Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer

Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II. Phenotyping and ribotyping with strains isolated during the study showed that only TNNIII7 and TNMB5 are similar to the pathogenic strain TNEMF6....

Genetic consequences of production of Pacific oyster larval in hatchery: drift and selective pressures related to rearing practices. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors were examined: the effects of discarding the smallest larvae (i.e. culling) and temperature effects. A mixed-family approach was used in order to infer the genetic composition of the larval population. The results show that high polymorphic microsatellite-based family assignment is a powerful tool for the study of bivalve larvae genetics. Artificial production from natural spawners could be a way of getting big quantities of spat of Pecten maximus, ln the method described, larval phase (until a size of 250 microns) occurs in a hatchery, post-Iarval phase (from 250 µ until 3 millimeters) in a nursery, and first growth period directly into the sea (from 2 mm to a size of 25-30 mm), The fifst results obtained, production of several thousand of individuals good enough for reseeding, seem to be transposable to a larger scale.

A comparative study of the reproductive cycle of Ruditapes decussatus under natural (South of Tunisia) and controlled conditions (hatchery). The aim of this study was to describe and compare the gametogenic activity of Ruditapes decussatus in situ and in hatchery. Criteria of maturity were characterized and the positive role of conditioning in the success of artificial reproduction was underlined. Effect of the probiotic strain Phaeobacter gallaeciensis after bacterial challenge on the complete larval development of Pecten maximus ArchiMer

The aim of this project was to evaluate the impact of probiotic (Phaeobacter gallaeciensis, X34 strain) treatment on the complete development (from veliger to metamorphosis) of Pecten maximus larvae in the context of a bacterial challenge and in conditions more representative of hatchery practices. To that effect, the present study was divided into two main steps. Mortalités massives en élevage larvaire de Lates Calcarifer ( Mise en évidence d'un virus neurotrope ) ArchiMer

Since 1986 high mortality of hatchery-reared sea bass, Lates Calcarifer, larvae appeared in Ifremer's Pacifie Oceanological Center (Tahiti). Light and electron microscopy show vacuolation and cells with cytoplasmic inclusion bodies in the brain, the cord and the eye relina of affected larvae. Numerous non-enveloped particles identified as viral units, icosahedral in morphology and measuring about 25 nm in diameter were found in the cytoplasm of the degenerating cells. In affected larvae, these particles were isolated by density grandient centrifugations. In vitro tests on cell lines and in vivo test on fish are performed with samples of diseased larvae. And immunisation of mice are made with purified immunoglobulins of Lates calcarifer.

Data are presented on the following subjects: environmental factors that sustain gametogenic activity and the time of year this activity occurs; biochemical changes that are associated with gametogenesis; the consistency of egg quality; and development of a simple test to predict egg quality.

Establishment of microbiota in larval culture of Pacific oyster, Crassostrea gigas ArchiMer

This study has two main objectives: (1) to implement a recycling aquaculture system (RS) for the larvae of the oyster Crassostrea gigas, and (2) to characterise the bacterial communities established in different compartments of this system. An RS with 25% fresh seawater addition per hour and another with no addition (0%) were compared with a flow-through system (FT). Larval survival was equivalent in RS and FT, but growth rate was 17% slower in RS than in FT. The physical chemical parameters remained stable, except for pH that decreased to 7.75 and salinity that increased to 37.5‰ in the RS 0%. Different pH levels of water in the incubation and larviculture of 'curimbatá' (Prochilodus lineatus) were tested to evaluate the effect of this variable on the survival and development of eggs and larvae. During incubation, pH of 5.0, 6.0, 7.0, 8.0, and 8.5 was tested. To observe the effects of pH on larvae with different hatchery histories, the following experimental design was utilised (pH incubation - pH larviculture): L 6-6; L 6-7; L 7-6; L 7-7; L 7-8; L 8-7; L 8-8; L 8.5-7, and L 8.5-8.5. Optimization of heterologous microsatellites in Piracanjuba PAB

Abstract: The objective of this work was to evaluate 41 microsatellite markers for heterologous amplifications in piracanjuba (Brycon orbignyanus). Some markers were tested for the first time. Loci were optimized for PCR conditions and applied to a sample of 49 individuals. Thirty-one loci resulted in PCR product formation, whereas ten loci yielded intelligible polymorphic patterns in the evaluated sample and can be used for amplifications in this species. From the evaluated markers, four loci (BoM1, BoM13, Bh6, and Bh16) are valid to be applied in the study of piracanjuba.

A completely randomized study was used with five treatments (0L:24D, 6L:18D, 12L:12D, 18L:6D, 24L:0D) and four replicates, with the aquaria as the experimental unit. This study evaluated the effects of hatching in low performance broiler chicks before industrial production on survival rate and productive performance. Twelve houses of 15,000 birds each, day-old chicks (Ross 308) were assigned to randomized complete block design (RCBD) in a 2×2 factorial arrangement with two factors.
