Comparative embryotoxicity and genotoxicity of the herbicide diuron and its metabolites in early life stages of Crassostrea gigas: implication of reactive oxygen species production ArchiMer
Behrens, Daphne; Rouxel, Julien; Burgeot, Thierry; Akcha, Farida.
Herbicides are one of the major classes of pollutants contaminating coastal waters over the world. Among them, diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a phenylurea herbicide frequently detected in oyster-producing area, known to be toxic for this important exploited non-target species. With the aim to investigate the mechanisms by which diuron displays its toxicity in oyster, the implication of both biotransformation and oxygen reactive species (ROS) production was studied considering embryotoxicity and genotoxicity as endpoints. Comparative embryotoxicity and genotoxicity of diuron and its main metabolites (DCPMU, DCPU and 3,4-DCA) were thus studied on oyster larvae by the embryo-larval bioassay on D larvae and the comet assay on trochophore...

Study of genetic damage in the japanese oyster induced by an environmentally-relevant exposure to diuron: evidence of vertical transmission of dna damage ArchiMer
Barranger, Audrey; Akcha, Farida; Rouxel, Julien; Brizard, Raphael; Maurouard, Elise; Pallud, Marie; Menard, Dominique; Tapie, N.; Budzinski, Helene; Burgeot, Thierry; Benabdelmouna, Abdellah.
Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Commercially-raised bivalves are particularly exposed to pollutants, among them pesticides, as shellfish farming zones are subject to considerable pressure from agricultural activities on the mainland. The aims of this study were to determine (1) the genotoxic effects of diuron exposure on oyster genitors and (2) the possible transmission of damaged DNA to offspring and its repercussions on oyster fitness. To investigate these points, oysters were exposed to concentrations of diuron close to those detected in the Marennes-Oleron Basin (two 7-day exposure pulses at 0.4 and 0.6 μg...

Etude des apports en herbicides et en nutriments par la Charente : Modélisation de la dispersion de l'atrazine dans le bassin de Marennes-Oléron ArchiMer
Munaron, Dominique.
Our study aims at making progress the estimate of herbicide and nutrient inputs to the Marennes-Oleron bay which could influence summer oysters mortalities, and to bring by modeling a better understanding of the dynamics of these compounds in the shellfish-farming area. The development of a strategy adapted to the Charente river and its estuary reveals that agricultural activity on the catchment area is omnipresent, as well on the level of the nitrogen and herbicides contributions. It is thus necessary to specify Charente fluxes, which is the main tributary of the Marennes-Oleron bay, in order to better characterize its impact on shellfishfarming activity. Use of a dispersion model will constitute an invaluable help in the risk assessment with respect to...

Barranger, Audrey.
The aims of this PhD were to determine (1) the effects of the herbicide diuron on the genome of the oyster, Crassostrea gigas, (2) the possible transmission to the offspring of damaged DNA and (3) its consequences on oyster physiology. The genotoxicity of diuron and the vertical transmission of DNA damage were highlighted after parental exposure to environmental concentrations of diuron during the gametogenesis. Primary DNA lesions were detected in both somatic cells and germ cells of exposed genitors. Detection of the oxidized base 8-oxodGuo in gonads shows that exposure to diuron led to an oxidative stress which likely explains the observed DNA strand breaks in spermatozoa. The transmission of these DNA damage was confirmed by fluorescent in situ...

Environmental epigenetic is an emerging field that studies the cause-effect relationship between environmental factors and heritable trait via an alteration in epigenetic marks. This field has received much attentions since the impact of environmental factors on different epigenetic marks have been shown to be associated with a broad range of phenotypic disorders in natural ecosystems. Chemical pollutants have been shown to affect immediate epigenetic information carriers of several aquatic species but the heritability of the chromatin marks and the consequences for long term adaptation remain open questions. In this work, we investigated the impact of the diuron herbicide on the DNA methylation pattern of spat from exposed Crassotrea gigas genitors. This...

Adsorption of glyphosate in chilean soils and its relationship with unoccupied phosphate binding sites PAB
Kogan,Marcelo; Metz,Alejandra; Ortega,Rodrigo.
The objective of this work was to investigate glyphosate adsorption by soils and its relationship with unoccupied binding sites for phosphate adsorption. Soil samples of three Chilean soils series - Valdivia (Andisol), Clarillo (Inceptisol) and Chicureo (Vertisol) - were incubated with different herbicide concentrations. Glyphosate remaining in solution was determined by adjusting a HPLC method with a UV detector. Experimental maximum adsorption capacity were 15,000, 14,300 and 4,700 mg g¹ for Valdivia, Clarillo, and Chicureo soils, respectively. Linear, Freundlich, and Langmuir models were used to describe glyphosate adsorption. Isotherms describing glyphosate adsorption differed among soils. Maximum adjusted adsorption capacity with the Langmuir model...

Resistance to ACCase inhibitors in Eleusine indica from Brazil involves a target site mutation Planta Daninha
Osuna,M.D.; Goulart,I.C.G.R.; Vidal,R.A.; Kalsing,A.; Ruiz Santaella,J.P.; De Prado,R..
Eleusine indica (goosegrass) is a diploid grass weed which has developed resistance to ACCase inhibitors during the last ten years due to the intensive and frequent use of sethoxydim to control grass weeds in soybean crops in Brazil. Plant dose-response assays confirmed the resistant behaviour of one biotype obtaining high resistance factor values: 143 (fenoxaprop), 126 (haloxyfop), 84 (sethoxydim) to 58 (fluazifop). ACCase in vitro assays indicated a target site resistance as the main cause of reduced susceptibility to ACCase inhibitors. PCR-generated fragments of the ACCase CT domain of the resistant and sensitive reference biotype were sequenced and compared. A point mutation was detected within the triplet of aspartate at the amino acid position 2078...

Estimation of Sulfentrazone Leaching in Isolated Application and in Mixture with Glyphosate Planta Daninha
LANGARO,A.C.; BARROS,J.P.A.; SOUZA,M.F.; SILVA,E.M.G.; SILVA,A.A..
ABSTRACT: As an alternative to control of resistant biotypes, farmers have resorted to the use of herbicides that have residual soil activity and good pre-emergence action. One of the most used mixtures in Brazil is that of sulfentrazone + glyphosate. When a herbicide has a pre-emergence action, small leaching of its molecules is necessary for its superficial incorporation into the soil, where most of the weed seeds are found. However, if excessive leaching occurs, the herbicide may have reduced efficiency in controlling weed, increasing the risks of groundwater contamination. In this study, sulfentrazone leaching was evaluated in columns with soil, in isolated applications and in mixture with formulations of glyphosate (Roundup Ready®, Roundup Ultra® and...

Morpho-Architectural Traits that Allow the Regeneration of Eustachys retusa (poaceae) in Systems with Intensive Glyphosate Application Planta Daninha
PANIGO,E.S.; ALESSO,C.A.; DELLAFERRERA,I.M.; OLIVELLA,J.; PERRETA,M.G..
ABSTRACT Eustachys retusa has low sensitivity to glyphosate at the reproductive stage. The purpose of this study was to analyze the effect of glyphosate on adult E. retusa plants in order to identify the traits that make possible their regeneration in no-tillage systems. For this purpose, reproductive-stage specimens from wild populations were treated with glyphosate at two different rates (480 and 1,200 g a.i. ha-1) including a non-treated control. Results demonstrated that glyphosate could control E. retusa plants at reproductive stage at 1,200 g a.i. ha-1 dose. However, a certain proportion of plants can survive despite having full chlorosis as a consequence of basal bud activation (bud bank) and the presence of reserves in the rhizome. These...

Integration of sunflower (Helianthus annuus) residues with a pre-plant herbicide enhances weed suppression in broad bean (Vicia faba) Planta Daninha
Alsaadawi,I.S; Khaliq,A; Al-Temimi,A.A; Matloob,A.
Field trial was conducted with the aim of utilizing allelopathic crop residues to reduce the use of synthetic herbicides in broad bean (Vicia faba) fields. Sunflower residue at 600 and 1,400 g m-2 and Treflan (trifluralin) at 50, 75 and 100% of recommended dose were incorporated into the soil alone or in combination with each other. Untreated plots were maintained as a control. Herbicide application in plots amended with sunflower residue had the least total weed count and biomass, which was even better than herbicide used alone. Integration of recommended dose of Treflan with sunflower residue at 1,400 g m-² produced maximum (987.5 g m-2) aboveground biomass of broad bean, which was 74 and 36% higher than control and recommended herbicide dose applied...

Dynamics of Clomazone Formulations Under Different Applications Conditions Planta Daninha
TROPALDI,L.; BRITO,I.P.F.S.; DIAS,R.C.; TRINDADE,M.L.B.; CARBONARI,C.A.; VELINI,E.D..
ABSTRACT: Maintenance of straw on the soil surface in sugarcane production areas can influence weed occurrence and herbicide dynamics. After application, considerable losses of clomazone can occur as a result of its physicochemical characteristics. For this reason, novel formulations have been developed. In addition to the conventional formulation, microencapsulated formulations are currently available. Thus, the aim of this study was to observe and compare the effect of clomazone formulations under different application conditions. For this purpose, the experiment was carried with 12 treatments in a completely randomized design, with four replications, using clomazone formulations (1,200 g a.i. ha-1), on soils with different moisture levels, in the...
