Immobilization of Beauveria bassiana Lipase on Silica Gel by Physical Adsorption BABT

Extracellular lipase from Beauveria bassianastrain CG481 was immobilized by using thirteen different immobilization protocols. Silica gel was chosen as the most suitable adsorbent with 94.8% of activity yield. The adsorption on silica gel did not change the optimum pH (8.5) and temperature (45ºC) values of the free lipase (FL) for lipolytic activity, and it showed higher activities in extreme conditions (pH 9.0 to 10.5, 60ºC). The lipase immobilized on silica gel (ILS) showed enhanced stability at pH 7.0 after 120 h incubation (69.0%) when compared to FL (33.3%). Glutaraldehyde has been used extensively as a crosslinking agent as well as for functionalization of matrices to immobilize β-galactosidase. Effect of raffinose and ultrasound pulses on invertase release by free and immobilized Saccharomyces cerevisiae in loofa (Luffa cylindrica) sponge BABT

This study investigated the effect of raffinose and ultrasound pulses on invertase release from free S. cerevisiae and S. cerevisiae immobilized in Luffa cylindrica. The free cell culture was submitted to 2% raffinose pulse and irradiated for 2 minutes at 0.12 and 0.46 h-1 dilution rates. The immobilized cell culture was submitted to raffinose pulse and irradiated for 1, 2 and 4 minutes, at 0.10 h-1 dilution rate. In immobilized cells, the raffinose pulse increased the invertase activity from 5.38 to 7.27 U/mg. Ultrasound application in free cell culture at the 0.12 h-1 dilution rate gave the best results. A comparative study on fungal laccases immobilized on chitosan BABT

The phenoloxidase enzyme laccase from the cultures of the Pleurotus ostreatus and Botryosphaeria sp. and a commercial laccase from Aspergillus sp. were immobilized on chitosan of pharmaceutical degree by adsorption followed by crosslinking. Different immobilization conditions in relation to the granulometry of support and amount of enzymatic laccase extract used were tested, aiming at reaching high enzymatic activity with the immobilized enzyme. Three reactors (R1, R2 and R3) were constructed in blue PVC pipes measuring 30 cm height and 150 mm diameter and filled with coffee capsules made of aluminum and plastic. The sewage from the pre-treatment phase of the wastewater treatment plant of the Federal University of Lavras fed the system. Comparative performance of microbial lipases immobilized on magnetic polysiloxane polyvinyl alcohol particles BABT

Microbial lipase from Mucor miehei and Candida rugosa were immobilized by covalent binding onto magnetized polysiloxane polyvinyl alcohol particles (POS-PVA). The resulting immobilized derivatives were evaluated in aqueous solution (olive oil hydrolysis) and organic solvent (butyl butyrate synthesis). Higher catalytic activities were found when the coupling procedure was made with M. miehei lipase. Immobilized M. miehei lipase also showed a better operational stability and a higher half-life than C. rugosa lipase after the successive batches of esterification. ABSTRACT Response surface methodology was used to optimize lincomycin production by Streptomyces lincolnensis NRRL ISP-5355 in submerged fermentation. Screening of fermentation medium components to find their relative effect on lincomycin production was done using Plackett-Burman design. Malt extract, dextrin, soluble starch and (NH4)2SO4 were the most significant nutrient influenced on lincomycin production. Central composite design was applied to determine optimal concentrations of these factors and the effect of their mutual interactions. The production of sorbitol by permeabilized and immobilized cells of Z. mobilis in sucrose BABT

The production of sorbitol by permeabilized and immobilized cells of Zymomonas mobilis in Luffa cylindrica was investigated in sucrose medium. A full 2³ factorial design was used to verify the influence of each factor and its interactions. The cell permeabilization showed a significant and negative effect upon the production of sorbitol, while the time of cultivation and the immobilization process were significant and positive. The results demonstrated that the cell immobilization and the time of cultivation of 36 h presented higher production of sorbitol.

This study evaluated the biological importance of immobilized urease enzyme over the free urease. The support material used for urease immobilization was alginate. Generally, the immobilization of urease in alginate gel showed a marked increase in Km and Vmax. However, the immobilized urease showed higher thermal stability than that of free enzyme. The rate of thermal inactivation of the immobilized enzyme decreased due to entrapment in gel matrix. Also, the activity of the immobilized urease was more stable in retention than that of the free enzyme during the storage in solution, although the activity of the immobilized enzyme was lower in comparison with the free enzyme. The objective of the present study was to determine to what extent, if any, swimming training applied before immobilization in a cast interferes with the rehabilitation process in rat muscles. Cyclosporin-A (CsA) is an immunosuppressive drug that acts as an inhibitor of calcineurin, a calcium phosphatase that has been suggested to play a role in skeletal muscle hypertrophy. The aim of the present study was to determine the effect of CsA administration (25 mg kg-1 day-1) on skeletal muscle mass and phenotype during disuse and recovery. We investigated the effects of immobilization on excitability and compound action potential (CAP) and the ultrastructure of the rat sciatic nerve. Xylofucoglucuronan from Spatoglossum schröederi algae was tested as a support for antibiotic immobilization. The polysaccharide (20 mg in 6 ml) was first activated using carbodiimide, 1-ethyl-3-(3-dimethylamino-propyl)carbodiimide methiodide (20 mg in 2 ml), under stirring for 1 h at 25ºC and pH from 4.5 to 5.0. We determined the effect of stretching applied once a week to the soleus muscle immobilized in the shortened position on muscle fiber morphology. Twenty-six male Wistar rats weighing 269 ± 26 g were divided into three groups.
