Rooting stimulation in muscadine grape cuttings Scientia Agricola
Castro,P.R.C.; Melotto,E.; Soares,F.C.; Passos,I.R.S.; Pommer,C.V..
The muscadine grape (Vitis rotundifolia Michx.) is native to Florida and has been cultivated for many years in U.S.A. It is harvested as single berries instead of in bunches and has small leaves. The fruit skin is thicker than those of bunch-type grapes. One reason for its popularity is that the muscadine is seldom seriously affected by diseases or insects. The muscadine grape is not readily propagated by hardwood cuttings, therefore the present work was carried out to develop a method for commercial propagation of its cuttings, using treatments with growth regulators (auxins) and low temperature. The results showed that the rooting of both basal and middle shoots of muscadine grape is better than the rooting of terminal sections of the shoots. Cuttings...

In vitro seed germination and seedling development of Annona crassiflora Mart. Scientia Agricola
Ribeiro,Márcia de Nazaré Oliveira; Pasqual,Moacir; Villa,Fabíola; Pio,Leila Aparecida Salles; Hilhorst,Henk William Maria.
Annona crassiflora Mart known as 'araticum', 'marolo' or 'field araticum' is a typical fruit from the Cerrado biome of Brazil with socio-economic and medicinal importance. Normally, Annona crassiflora is propagated through seeds. However, due to a deep dormancy that the seeds display at dispersion and the difficulty to obtain uniform plants in a short time period, micropropagation may be a feasible alternative. Concentrations of gibberellic acid (GA3) and naphthalene-acetic acid (NAA) and their interactive effects on in vitro seed germination and seedling development of Annona crassiflora were studied. Mature fruits of Annona crassiflora were depulped and the seeds washed in clear water and dried at room temperature. Seed coat was removed and the seeds...

Abscisic acid and ethephon treatments applied to ‘Verdejo’ white grapes affect the quality of wine in different ways Scientia Agricola
González,Raquel; González,María-Rosa; Martín,Pedro.
ABSTRACT: Exogenous ethephon and abscisic acid (ABA) treatments are used worldwide to improve the coloring of red table grapes, but there is little information about their impact on the quality of white grapes and wines. The aim of this work was to evaluate the effects of exogenous ethephon and ABA applications at veraison on the composition of ‘Verdejo’ white grapes, and on the quality of their wines. To attain this objective, a field trial was carried out in a ‘Verdejo’/110 Richter vineyard located in north-central Spain. Two levels of ethephon (0 and 1500 mg L−1) and ABA (0 and 800 mg L−1) were sprayed on clusters at veraison, in a factorial design for three consecutive seasons. Ethephon and ABA had additive effects, decreasing titratable acidity and...

Griebeler,Ana Gláucia; Consatti,Guilherme; Freitas,Elisete Maria de; Sperotto,Raul Antonio.
In the state of Rio Grande do Sul, Brazil, yerba mate (Ilex paraguariensis A. St.-Hil.) is an economically and culturally important species. Because of the difficulty in obtaining plantlets from seeds, micropropagation has emerged as an alternative that could increase production. This work aimed to evaluate the effect that treatment with 6-benzyladenine (BA), at concentrations of 2.2, 8.9, and 20 µM and associated with naphthaleneacetic acid (NAA, 0.5 µM), has on the in vitro growth and leaf emission of I. paraguariensis nodal segments cultivated in 0.5 X Murashige and Skoog (MS) medium or 0.5 X Woody Plant Medium (WPM). The most efficient treatment was MS medium with 8.9 µM BA+NAA, in which I. paraguariensis presented greater shoot elongation and higher...

Callus induction and plant regeneration of tomato (Lycopersicon esculentum cv. IPA 5) via anther culture Ciência Rural
Brasileiro,Ana Christina Rabello; Willadino,Lilia; Carvalheira,Gianna Griz; Guerra,Marcelo.
Different growth regulators combinations were tested on the production of anther callus in tomato cultivar IPA 5. Calli were induced on media supplemented with 1.0mgL-1 gibberellic acid (GA3), 0.05mgL-1 alpha-naphthaleneacetic acid (NAA) plus 0.1mgL-1 6-benzylaminopurine (BAP), or with 1.0mgL-1 BAP plus 1.0mgL-1 NAA. The medium containing 1.0mgL-1 BAP and 1.0mgL-1 NAA produced the highest calli frequency, and promoted plant regeneration by indirect organogenesis, when calli were transferred to 0.01mgL-1 BAP and 0.001mgL-1 NAA. Plants regenerated presented tetraploid cells and rare diploid cells. These tetraploid plants could be used as source for further obtainment of trisomic lines, for the purpose of genic localization studies and protein compounds...

Adaptation of in vitro regeneration protocol for Brazilian wheat genotypes Ciência Rural
Roesler,Eduardo André; Manfroi,Ernandes; Morás,Andréa; Didoné,Dielli Aparecida; Grando,Magali Ferrari; Lau,Elene Yamazaki.
ABSTRACT: The availability of an efficient protocol for in vitro regeneration is imperative for genetic transformation. Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. Furthermore, combinations of 2,4-D and picloram in the callus induction medium were tested in order to improve regeneration efficiency. Genotypes with higher regeneration efficiency were BR18-Terena and PF020037, yielding 0.42 and 1.13 shoots per explant...

Effects of cytokinins on in vitro mineral accumulation and bud development in Annona glabra L. Ciência e Agrotecnologia
Oliveira,Lenaldo Muniz de; Paiva,Renato; Santana,José Raniere Ferreira de; Pereira,Flávia Dionísio; Nogueira,Raírys Cravo; Silva,Luciano Coutinho.
Annona glabra is a tropical species that has significant agronomic potential in terms of furnishing fruits for in natura consumption and for the production of phyto-pharmaceuticals. In vitro cultivation has been considered the most promising form of propagation for this species, although large scale utilization of this technique is currently limited by high rates of leaf abscission, reduced rates of explant multiplication and slow bud growth. The present work evaluated the effects of different cytokinins on mineral accumulation in shoots of A. glabra cultivated in vitro, and their effects on growth and survival of these plants. Buds of A. glabra were cultivated in Wood Plant Medium (WPM) in the presence of 6-benzilaminopurine (BAP), thidiazuron (TDZ),...

Tissue culture of Cecropia glaziovii Sneth (urticaceae): vegetative micropropagation and plant regeneration from callus Ciência e Agrotecnologia
Alves,Marcos Nopper.
Cecropia glaziovii is a tree with used in Brazilian popular medicine. Methods allowing the clonal propagation of this species are of great interest for superior genotype multiplication and perpetuation. For this reason, we examined the effect of different culture media and different types of explants on adventitious shoot regeneration from callus and buds of C. glaziovii. Leaves, petioles and stipules obtained from aseptically grown seedlings or from pre-sterilized plants were used to initiate cultures. Adventitious shoot regeneration was achieved when apical and axillary buds were inoculated on gelled Murashige & Skoog (MS) medium supplemented with 6-benzylaminopurine alone (BAP) (1.0, 5.0 or 10.0 mg L-1) or combined with -naphthalene acetic acid...
