The Digestive Function of Pseudoplatystoma punctifer Early Juveniles Is Differentially Modulated by Dietary Protein, Lipid and Carbohydrate Content and Their Ratios ArchiMer
Castro-ruiz, Diana; Andree, Karl B.; Solovyev, Mikhail M.; Fernández-méndez, Christian; García-dávila, Carmen; Cahu, Chantal; Gisbert, Enric; Darias, Maria J..
Pseudoplatystoma punctifer is an Amazonian catfish highly appreciated for its high flesh quality, size, and commercial value. Its aquaculture is pursued to satisfy the demands of an increasing population in the region. However, knowledge of the nutritional needs during the early life stages is necessary for improving growth and reducing the incidence of cannibalism, factors that limit the success of its commercial farming. This study aimed at evaluating the influence of four diets containing different protein and lipid levels (30:15, 30:10, 45:15, or 45:10 in %) in the digestive physiology and performance of early juveniles. The results showed that the dietary protein:lipid as well as carbohydrate levels and ratios influenced differently the whole-body...

Evaluating the histological-based condition of wild collected larval fish: A synthetic approach applied to common sole (Solea solea) ArchiMer
Di Pane, Julien; Gendrot, F.; Giraldo, Carolina; Marchal, Paul; Koubbi, Philippe; Loots, Christophe.
In the present study, the histological-based condition of common sole (Solea solea) larvae in the English Channel was evaluated to assess the relevance of experimentally-calibrated indices for wild collected specimens. Based on topographical observation of foregut, midgut, hindgut, liver and pancreas, each of the 202 larvae analysed was attributed one of the six grades of a condition index that was developed for sole in the nineties. In parallel, a synthetic table of 20 criteria historically used as condition indicators was created from an extensive review of the literature. These criteria were related to tissular integrity, cells height, nuclei, presence of prey and vacuolisation of the same five target organs. For each larva, each criterion was scored on...

Effects of an environmentally relevant concentration of diuron on oyster genitors during gametogenesis: responses of early molecular and cellular markers and physiological impacts ArchiMer
Akcha, Farida; Barranger, Audrey; Bachere, Evelyne; Berthelin, C. Heude; Piquemal, D.; Alonso, P.; Sallan, R. Rondon; Dimastrogiovanni, G.; Porte, C.; Menard, Dominique; Szczybelski, A.; Benabdelmouna, Abdellah; Auffret, M.; Rouxel, Julien; Burgeot, Thierry.
Genitors of the Pacific oyster Crassostrea gigas were submitted during gametogenesis to a short pulse exposure to the herbicide diuron at a realistic environmental concentration. Histological analysis showed no effect of diuron on gametogenesis course, sex ratio and reproductive effort. A non-significant increase in testosterone and progesterone levels was observed in genitors exposed to the herbicide. At cell level, diuron exposure was shown to modulate the phagocytic activity of circulating hemocytes. The results of a transcriptional analysis showed that diuron affected the expression of genes belonging to functions known to play a major role during oyster gametogenesis such as gene transcription regulation, DNA replication and repair, DNA methylation...

Histogenese de l'organe Y (glande de mue) chez l'embryon de la crevette Palaemon serratus (Pennant) (crustace decapode Natantia) ArchiMer
Leroux, A.
The Y organ appears during the embryonic development of the prawn (P. serratus ) at a stage approximately C, according to Richard's definition (1974). 3 weeks separate this stage from hatching at a temperature of 20 degree C. The organ formation is realized over 48 hours and is marked by an histological modification and a small mitotic crisis in the epidermal tissue from which it differentiates. Its metameric localization is mainly maxillular.

Effects of bioactive extracellular compounds and paralytic shellfish toxins produced by Alexandrium minutum on growth and behaviour of juvenile great scallops Pecten maximus ArchiMer
Borcier, Elodie; Morvezen, Romain; Boudry, Pierre; Miner, Philippe; Charrier, Gregory; Laroche, Jean; Hegaret, Helene.
Dinoflagellates of the genus Alexandrium are a major cause of harmful algal blooms (HABs) that have increasingly disrupted coastal ecosystems for the last several decades. Microalgae from the genus Alexandrium are known to produce paralytic shellfish toxins (PST) but also bioactive extracellular compounds (BEC) that can display cytotoxic, allelopathic, ichtyotoxic or haemolytic effects upon marine organisms. The objective of this experimental study was to assess the effects of PST and BEC produced by A. minutum upon juvenile great scallops Pecten maximus. Scallops were exposed for one week to two different strains of A. minutum, the first producing both PST and BEC and the second producing only BEC. Escape response to starfish, daily shell growth,...

Trophic relationships gonad-intestinal loop in Pecten maximus . ArchiMer
Lepennec, M; Dorange, G; Beninger, P; Donval, A; Widowati, I.
Histological, histochemical and ultrastructural studies were performed on the region of the gonad traversed by the intestine in Pecten maximus . The results indicate a digestive and absorptive function of intestine epithelial cells and a transfer of metabolites to the surrounding gonad tissue. Several possible mechanisms of translocation are presented, including diffusion into the haemolymph and transfer via vacuolated cells of the gonad interstitial connective tissue.

First report of a Mikrocytos-like parasite in European oysters Ostrea edulis from Canada after transport and quarantine in France ArchiMer
Gagne, N; Cochennec, Nathalie; Stephenson, M; Mcgladdery, S; Meyer, G; Bower, S.
As part of a disease resistance experiment, 112 apparently healthy European flat oysters Ostrea edulis L. were exported from Canada (Nova Scotia) into France to test their susceptibility to Bonamia ostreae infection. Twelve oysters died in transit and 17 others died within 2 wk of laboratory quarantine acclimation. All oysters were examined histologically, and the 17 that died during quarantine were assayed for microcells (Bonamia sp. and Mikrocytos mackini) using molecular techniques. A microcell parasite was detected in the connective tissue of 5 of the 112 oysters. Morphological appearance, tissue affinity and molecular characterization through PCR, in situ hybridization (ISH), fluorescence in situ hybridization (FISH) and sequencing revealed a protist...

A histological study on the development of the digestive system of Pseudosciaena crocea larvae and juveniles ArchiMer
Mai, Kangsen; Yu, Hairui; Ma, Hongming; Duan, Qingyuan; Gisbert, Enric; Zambonino, Jose-luis; Cahu, Chantal.
The ontogenetic development of the gut and accessory organs in large yellow croaker Pseudosciaena crocea was investigated using light microscopy from hatching up to the juvenile stage (40 days post hatch, dph). At 3 dph (mean +/- S.D., 4.1 +/- 0.1 mm total length, L-T), coinciding with the buccopharynx opening, larvae started to feed exogenously, and the gut consisted of a well-developed buccopharynx, a partially-differentiated oesophagus and an intestine divided in three regions (anterior intestine, intermediate intestine and rectum). Yolk reserves were not completely depleted at the onset of exogenous feeding, and a period of mixed nutrition was observed up to 6 dph (4.3 +/- 0.1 mm L-T) when yolk was definitively exhausted. Important morphological...

Histological study of a cellular reaction in Ruditpaes decussatus infected by a protozoan ArchiMer
Chagot, Dominique; Comps, Michel; Boulo, Viviane; Ruano, Francisco; Grizel, Henri.
An infection caused by a Perkinsus -like parasite has been observed in Ruditapes decussatus from Portugal (Comps and Chagot, 1987). Following a pathological study carried out on clams affected by mortalities in the Algarve area, this parasite was found again, inducing a reaction process in the host. Histological examination reveals the parasite in the connective tissue of different organs. The adjoining tissues, particularly the epithelia, are not affected.

Classical and computed tomographic anatomical analyses in a not-so-cryptic Alviniconcha species complex from hydrothermal vents in the SW Pacific ArchiMer
Laming, Sven; Hourdez, Stéphane; Cambon-bonavita, Marie-anne; Pradillon, Florence.
The chemosymbiotic gastropod Alviniconcha (Provannidae), first described in 1988, is one of the most emblematic hydrothermal-vent taxa described from the Central Indian Ridge and the Southwest (SW) Pacific. Symbiotic bacteria found in the gill of Alviniconcha are thought to be their principal source of nutrition. In the SW Pacific, species distributions for A. kojimai, A. boucheti – and to a lesser extent A. strummeri – overlap. While Alviniconcha species do not appear to truly co-exist in these highly energetic but spatially limited habitats, certain species regularly co-occur within a single vent field and in rare instances, the same edifice. Past research suggests that SW-Pacific Alviniconcha species might aggregate around fluids with distinct...

Influence of lupin and rapeseed meals on the integrity of digestive tract and organs in gilthead seabream (Sparus aurata L.) and goldfish (Carassius auratus L.) juveniles ArchiMer
Omnes, Marie-helene; Silva, Francisco; Moriceau, Jacques; Aguirre, P.; Kaushik, Sadasivam; Gatesoupe, F. J..
Two rearing trials were conducted to evaluate the effects of dietary lupin (LM) and rapeseed (RM) meals in gilthead seabream (Sparus aurata L.) and goldfish (Carassius auratus L.) juveniles. Each plant meal was incorporated at the rate of 200 g kg−1 in two distinct diets, which were compared with a fishmeal-based diet as control. After 1 month, the two plant diets did not influence the whole body growth, but the digestive systems were affected. The splenosomatic index was reduced with two plant meals in goldfish and with RM in seabream. The hepatosomatic index was only reduced in LM-fed seabream. Cellular characteristics were also affected. The largest liver cells were observed in RM-fed goldfish suggesting changes in metabolic function. The LM and RM...

Evaluation of the gonad volume throughout magnetic resonance imaging in Crassostrea gigas and comparison with the histological method ArchiMer
Flahauw, Emilie; Davenel, Armel; Quellec, Stéphane; Quillien, Virgile; Degremont, Lionel; Lapegue, Sylvie; Hatt, Philippe-jacques.
Biological parameters of the cupped oyster Crassostrea gigas such as growth, reproductive effort (duration of the gametogenesis, date of spawning, and fecundity) differ widely among oysters in the same location. Furthermore, most of the traditional methods, like histological observations, could lead to large variability on the observations and the measurements ofthese traits, and large uncertainties on their changes over time. These can be drastically reduced by using Magnetic Resonance Imaging (MRI) which is a non invasive method allowing to measure the same individual during its development. Forty oysters were assessed using MRI for the volume of whole soft body, the intra valves space, the adductor muscle, and the gonad, which also include the digestive...

Azaspiracid accumulation, detoxification and biotransformation in blue mussels (Mytilus edulis) experimentally fed Azadinium spinosum ArchiMer
Jauffrais, Thierry; Marcaillou-le Baut, Claire; Herrenknecht, Christine; Truquet, Philippe; Sechet, Veronique; Nicolau, Elodie; Tillmann, Urban; Hess, Philipp.
Azadinium spinosum (Elbrächter and Tillmann), a small marine dinoflagellate, has been recently described as a de novo producer of azaspiracid-1 and -2 (AZA1 and -2) diarrhoeic toxins. A culture of A. spinosum was established in our laboratory and optimised for pilot-scale production of this organism, to evaluate and understand AZA1 and -2 accumulation and biotransformation in blue mussels (Mytilus edulis) fed with A. spinosum. Adult mussels were continuously exposed to A. spinosum over 1 week in 160 L cylindrical conical tanks. Three different diets were tested for contamination: 5000, 10 000 cells mL−1 of A. spinosum and a mixture of 5000 cells mL−1 of A. spinosum with 5000 cells mL−1 of Isochrysis aff. galbana (T-Iso, CCAP 927/14). Comparison of dietary phospholipids and neutral lipids: effects on gut, liver and pancreas histology in Atlantic cod (Gadus morha L.) larvae ArchiMer
Wold, P. -a.; Hoehne-reitan, K.; Cahu, Chantal; Zambonino, Jose-luis; Rainuzzo, J.; Kjorsvik, E..
The aim of the present study was to compare effects of dietary n-3 highly unsaturated fatty acids (HUFA) being incorporated in the phospholipid (PL) or in the neutral lipid (NL) fraction of the larval feed, on larval growth and histology of digestive organs in Atlantic cod (Gadus morhua L.) larvae. Three isoproteic and isolipidic diets, labelled according to the percentage of n-3 docosahexaenoic acid and eicosapentaenoic acid contained in NL1 or in PL1 and PL3 of the diets, were fed to cod larvae from 17 days post hatching (dph) to 45 dph. In the liver, hepatocytes and their nuclei were smaller in NL1 larvae compared with the PL larvae; the mitochondrial membrane structures were less dense and the amount of lipids observed in the liver was significantly...

Reproductive dynamics of swordfish (Xiphias gladius) in the southwestern Indian Ocean (Reunion Island). Part 1: oocyte development, sexual maturity and spawning ArchiMer
Poisson, Francois; Fauvel, Christian.
The reproductive dynamics of swordfish (Xiphias gladius) was investigated from catches of the Reunion Island-based longline swordfish fishery between 19-25 degrees S and 48-54 degrees E. Thus, 1727 swordfish (size range 75-289 cm, lower jaw-to-fork length, LJFL) were sampled on board commercial fishing vessels during the period of May 1998 to January 2001. Reproductive activity was assessed using macroscopic gonad characteristics, trends of gonadal indexes for both sexes, oocyte size-frequency distributions and microscopic investigation of oocyte development stages. Size at first maturity (L-50) for female and male swordfish was estimated in Indian Ocean for the first time. L50 was estimated at 170 cm and 120 cm (LJFL) for females and males, respectively....

Histology, cytogenetics and cytofluorymetry in diagnosis of neoplasia in Macoma balthica (Bivalvia, L.) from the southern Baltic Sea ArchiMer
Katarzyna, S; Renault, Tristan; Wolowicz, M.
This study presents results of histopathologic, cytogenetics and flow cytometry analyses performed on Macoma balthica collected from the Gulf of Gdansk (Baltic Sea) in 2003 in order to compare the techniques for diagnosis of neoplasia. The proportion of affected clams gave a crude prevalence of 15.7 %. The four stages of the disease defined by histology and three stages of neoplasia defined by flow cytometry were reported. Stage I defined by flow cytometry corresponded to stages I and II defined by histology. Chromosome analysis did not lead to a staging of neoplastic progression. Both cytogenetics and flow cytometry indicated a difference in the DNA content of non-neoplastic and neoplastic cells. Cytogenetics examination marked that the range of...
