A New Glider-Compatible Optical Sensor for Dissolved Organic Matter Measurements: Test Case from the NW Mediterranean Sea ArchiMer
Cyr, Frédéric; Tedetti, Marc; Besson, Florent; Beguery, Laurent; Doglioli, Andrea M.; Petrenko, Anne A.; Goutx, Madeleine.
The MiniFluo-UV is a new glider-compatible optical sensor for measurements of dissolved organic matter (DOM) in natural waters. The working principle, sensor design and challenges faced during the validation phase are reported. The first in situ application of the sensor during three glider deployments in the NW Mediterranean sea (spring, summer, and fall) are also presented. For these campaigns, the two channels of the sensor were adjusted to target Tryptophan-like (excitation/emission wavelengths λEx/λEm: 275/340 nm) and Phenanthrene-like (λEx/λEm: 255/360 nm) fluorescence. These were chosen because they represent fluorophores of interest commonly found in seawater. While Tryptophan (an amino-acid believed to be a by-product of biological activity) is...

Hydrophobicity of an Entisol under loblolly pine (Pinus taeda) plantation PFB - Pesquisa Florestal Brasileira
Maia, Claudia Maria Branco de Freitas; Fukamachi, Cristiane Regina Budziak; Cambronero, Yorleni Chang; Dedecek, Renato Antônio; Mangrich, Antônio Sálvio; Narimoto, Kelly Mayumi; Milori, Débora M.B.P.; Simões, Marcelo Luiz.
The understanding of soil carbon stabilization processes can be very useful in the development of mitigation techniques for CO2 emissions and global warming. The greater the hydrophobicity of soil organic matter the more stabilized soil organic carbon. Therefore,  hydrophobicity can be a sensitive index to characterizethe ‘quality’ of soil organic matter. In this context, the present work aimed to characterize the chemical  structures of humic acids collected at three different depths in a hydrophobic Entisol (Neossolo) under  loblolly plantation. The results of spectroscopic and chemical analyses (UV-Vis, fluorescence, EPR and X-ray diffractometry) indicated that, as soil depth increased, so did the content of conjugated organic structures, aromatic...

Physiologic parameters of soybean of determinate and indeterminate growth habit subjected to levels of soil moisture PAB
Machado Júnior,Carlos Sebastião; Silva,Cláudio Ricardo da; Sanches,Maria Cristina; Hamawaki,Osvaldo Toshiyuki; Sousa,Larissa Barbosa de.
Abstract: The objective of this work was to evaluate the physiological and production parameters of soybean (Glycine max) cultivars of determinate and indeterminate growth habits, in three levels of soil moisture. Three cultivars of determinate growth habit and three of indeterminate growth, at the beginning of the reproductive stage, were subjected to three levels of soil moisture: control, moisture at field capacity; moderated, 50% of the control level; and severe, 25% of the control level. Leaf gas exchange, xylem-water potential, chlorophyll content, chlorophyll a fluorescence, plant dry matter, and grain yield per plant were evaluated. The cultivars of indeterminate growth habit showed the greatest values for stomatal conductance and CO2 assimilation...

Localization and compartmentation of Al in the leaves and roots of tea plants Phyton
Hajiboland,R; Poschenrieder,C.
Under acid soil conditions, solubility of aluminum (Al) increases leading to toxicity for plants. Al accumulator species such as tea, however, accumulate high levels of Al in tissues without toxicity symptoms. In this work, Al localization and compartmentation were studied in tea [Camellia sinensis (L.) O. Kuntze] grown hydroponically at 0 or 100 µM Al for eight weeks. Plant dry matter production was significantly higher in the presence of Al and accumulated up to 1.21 and 6.18 mg Al/g DW in the leaves and roots, respectively. About 40-50% of Al was partitioned into cell wall (CW)-bound fraction without any difference among leaves of different age and roots. A significant increase of the soluble phenolics fraction by Al was observed in both leaves and...

Equipment based on high power UV and white light LEDs to collect and observe scorpions (Arachnida: Scorpiones) and other fluorescent organisms Rev. Bras. Zool.
Ramires,Eduardo N.; Peracetta,Luiz F.; Nogas,Cláudio; Navarro-Silva,Mário A.; Paladini,Edson P..
We introduce a new, high quality, low cost and versatile LEDs-based handset device that emits high power UV and white light, which can be used interchangeably. It offers power control and has long battery life. Even though it is optimized to detect and collect scorpions under low light conditions, it can also be used with other groups of fluorescent organisms. The device achieved superior performance in field and laboratory trials when compared with a 12 LEDs low power UV flashlight, and a 46 W black fluorescent light lamp, to locate Tityus serrulatus Lutz & Mello, 1922.

Canellas,Luciano Pasqualoto; Espindola,José Antonio Azevedo; Rezende,Carlos Eduardo; Camargo,Plínio Barbosa de; Zandonadi,Daniel Basílio; Rumjanek,Victor Marcos; Guerra,José Guilherme Marinho; Teixeira,Marcelo Grandi; Braz-Filho,Raimundo.
Using herbaceous legumes in agricultural systems yields great quantities of plant residues, allowing changes in soil organic matter quality and content over the years. This study was conducted on an Ultisol, at Seropédica, RJ, Brazil, to evaluate the effects of different perennial herbaceous legumes on soil organic matter quality. A factorial scheme with three replications was used to evaluate the species: forage groundnut cv. BR-14951 (Arachis pintoi), tropical kudzu (Pueraria phaseoloides), and siratro (Macroptilium atropurpureum). After the first cut, each plot was divided into two subplots; plants were cut and left on the soil surface or cut and removed. Soil samples of a closed area covered by spontaneous vegetation (mainly C3 plants) or by Panicum...

Effect of salinity and priming on seedling growth in rapeseed (Brassica napus var oleifera Del.) - doi: 10.4025/actasciagron.v35i4.17655 Agronomy
Benincasa, Paolo; University of Perugia; Pace, Roberta; University of Perugia; Quinet, Muriel; Université Catholique de Louvain; Lutts, Stanley; Université Catholique de Louvain.
Experiments were performed to examine the effect of salt stress and GA3-priming on initial growth of two rapeseed cultivars, one tolerant and one sensitive to salt stress during germination. Seedlings from seeds germinated in salty (as NaCl) and non salty substrate were grown in salty and non salty hydroponics. Salt stress reduced seedling growth of the two genotypes consistently with their degree of stress tolerance during germination. Seedlings from stress sensitive seeds germinated under high salinity showed a rapid recover of growth in non stressing conditions. The effect of salt stress on shoot/root ratio was controversial, increased for lab and decreased for greenhouse experiments, probably due to different timing of stress application and additional...

Chlorophyll a spatial inference using artificial neural network from multispectral images and in situ measurements Anais da ABC (AABC)
FERREIRA,MONIQUE S.; GALO,MARIA DE LOURDES B.T..
Considering the importance of monitoring the water quality parameters, remote sensing is a practicable alternative to limnological variables detection, which interacts with electromagnetic radiation, called optically active components (OAC). Among these, the phytoplankton pigment chlorophyll a is the most representative pigment of photosynthetic activity in all classes of algae. In this sense, this work aims to develop a method of spatial inference of chlorophyll a concentration using Artificial Neural Networks (ANN). To achieve this purpose, a multispectral image and fluorometric measurements were used as input data. The multispectral image was processed and the net training and validation dataset were carefully chosen. From this, the neural net...

Photosynthetic response of soybean to twospotted spider mite (Acari: Tetranychydae) injury BABT
Bueno,Adeney de Freitas; Bueno,Regiane Cristina Oliveira de Freitas; Nabity,Paul David; Higley,Leon George; Fernandes,Odair Aparecido.
The twospotted spider mite Tetranychus urticae Koch is a common pest on soybean plants. To clarify plant-arthropod interaction on mite-soybean system, leaf fluorescence, photosynthetic responses to variable carbon dioxide levels, and chlorophyll content were evaluated. Significant photosynthetic rate reduction was observed due to stomatal limitation. Stomatal closure was the major plant physiological response. As a consequence, there was reduction in photosynthetic rates. Surprisingly, plants did not show chlorophyll content reduction associated with photosynthetic impairment. No differences in fluorescence data indicate that T. urticae injury did not impair the function of light harvesting and photoelectron transport. These results showed that T. urticae...

Effect of Hydrostatic Pressure on the Fluorescence of Tryptophan in the Presence of Metal Ions Running title: Effect of pressure on the tryptophan BABT
Bao,Chengman; Tang,Xinhui; Ye,Shuming.
ABSTRACT The effect of pressure on the fluorescence of tryptophan in the presence of metal ions was studied by fluorescence spectrometry. It was found that at 60 MPa, the fluorescence intensity of M/Trp mixtures (M represented metal ions) increased compared to that at atmosphere pressure. The relative fluorescence efficiency of M/Trp mixtures increased with pressure. When the M/Trp ratio was above 10:1, the relative fluorescence efficiency in decreasing order was Cu2+/Trp mixtures, Ni2+/Trp mixtures and Mg2+ (K+)/Trp mixtures. When the ratio was below 10:1, the decreasing order was Cu2+/Trp mixtures and Ni2+ (Mg2+, K+)/Trp mixtures. The relative fluorescence efficiency increased with the concentration of Cu2+ and Ni2+. The variation was relate to the...

Trypan blue exclusion assay by flow cytometry BJMBR
Avelar-Freitas,B.A.; Almeida,V.G.; Pinto,M.C.X.; Mourão,F.A.G.; Massensini,A.R.; Martins-Filho,O.A.; Rocha-Vieira,E.; Brito-Melo,G.E.A..
Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and...

Influence of chromium in Laguncularia racemosa (L). Gaertn f. physiology Braz. J. Plant Physiol.
Rocha,Andréa C.; Canal,Emerson C.; Campostrini,Eliemar; Reis,Fabrício O.; Cuzzuol,Geraldo Rogério F..
Most studies of chromium have been realized in the sediments and in mangrove trees. However, little is known about its effect on growth and metabolism in this vegetation. L. racemosa plants were grown in solution with 20% of ionic strength of Hoagland and Arnon containing 100 mM NaCl and supplemented with 0.00, 0.05 and 0.50 mg L-1 of Cr+3. The growth, the pigments and gas exchange measurements showed no changes in response to doses of chromium. After 30 days the initiation of treatments, plants of L. racemosa had average 0.559 g of total dry weight, 1.34 mg L-1 of total chlorophyll and 7.9 µmol m-2 s-1 of A (CO2 photosynthetic assimilation). The photochemical potential efficiency was affected by the intermediate dose of 0.05 mg L-1 of Cr+3 (Fv/Fm=0.73)...

Morphological analysis and photosynthetic performance of improved papaya genotypes Braz. J. Plant Physiol.
Torres Netto,Alena; Campostrini,Eliemar; Azevedo,Letícia C.; Souza,Marcelo A.; Ramalho,José C.; Chaves,Manuela M..
Brazil is the largest world producer of papaya and the third largest exporter despite of only 1.5 to 2.0% of its production is exported. Such an underexplored exportation potential highlight the necessity for physiological studies on new cultivars and hybrids to verify their agronomic and commercial viability. Two Brazilian states, Bahia and Espirito Santo, are responsible for 80% of national production. Papaya can also be an agricultural alternative to north / northeast of Rio de Janeiro, because the region is close to consumer centers and have similar environmental conditions of the most productive regions. Nevertheless, it is worth to develop cultivars that can express the highest yield potential in this region. The aim of this work was to characterize...
