Seed storage proteins in coffee Rev. Bras. Fisiol. Veg.
BAÚ,SANDRA M. T.; MAZZAFERA,PAULO; SANTORO,LUIZ G..
It has been reported that Coffea arabica seeds contain as the main reserve protein, a legumin-like protein, constituted of two subunits, alpha and beta, of approximately 35 and 20 kDa. In this work the seed proteins of several coffee species and varieties were investigated by SDS-PAGE and gel filtration. No differences were observed in the electrophoretic profiles among varieties of C. arabica, however, marked differences were observed among species, or even among individuals of some species. In general, the molecular weight of the subunits alpha and beta accounted for a monomer of 48 to 62 kDa. However, native molecular weight obtained by gel filtration showed that for most of the species there is association of 6 of such proteins, in a hexamer. The most...

Ploidy level and caffeine content in leaves of Coffea Scientia Agricola
Silvarolla,Maria Bernadete; Mazzafera,Paulo; Lima,Marinez Muraro Alves de; Medina Filho,Herculano Penna; Fazuoli,Luiz Carlos.
Several species of Coffea and varieties of C. arabica differing in chromosome number had their caffeine content determined in the leaves. For Coffea arabica var. "Angustifolia", var. "Caturra" and var. "Icatu", it was observed caffeine decrease from the haploid (2n = 22) to tetraploid leaves (2n = 44). Caffeine in the tetraploid "Angustifolia" was decreased by 50% when compared to the haploid. Caffeine reduction was also observed in leaves of C. canephora var. "Kouilou" as chromosome number was increased (2n = 22 ® 2n = 44). In this case, caffeine in the leaves of the diploid genotype was close to 4 times higher than in the tetraploid. On the other hand it was observed an increase of the alkaloid when the chromosome number was doubled in the C. canephora...

OIL CONTENT OF GREEN BEANS FROM SOME COFFEE SPECIES Bragantia
MAZZAFERA,PAULO; SOAVE,DAYSE; ZULLO,MARCO ANTONIO TEIXEIRA; GUERREIRO FILHO,OLIVEIRO.
The oil content was determined in seeds of several continental African species of the coffee germplasm bank of Instituto Agronômico de Campinas, State of São Paulo, Brazil. Oil was extracted from seeds with hexane in Soxhlet apparatus. Due to the economic importance, C. arabica and C. canephora have been the best studied species concerning oil content and composition, and the results obtained are in agreement with the reported in the literature. On the other hand, only one report in the literature describes the results of oil analyses in other few species of the African continent, although it does not allow comparison with our results. The oil content of most of the species varied from 9 to 15%, therefore, similar to the range observed for C. arabica and...

Queiroz-Voltan,Rachel Benetti; Cabral,Luciane Perosin; Fazuoli,Luiz Carlos; Paradela Filho,Osvaldo.

Medina Filho,Herculano Penna; Carvalho,Alcides; Ballve,Rosa Maria Lizana; Bordignon,Rita; Silvarolla,Maria Bernadete; Lima,Marinez Muraro Alves de; Fazuoli,Luiz Carlos.

Cytogenetics of coffee Braz. J. Plant Physiol.
Pinto-Maglio,Cecília A. F..
The genus Coffea L. has around 100 native species distributed in tropical and subtropical areas in Africa, and the most important economic species are C. arabica and C. canephora. C. arabica is exceptional in the genus since it is the only species so far analyzed which is self-compatible, and a natural polyploid with 2n=4x=44 chromosomes; it is considered to be a segmental allopolyploid because it presents a disomic inheritance and a regular meiotic behavior. All other species in the genus are self-incompatible diploids with 2n=2x=22. Cytogenetic studies in Coffea, undertaken since 1912, have followed various phases: initial studies were limited only to establishing chromosome counts. Subsequent studies characterized the karyotypes of various species using...

Impacts of drought and temperature stress on coffee physiology and production: a review Braz. J. Plant Physiol.
DaMatta,Fábio M.; Ramalho,José D. Cochicho.
Overall, drought and unfavourable temperatures are the major climatic limitations for coffee production. These limitations are expected to become increasingly important in several coffee growing regions due to the recognized changes in global climate, and also because coffee cultivation has spread towards marginal lands, where water shortage and unfavourable temperatures constitute major constraints to coffee yield. In this review, we examine the impacts of such limitations on the physiology, and consequently on the production of mainly Coffea arabica and C. canephora, which account for about 99 % of the world coffee bean production. The first section deals with climatic factors and the coffee plant’s requirements. The importance of controlling oxidative...

The lipid fraction of the coffee bean Braz. J. Plant Physiol.
Speer,Karl; Kölling-Speer,Isabelle.
The lipid fraction of coffee is composed mainly of triacylglycerols, sterols and tocopherols, the typical components found in all common edible vegetable oils. Additionally, the so-called coffee oil contains diterpenes of the kaurene family in proportions of up to 20 % of the total lipids. Diterpenes are of interest because of their analytical and physiological effects. The composition of the main lipid components of the two most important coffee species, Coffea arabica and Coffea canphora var. Robusta is presented. In addition, the influences of typical processes like roasting and steaming on selected lipid components as well as the effects of the storage of green coffee beans under different conditions will be described. Furthermore, new findings...

Metabolism of alkaloids in coffee plants Braz. J. Plant Physiol.
Ashihara,Hiroshi.
Coffee beans contain two types of alkaloids, caffeine and trigonelline, as major components. This review describes the distribution and metabolism of these compounds. Caffeine is synthesised from xanthosine derived from purine nucleotides. The major biosynthetic route is xanthosine -> 7-methylxanthosine -> 7-methylxanthine -> theobromine -> caffeine. Degradation activity of caffeine in coffee plants is very low, but catabolism of theophylline is always present. Theophylline is converted to xanthine, and then enters the conventional purine degradation pathway. A recent development in caffeine research is the successful cloning of genes of N-methyltransferases and characterization of recombinant proteins of these genes. Possible biotechnological...
