Disseminated candidiasis is a serious problem in public health that results from the invasion of Candida species, in particular Candida albicans. The aim of this study was to compare the efficacies of Zataria multiflora essential oil and itraconazole in clearing C. albicans from the visceral organs of BALB/c mice suffered from disseminated candidiasis. Zataria multiflora essential oil was extracted using Clevenger-type apparatus and analyzed by gas chromatography mass spectrometry (GC-MS). Analysis of the in vitro adherence of Streptococcus mutans and Candida albicans BJM

The objective of the present study was to investigate the in vitro adherence capacity of Streptococcus mutans and Candida albicans. Adherence assays were conducted on dental surfaces and analyzed by scanning electron microscopy (SEM). Extracted human teeth were inoculated with Streptococcus mutans or Candida albicans and with both species simultaneously, and incubated at 37ºC for 21 days. Biotyping and genotypic diversity among oral Candida albicans strains from caries-free and caries-active healthy children BJM

Candida albicans oral strains collected from caries-free and caries-active healthy children ranging from 24 to 36 months old, were studied. The aim of the study was to determine proteinase and phospholipase activities produced by Candida albicans in the two groups and to determine the phenotypic diversity of these enzymes based on genetic polymorphism using the AP-PCR method. The genetic diversity of C. albicans oral isolates from 75 healthy schoolchildren from eight schools located in different geographic areas of Piracicaba city, São Paulo state, Brazil, was established using isoenzymes marker (Multilocus Enzyme Electrophoresis - MLEE) and cluster analysis. Candida dubliniensis is an emerging pathogen capable of causing superficial as well as systemic infections. Due to its close similarity to C. albcians, conventional methods based on phenotypic traits are not always reliable in identification of C. dubliniensis. In this study, we developed a PCR-restriction fragment length polymorphism (RFLP) assay to identify and discriminate between the two closely related species. EXPERIMENTAL MODEL OF CANDIDA ALBICANS (SEROTYPES A AND B) ADHERENCE IN VITRO BJM

The adherence of Candida albicans is considered an important virulence factor since it determines the colonization of the yeast in various regions of the host, being the initial cause of the infection. In this work we studied the adherence of C. albicans (serotypes A and B) to HeLa and Vero cells after culture for 18 h and 30 h in chemically defined medium (YNB) plus carbohydrate sources such as glucose, sucrose, maltose, galactose and mannose (500 mM). Each assay was performed in triplicate. The best results were obtained with the medium containing glucose. The adherence was greatest in 18 h cultures at 37ºC and at 1 h contact time. The interaction of yeast cells with the host cells was studied by scanning microscopy.

In current research, nine basic esters of para-alkoxyphenylcarbamic acid with incorporated 4-(4fluoro-/3-trifluoromethylphenyl)piperazin-1-yl fragment, 6i-6m and 8f-8i, were screened for their in vitro antimicrobial activity against Candida albicans, Staphylococcus aureus and Escherichia coli, respectively. Several molecular methods, such as Southern blotting hybridization, Multilocus Sequence Typing, and DNA microsatellite analysis, have been employed to genotype Candida albicans. The genotype analysis allows to group strains in clades, that is, a group composed of one ancestor and its descendants. These genotype studies demonstrate that clades distribution is influenced by geographic area as well as that antifungal resistance is associated with particular clades. These findings suggested that C. albicans reproduces mainly in a clonal manner, with certain degree of DNA microevolution. With the purpose to evaluate Candida albicans virulence in vivo, two strains selected were based on their phospholipase and proteinase activity, and used in a model of experimental infection. One strain, isolated from vaginal secretion, was stocked at the URM Culture Collection for 43 years and presented high phospholipase (Pz=0.217) and proteinase (1.386 U.mL-1) activity. The other strain was a fresh strain isolated from oropharyngeal secretion of an AIDS patient, and presented low phospholipase (Pz=0.482) and proteinase (0.780 U.mL-1) activity. Isolation of Candida spp. from denture-related stomatitis in Pará, Brazil BJM

ABSTRACT The aim of this study was to isolate and identify Candida species from the oral cavity of denture wearers with denture-related stomatitis who were attended at the University Federal of Pará (Belém City, Pará State, Brazil). A total of 36 denture wearers with denture-related stomatitis were included, and type I (50%), type II (33%) and type III (17%) stomatitis were observed. Candida spp. were isolated from 89% of the cases and included five different Candida species. In the present investigation, the basic esters of meta-alkoxyphenylcarbamic acid bearing variously substituted N-phenylpiperazine fragment were screened for their in vitro antimicrobial activity against Staphylococcus aureus, Escherichia coli and Candida albicans, respectively. The most effective against Escherichia coli was found the compound 6d (MIC=195,3 μg/mL) bearing simultaneously para-fluoro substituent at the 4‑phenylpiperazin-1-yl core and meta-methoxy side chain in the lipophilic part of the molecule. Scanning electron microscope (SEM) observations were used to analyze particular morphologies of Candida albicans clinical isolate (strain 82) and mutants defective in hyphae-promoting genes EFG1 (strain HLC52) and/ or CPH1 (strains HLC54 and Can16). Transcription factors Efg1 and Cph1 play role in regulating filamentation and adhesion of C. albicans' morphologies. Propolis has been used in traditional medicine for many centuries because of its benefcial health properties, including its antimicrobial capacity. Prosthesis stomatitis affects a signifcant percentage of users of removable dentures; Candida albicans is the most common fungal species associated with the development of this pathology.
