Bovine Oocytes and Early Embryos Express mRNA Encoding Glycerol Kinase but Addition of Glycerol to the Culture Media Interferes with Oocyte Maturation OAK
OKAWARA, Sumika; HAMANO, Seizo; TETSUKA, Masafumi; 手塚, 雅文.
Glycerol plays multi-functional roles in cellular physiology. Other than forming the backbone molecule for glycerophospholipid and triglyceride (TG), glycerol acts as an energy substrate for glycolysis. Spermatozoa are known to utilize glycerol for energy production, but there are no reports of this in oocytes. In this study, the value of glycerol as an energy substrate for bovine oocyte maturation (Exp. 1) and the gene expression of glycerol kinase (GK), an enzyme crucial for cellular glycerol utilization, in bovine oocytes and early embryos (Exp. 2) were examined. In Exp. 1, in vitro maturation (IVM) was conducted using synthetic oviduct fluid supplemented with/without glucose (1.5 mM) and/or glycerol (1.0 mM), and maturation rate, degree of cumulus...

Preimplantation Development of Embryos in Labrador Retrievers OAK
ABE, Yasuyuki; SUWA, Yoshinori; YANAGIMOTO-UETA, Yoshiko; SUZUKI, Hiroshi; 鈴木, 宏志.
Preimplantation development of canine embryos is not well understood. To understand the timing of preattachment embryogenesis relative to the luteinizing hormone (LH) surge, early embryonic development was examined in Labrador Retrievers after artificial insemination. The embryos migrated from the oviduct to the uterus beginning on day 11 after the LH surge. This transport must be completed within 24 h. By day 13 after the LH surge, all of the embryos had moved and were localized in the uterus. The embryos developed to the morula stage within 11-13 days and to the blastocyst stage within 14 days after the LH surge, respectively. These findings add to the current understanding concerning the physiology of preimplantation development and should help further...

Bovine embryo induces an anti-inflammatory response in uterine epithelial cells and immune cells in vitro: Possible involvement of interferon tau as an intermediator OAK
Talukder, Anup K.; Yousef, Mohamed S.; Rashid, Mohammad B.; Awai, Kensuke; Acosta, Tomas J.; Shimizu, Takashi; Okuda, Kiyoshi; Shimada, Masayuki; Imakawa, Kazuhiko; Miyamoto, Akio.
Recent observations suggest that the bovine uterus starts to react to the early embryo immediately after its arrival from the oviduct. The present study aimed to investigate the effect of the early developing embryo on the immune-related gene profile in bovine uterine epithelial cells (BUECs) in vitro, and to further examine the impact of conditioned media (CM), either from embryo-BUEC co-culture or embryo culture alone, on gene expression in peripheral blood mononuclear cells (PBMCs). First, BUECs were co-cultured with morulae (n = 10) for D5-D9 (D0 = IVF), and gene expression in BUECs was analyzed. Subsequently, PBMCs were cultured in CM from embryo-BUEC co-culture or D5-D9 embryo culture, and gene expression was evaluated. In BUECs, the embryo induced...

Viability and vigor of jamun (Syzygium cumini) seeds Rev. Bras. Bot.
Sivasubramaniam,K.; Selvarani,K..
Jamun (Syzygium cumini L. Skeels) (Black plum, Damson plum) fruits weigh between 2-5 g at maturity. Fresh seeds represented 20-80% of the total fruit weight; the seed coat and cotyledons contributed 6% and 94% to the total seed weight respectively, while the weight of the embryonic axis was insignificant. Only the embryonic axis stained with Tetrazolium, not the cotyledons. The seeds are polyembryonic with up to four embryos, of which at most three germinate. Decoated seeds germinated faster than coated seeds under nursery conditions, with high significant germination percentages, dry matter production rates and vigor indices. The lack of staining of the cotyledon by tetrazolium was probably due to the presence of an impermeable layer. Decoating seeds for...

Effect of breeder age on eggshell thickness, surface temperature, hatchability and chick weigh Rev. Bras. Ciênc. Avic.
Gualhanone,A; Furlan,RL; Fernandez-Alarcon,MF; Macari,M.
Two experiments were carried out to study the effect of breeder age on incubation parameters (hatchability, eggshell thickness, egg surface temperature and chick weight). In Exp. 1, fertile eggs (30- and 60-wk-old breeders) were incubated at three different temperatures (36.8, 37.8 and 38.8 ºC). Eggshell surface temperature was measured by attaching a thermocouple to the shell and data were collected in a datalogger every ten minutes. This study was conducted according to a 3 x 2 factorial design (three temperatures and two breeder ages). Data revealed that eggshell surface temperature changed according to incubation temperature, with the main increase occurring between 10 and 13 days of incubation, and that the maximum increase in eggshell surface...

Effect of Broiler Breeder Age on the Intestinal Mucosa Development of the Embryos at 20 Days of Incubation Rev. Bras. Ciênc. Avic.
Maiorka,A; Silva,AVF; Santin,E; Bruno,LDG; Boleli,IC; Macari,M.
ABSTRACT This study investigated the effect of broiler breeder age on the morphological development of the small intestine broiler embryos (villus height, crypt depth, microvillus height, and villus density) at 20 day of incubation. Eggs obtained from 30- and 60-wk-old broiler breeders were used. The results showed that embryos from older broiler breeders presented longer villi in the duodenum, jejunum, and ileum compared with younger broiler breeders. In addition, embryos from older broiler breeders presented deeper crypts in the jejunum and ileum, longer microvilli in jejunal enterocytes, and lower villus density (microvillus number/mm2) in the duodenum and ileum than younger breeders. These results suggest that breeder age influences the gut mucosa...

Onbaşılar,EE; Güngör,ÖF; Demir,T; Erbay,FK Elibol; Kahraman,M; Taban,S; Çalık,A; Pirpanahi,M.
ABSTRACT Layer hybrids are more superior in the egg yield than pure breeds. But this superiority causes some problems in the bones during the production period. There are a lot of differences among genotypes in the production period that have been examined extensively, however, the differences throughout incubation period aren’t known yet. Thus, the objective of this study was to evaluate the differences in femur properties of pure breed and layer hybrid embryos during the incubation period. A total of 354 fresh hatching eggs were used from one hybrid (Lohman White) and two pure breeds (Denizli & Gerze). The eggs were incubated. Hatching eggs from each genotype at the beginning of embryonic days (E) 19 and E21 were examined. At these embryonic ages, 12...

Identification of plantlet genetic origin in polyembryonic mango (Mangifera indica, L.) cv. Rosinha seeds using RAPD markers Rev. Bras. Frutic.
Cordeiro,Maria Cristina Rocha; Pinto,Alberto Carlos Queiróz; Ramos,Víctor Hugo Vargas; Faleiro,Fábio Gelape; Fraga,Lilia Marta Santos.
Mango can be propagated by seeds or by grafting. For commercial purpose, grafting is the most appropriate method because it maintains the genetic characters from the propagated variety. To obtain grafted mango it is important to use polyembryonic varieties as rootstock since they produce a zygotic and many nucellar plantlets. The nucellar plantlets maintain the genetics of the mother-plant thus, are preferred for grafting since they supposedly give more uniformity to the orchard. In general, nurserymen use the most vigorous plantelet to graft, believing that they are nucellar. But, orchard disuniformities on height and yield are very common among mango trees of commercial orchards in Northeast region. The objective of this paper was to identify the...

Relationship between eggplant seed morphology and germination Rev. bras. sementes
Silva,Vanessa Neumann; Cicero,Silvio Moure; Bennett,Mark.
Structural differences such as abnormalities, damage and free spaces in seeds may affect germination. The aim of this study was to study the relationship between eggplant seed morphology and seed germination. Ten seed lots of the eggplant cultivar Embu were evaluated by X-ray image analysis and the germination test. Seed image analysis was performed by Image Pro Plus® software and the whole seed area and free space between the embryo and endosperm were measured. The internal seed area filled by the embryo and endosperm was calculated from the difference between the whole seed and free space areas. Based on these results and visual seed analysis, seeds were classified into three categories and information on germination was obtained for each one. X-ray...

In vitro embryo production in small ruminants R. Bras. Zootec.
Freitas,Vicente José de Figueirêdo; Melo,Luciana Magalhães.
This paper reviews the technical bases of in vitro embryo production in small ruminants with special attention to the results obtained by our group in Northeastern Brazil. The laparoscopic oocyte recovery in hormonally treated live animals indicates a promising future for the application of this technique to genetic improvement program. New molecular biology tools should provide information to improve the efficiency of in vitro maturation. Furthers efforts have to be made to improve the oocyte maturation and to standardize the semen-capacitating process.
