The gram-positive spore-forming bacteria, Bacillus thuringiensis (Bt) strains produced novel cellulases which could liberate glucose from soluble cellulose, carboxymethyl cellulose (CMC), and insoluble crystalline cellulose. The maximal cellulase activities were obtained after 60 hrs incubation at 28ºC in a LB broth medium with 1% CMC. Maximum CMCase activities were got at 40ºC and pH 4.0, respectively, and more than 50% of its maximal activity was retained at 40-60ºC for 1 hr, while approximately 40% of its maximal activity was also retained after incubating at 70ºC for 1 hr. Most metal ions and reagents such as Ca2+, Mg2+, Cd2+, Pb2+, Zn2+, Cu2+, EDTA, and SDS inhibited the enzyme activities, but K+ and Mn2+ activated the activities. The enzymes from... |

Aberdeen Angus beef cattle from the Brazilian herd were studied genetically using restriction fragment length polymorphism (RFLP) of the kappa-casein - HinfI (CSN3 - HinfI), beta-lactoglobulin - HaeIII (LGB - HaeIII) and growth hormone AluI (GH- AluI) genes, as well as four microsatellites (TEXAN15, CSFM50, BM1224 and BM7160). The RFLP genotypes were determined using the polymerase chain reaction (PCR) followed by digestion with restriction endonucleases and electrophoresis in agarose gels. The occurrence of hardseedness and the low percentage of seed germination are major challenges when growing okra. For this reason, five elite lines of okra developed at the Campinas Agronomic Institute (IAC), three with angular pods (lines 8.1, 20.1.2 and 47.1) and two with cylindrical pods (lines 13.1.2 and 20.1.R), were evaluated for seed quality and compared to two commercial seed samples (Horticeres Sementes and Sakata Seed Sudamerica) of cultivar Santa Cruz 47. ABSTRACT In a collection of costeño peppers conserved by indigenous producers from the municipalities of Santa Maria Tonameca and Santo Domingo de Morelos, Oaxaca, Mexico, a description and classification of agromorphological variation was undertaken by characterization of 46 populations in a greenhouse distributed under complete randomized block design with three replicates. Resumo: Esta publicação tem por objetivo descrever a história, a composição da Coleção de trabalho de Microrganismos Patogênicos a Caprinos e Ovinos, e de que forma ela tem conservado, caracterizado, documentado e utilizado seus isolados. A coleção foi criada em 2012 e atualmente é composta por 141 cepas bacterianas e seis cepas virais, pertencentes às espécies Corynebacterium pseudotuberculosis, Staphylococcus aureus e ao gênero Lentivirus. Polysaccharides are highly heat-sensitive macromolecules, so high temperature treatments are greatly destructive and cause considerable damage, such as a great decrease in both viscosity and molecular weight of the polymer. The technical feasibility of the production of exopolysaccharides by deep-sea bacteria Vibrio diabolicus and Alteromonas infernus was previously demonstrated using a bioproduct manufacturing process. This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Marine bacteria are a rich source of bioactive metabolites. However, the microbial diversity of marine ecosystem still needs to be explored. The aim of this study was to isolate and characterize bacteria with antimicrobial activities from various marine coastal environment of New Caledonia. We obtained 493 marine isolates from various environments and samples of which 63 (12.8%) presented an antibacterial activity against a panel of reference pathogenic strains (Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Enterococcus faecalis).
