostreae. Le diagnostic de la maladie repose essentiellement sur deux techniques (histologie et la cytologie par apposition de tissus cardiaque), cependant la sensibilité de ces technique reste relativement faible. C'est pourquoi il est nécessaire de mettre au point une technique plus sensible, afin de pouvoir établir un diagnostic plus précoce de la maladie. Bonamia ostreae and Ostrea edulis : a stable host-parasite system in France? ArchiMer

Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. After its first description in June 1979 in L'Ile Tudy, Brittany, the parasite rapidly spread to all French oyster farming areas and in other European countries through transfers of live molluscs. Bonamiosis and marteiliosis, another protozoan disease appeared in the seventies, drastically reduced the French flat oyster production from 20 000 tonnes in 1970 to less than 2000 tonnes after 1981. The history of the French oyster production highlights the fragility of this production against overexploitation and disease outbreaks. In particular, the production of flot oyster, Ostrea edulis, has decreased following the emergence of two parasitic diseases including bonamiosis. The means to fight against bonamiosis are relatively limited. They are mainly based on oyster health surveillance to limit the spread of the disease. However, the use of predictive models of disease progression in infected area would help to improve stock management and minimize the impact pathogens. Infection with the protozoan parasite Bonamia ostreae modifies in vitro haemocyte activities of flat oyster Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan parasite, infecting haemocytes of the European flat oyster Ostrea edulis. Oyster defence mechanisms mainly rely on haemocytes. In the present study in vitro interactions between parasites and flat oyster haemocytes were investigated using flow cytometry and light microscopy. Haemocyte parameters including: non specific esterase activity, reactive oxygen species (ROS) production and phagocytosis were monitored using flow cytometry after 2 hour cell incubation with live and dead B. ostreae. The epizootiological aspect, the impact of diseases on Breton oyster farming and the research for appropriate prophylaxes is addressed. The morphology, the ultrastructure and certain phases of the growth cycle are described for the two parasites, as well as their cyto- and histopathogenic effects. Two successive diseases struck the flat oyster Ostrea edulis farms in Brittany. These two epizooties lead to some changes in the farming practices. The oyster culture developed towards the open ocean, in the sublitoral part. New techniques have been developed for the culture and collection. The flat oyster has been partially replaced by the cupped oyster Crassostrea gigas whose production went from 3 000 to 16 000 tonnes. They also had heavy consequences on the regional economy. The important mortalities decimated the stocks. The production dropped by about 17 000 tonnes from 1910 to 1982. The parasite, Bonamia ostreae, was diagnosed in several South Brittany flat oyster's samples, from gills tissues's prints, stained in May-Grunwald-Giemsa and from corresponding digestive glands sections. The comparaison of results shows quantitative differences relating to the infested oyster's rate, and qualitative differences relating to the early stages of infestation. Bonamiosis spread rapidly among oysters farming in France and Europ. Another species, Bonamia sp. has been detected in New Zealand and Australia. The protozoan Bonamia ostreae (Ascetospora) is the etiological agent of bonamiosis, or hemocyte disease of the European fiat oyster, Ostrea edulis (Pichot et al., 1980). Since this infection was first reported from the coast of Brittany (Comps et al., 1980), the parasite has spread rapidly along the French Atlantic coasts (Grizel & Tigé, 1982), and in the European countries including the UK, Holland, lreland and Spain (Bannister & Key, 1982; Van Banning, 1982; Polanco et al., 1984). Whole-genome amplification: a useful approach to characterize new genes in unculturable protozoan parasites such as Bonamia exitiosa ArchiMer
