The MiniFluo-UV is a new glider-compatible optical sensor for measurements of dissolved organic matter (DOM) in natural waters. The working principle, sensor design and challenges faced during the validation phase are reported. The first in situ application of the sensor during three glider deployments in the NW Mediterranean sea (spring, summer, and fall) are also presented. For these campaigns, the two channels of the sensor were adjusted to target Tryptophan-like (excitation/emission wavelengths λEx/λEm: 275/340 nm) and Phenanthrene-like (λEx/λEm: 255/360 nm) fluorescence. These were chosen because they represent fluorophores of interest commonly found in seawater. While Tryptophan (an amino-acid believed to be a by-product of biological activity) is... |

DELVOYE, L., 1992. Endolithic algae in living stony corals: Algal concentrations under influence of depth-dependent light conditions and coral tissue fluorescence in Agaricia agaricites (L) and Meandrina meandrites (L.) (Sclereactinia, Anthozoa). Studies Nat. Hist. Caribbean Region 71, Amsterdam 1992: 24-41. The relation between Scleractinians and their endolithic algae was studied in the depth range of 10 to 35 m in Curaçao. Endolithic algal concentrations are found in the skeleton under the living tissue of stony corals and never in dead parts or in coral rubble. The influences of depth-dependent light conditions and coral tissue fluorescence on endolithic algal concentrations were studied in a non-fluorescent and a fluorescent form of both Agaricia... |

The understanding of soil carbon stabilization processes can be very useful in the development of mitigation techniques for CO2 emissions and global warming. The greater the hydrophobicity of soil organic matter the more stabilized soil organic carbon. Therefore, hydrophobicity can be a sensitive index to characterizethe ‘quality’ of soil organic matter. In this context, the present work aimed to characterize the chemical structures of humic acids collected at three different depths in a hydrophobic Entisol (Neossolo) under loblolly plantation. The results of spectroscopic and chemical analyses (UV-Vis, fluorescence, EPR and X-ray diffractometry) indicated that, as soil depth increased, so did the content of conjugated organic structures, aromatic... |

Abstract: The objective of this work was to evaluate the physiological and production parameters of soybean (Glycine max) cultivars of determinate and indeterminate growth habits, in three levels of soil moisture. Three cultivars of determinate growth habit and three of indeterminate growth, at the beginning of the reproductive stage, were subjected to three levels of soil moisture: control, moisture at field capacity; moderated, 50% of the control level; and severe, 25% of the control level. Leaf gas exchange, xylem-water potential, chlorophyll content, chlorophyll a fluorescence, plant dry matter, and grain yield per plant were evaluated. The cultivars of indeterminate growth habit showed the greatest values for stomatal conductance and CO2 assimilation... |

Abstract Eugenia dysenterica DC is a fruiting species endemic to the Brazilian Cerrado, belonging to the Myrtaceae family and popularly known as Cagaiteira. It has medicinal and antifungal properties, and has an important function in the ecosystem. Nevertheless, there are few studies about the maintenance of this species. The aim of this study was to evaluate the growth, nutrition, quality and physiology of E. dysenterica seedlings grown in fine vermiculite and rice husk-based substrates in the following combinations: 1:0, 3:1, 1:1, and 1:3, in addition to Trimix® commercial substrate and vermiculite only. The physical attributes of substrates (dry and moist densities, available water, remaining water, aeration space and total porosity), seedling emergence... |

We introduce a new, high quality, low cost and versatile LEDs-based handset device that emits high power UV and white light, which can be used interchangeably. It offers power control and has long battery life. Even though it is optimized to detect and collect scorpions under low light conditions, it can also be used with other groups of fluorescent organisms. The device achieved superior performance in field and laboratory trials when compared with a 12 LEDs low power UV flashlight, and a 46 W black fluorescent light lamp, to locate Tityus serrulatus Lutz & Mello, 1922. |

Using herbaceous legumes in agricultural systems yields great quantities of plant residues, allowing changes in soil organic matter quality and content over the years. This study was conducted on an Ultisol, at Seropédica, RJ, Brazil, to evaluate the effects of different perennial herbaceous legumes on soil organic matter quality. A factorial scheme with three replications was used to evaluate the species: forage groundnut cv. BR-14951 (Arachis pintoi), tropical kudzu (Pueraria phaseoloides), and siratro (Macroptilium atropurpureum). After the first cut, each plot was divided into two subplots; plants were cut and left on the soil surface or cut and removed. Soil samples of a closed area covered by spontaneous vegetation (mainly C3 plants) or by Panicum... |

The twospotted spider mite Tetranychus urticae Koch is a common pest on soybean plants. To clarify plant-arthropod interaction on mite-soybean system, leaf fluorescence, photosynthetic responses to variable carbon dioxide levels, and chlorophyll content were evaluated. Significant photosynthetic rate reduction was observed due to stomatal limitation. Stomatal closure was the major plant physiological response. As a consequence, there was reduction in photosynthetic rates. Surprisingly, plants did not show chlorophyll content reduction associated with photosynthetic impairment. No differences in fluorescence data indicate that T. urticae injury did not impair the function of light harvesting and photoelectron transport. These results showed that T. urticae... |

ABSTRACT The effect of pressure on the fluorescence of tryptophan in the presence of metal ions was studied by fluorescence spectrometry. It was found that at 60 MPa, the fluorescence intensity of M/Trp mixtures (M represented metal ions) increased compared to that at atmosphere pressure. The relative fluorescence efficiency of M/Trp mixtures increased with pressure. When the M/Trp ratio was above 10:1, the relative fluorescence efficiency in decreasing order was Cu2+/Trp mixtures, Ni2+/Trp mixtures and Mg2+ (K+)/Trp mixtures. When the ratio was below 10:1, the decreasing order was Cu2+/Trp mixtures and Ni2+ (Mg2+, K+)/Trp mixtures. The relative fluorescence efficiency increased with the concentration of Cu2+ and Ni2+. The variation was relate to the... |

Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and... |

Most studies of chromium have been realized in the sediments and in mangrove trees. However, little is known about its effect on growth and metabolism in this vegetation. L. racemosa plants were grown in solution with 20% of ionic strength of Hoagland and Arnon containing 100 mM NaCl and supplemented with 0.00, 0.05 and 0.50 mg L-1 of Cr+3. The growth, the pigments and gas exchange measurements showed no changes in response to doses of chromium. After 30 days the initiation of treatments, plants of L. racemosa had average 0.559 g of total dry weight, 1.34 mg L-1 of total chlorophyll and 7.9 µmol m-2 s-1 of A (CO2 photosynthetic assimilation). The photochemical potential efficiency was affected by the intermediate dose of 0.05 mg L-1 of Cr+3 (Fv/Fm=0.73)... |

Brazil is the largest world producer of papaya and the third largest exporter despite of only 1.5 to 2.0% of its production is exported. Such an underexplored exportation potential highlight the necessity for physiological studies on new cultivars and hybrids to verify their agronomic and commercial viability. Two Brazilian states, Bahia and Espirito Santo, are responsible for 80% of national production. Papaya can also be an agricultural alternative to north / northeast of Rio de Janeiro, because the region is close to consumer centers and have similar environmental conditions of the most productive regions. Nevertheless, it is worth to develop cultivars that can express the highest yield potential in this region. The aim of this work was to characterize... |

The effects of drought and rehydration on Canavalia ensiformis (L.) D.C. (jack bean) plants were evaluated using the following gas exchange parameters: net carbon assimilation rate (A), stomatal conductance (g s), Ci/Ca ratio and transpiration rate (E); chlorophyll a fluorescence: Fv/Fm and Fv/F0 ratio. The plants were cultivated under greenhouse conditions and after 30 days from the emergence, irrigation was suspended in the plants submitted to drought, to obtain the following predawn leaf water potential (psipd): -0.40 MPa (control), -1.00 MPa (moderate drought) and -2.30 Mpa (severe drought). Afterwards, the gas exchange and fluorescence analysis were initiated , and 24 h after rehydration the same analyses were repeated. The A, E, g s and Ci/Ca values... |
