Summer mortality associated with juveniles of the oyster Crassostrea gigas is the result of a complex interaction between the host, pathogens and environment. Divergent selection based on summer survival have been applied to produce families with resistant and susceptible progeny. Here, we describe the use of suppression subtractive hybridization to generate 150 clones that were differentially regulated between resistant and susceptible F2 progeny. The nucleotide sequence of these 150 clones was determined. Among the 25 unique sequences that matched with the products of known genes, those putatively implicated in energy generation (12%) and immune function (16%), systems suspected to be implicated in summer mortality, were extended and analyzed by real... |

The aim of this study was to determine whether mortality observed during the larval development of reared bluefin tuna (Thunnus thynnus) could be related to improper expression profiles of key genes involved in digestive or antioxidant response capabilities. Tuna larvae were sampled at hatching, 2, 5, 10, 15 and 20 dph (days post hatching) for the relative quantification of transcripts encoded by genes involved in digestive [trypsinogen 1 (TRYP1), alpha-amylase (AMY), aminopeptidase N (ANPEP)] and antioxidant [catalase (CAT)] functions. The levels of expression of ANPEP related to the development and maturation of intestinal function increased from 5 to 20 dph. Furthermore, AMY and TRYP1 genes, which are pancreatic enzymes implicated in carbohydrate and... |

The geographical structure of 15 natural populations of the flat oyster (Ostrea edulis L.) was assessed by single-strand conformation polymorphism (SSCP) of a 313-base-pair (bp) fragment of the mitochondrial 12S-rRNA gene. Fourteen haplotypes were observed, with one being dominant in the Mediterranean samples and another one in the Atlantic populations. The geographically extreme populations sampled in Norway and the Black Sea appeared differentiated by exhibiting the dominance of a third group of haplotypes. The results were compared to available microsatellite data at five loci. The Atlantic/Mediterranean differentiation pattern was qualitatively the same with both types of markers, confirming an isolation-by-distance pattern. The average mitochondrial... |

The history of French oyster culture shows very well how fragile this farming is to stock overexploitation and diseases. The production of flat oyster, Ostrea edulis, fell dramatically after the appearance of two parasitic diseases: arteiliosis and bonamiosis. The curative means against bonamiosis (Bonamia ostreae) are restricted and imply a good knowledge of the parasite's life cycle and of its interaction with its host. Studying the expression of the parasite's and the host's genes during the infection seems essential in providing new adapted measures in order to maintain, if not relaunch the flat oyster culture. In this context, this thesis aims to understand the flat oyster-parasite interactions, and especially, the molecular basis of the resistance to... |

Summer mortality of Crassostrea gigas is the result of a complex interaction between oysters, their environment, and pathogens. A high heritability was estimated for resistance to summer mortality, which provided an opportunity to develop lines of oysters that were resistant (R) or susceptible (S) to summer mortality. Previous genome-wide expression profiling study of R and S oyster gonads highlighted reproduction and antioxidant defense as constitutive pathways that operate differentially between these two lines. Here, we show that signaling in innate immunity also operates differentially between these lines, and we hypothesize that this is at the main determinant of their difference in survival in the field. A reanalysis of our published microarray data... |

The main objective of this project was to develop the tools for the in-depth understanding of mollusc biology and pathology. Such a goal has been reached in other biological systems through the development of molecular biology and cell culture. Indeed, development of a mollusc cell culture system is an essential step to approach many of the problems related to mollusc pathology, in particular to use molecular biology tools to study viral infection of these organisms. No continous cell line is available from bivalves and while cell cultures have been obtained from these organisms they are difficult to develop and labor intensive, survival cultures being maintained only for a few weeks. Our goal has been to study in detail many of the parameters involved in... |

This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively.... |

To investigate the control at the mRNA level of glycogen metabolism in the cupped oyster Crassostrea gigas, we report in the present paper the cloning and characterization of glycogen phosphorylase and synthase cDNAs (Cg-GPH and Cg-GYS, respectively, transcripts of main enzymes for glycogen use and storage), and their first expression profiles depending on oyster tissues and seasons. A strong expression of both genes was observed in the labial palps and the gonad in accordance with specific cells located in both tissues and ability to store glucose. Cg-GPH expression was also found mainly in muscle suggesting ability to use glycogen as readily available glucose to. supply its activity. For seasonal examinations, expression of Cg-GYS and Cg-GPH genes... |

Bacillus thuringiensis harbors genes encoding Cry proteins found in chromosomes or plasmids of different sizes (4-150 Mb). Although the smaller plasmids are more abundant in B. thuringiensis, their specific function is unknown. As for the megaplasmids, their main recognized function is to harbor cry genes, although the sequencing of some of these plasmids indicates the occurrence of other important genes. This work used a new protocol for practical and rapid extraction of plasmid DNA in order to characterize the plasmid patterns of Brazilian strains belonging to Embrapa Milho e Sorgo research center B. thuringiensis bank. We tried to further assess the relationship of plasmid patterns with strains belonging to the same serovars and strains causing 100% and... |
