Cytogenetic analysis of three catfish species of the family Pseudopimelodidae (Teleostei, Siluriformes) Genet. Mol. Biol.
Martinez,Emanuel Ricardo Monteiro; Nirchio,Mauro; Granado,Angel; Foresti,Fausto; Oliveira,Claudio.
Cytogenetic analyses performed in Cephalosilurus apurensis, Microglanis aff. cottoides and Pseudopimelodus bufonius revealed that the three species have 2n = 54 chromosomes: C. apurensis presented six metacentric (M), 28 submetacentric (SM), 14 subtelolocentric (ST), and six acrocentric (A) chromosomes, while M. aff. cottoides showed 10M, 32SM, 10ST and 2A, and P. bufonius had 12M, 30SM and 12ST. The nucleolus organizer regions (NORs) were present on the short arm of a middle-sized ST pair, identified as pair 19, in C. apurensis NORs were found on the short arm of a middle-sized ST (pair 23) and on the long arm of a middle-sized ST (pair 22) in M. aff. cotttoides and on the short arm of three middle-sized ST pairs, identified as pairs 9, 10 and 11, in P....

Detection of diploid males in a natural colony of the cleptobiotic bee Lestrimelitta sp (Hymenoptera, Apidae) Genet. Mol. Biol.
Tavares,Mara Garcia; Carvalho,Carlos Roberto; Soares,Fernanda Aparecida Ferrari; Fernandes,Anderson.
When working at quantifying the genome size of stingless bees, it was observed that males of Lestrimelitta sp possessed the same amount of nuclear DNA as the females. Thus, we used flow cytometry (FCM) and cytogenetic analysis to confirm the ploidy of these individuals. The males analyzed proved to be diploid, since, through cytometric analysis, it was demonstrated that the mean genome size of both males and females was the same (C = 0.463 pg), and, furthermore, cytogenetic analysis demonstrated that both had 2n = 28 chromosomes.

Mapping of ribosomal genes and chromosomal markers in three species of the genus Serrasalmus (Characidae, Serrasalminae) from the Amazon basin Genet. Mol. Biol.
Nakayama,Celeste M.; Feldberg,Eliana; Bertollo,Luiz Antonio C..
Karyotypic characteristics of three species of the genus serrasalmus ( S. altispinnis , S. gouldingi and S. Serrulatus ) from the middle and lower Negro River, Amazon Basin, were investigated using different staining techniques and Fluorescent in situ hybridization with 5S and 18S rDNA probes. The diploid number was invariably 2n = 60 and the fundamental number was FN = 110. Nevertheless, the karyotypes differed from each other in composition: 24m, 20sm, 6st, 10a in S. altispinnis ; 22m, 22sm, 6st, 10a in S. gouldingi and 20m, 22sm, 8st, 10a in S. serrulatus. The karyotype of S. altispinnis differed from the one previously described in a population from the Pitinga River. C-positive constitutive heterochromatin was mainly pericentromeric in...

Esterase-D and chromosome patterns in Central Amazon piranha (Serrasalmus rhombeus Linnaeus, 1766) from Lake Catalão Genet. Mol. Biol.
Teixeira,Aylton Saturnino; Nakayama,Celeste Mutuko; Porto,Jorge Ivan Rebelo; Feldberg,Eliana.
This study presents additional genetic data on piranha (Serrasalmus rhombeus Linnaeus, 1766) complex previously diagnosed due to the presence of distinct cytotypes 2n = 58 and 2n = 60. Three esterase-D enzyme loci (Est-D1, Est-D2 and Est-D3) were examined and complemented with chromosomal data from 66 piranha specimens collected from Lake Catalão. For all specimens the Est-D1 and Est-D2 loci were monomorphic. In contrast, the Est-D3 locus was polymorphic with genotypes and alleles being differentially distributed in the previously described cytotypes and served as the basis for detecting a new cytotype (2n = 60 B). In cytotype 2n = 58 the Est-D3 locus was also polymorphic and presented Mendelian allelic segregation with four genotypes (Est-D3(11),...

Comparative cytogenetics of Hoplias malabaricus (Pisces, Erythrinidae): a population analysis in adjacent hydrographic basins Genet. Mol. Biol.
Vicari,Marcelo Ricardo; Artoni,Roberto Ferreira; Bertollo,Luiz Antonio Carlos.
The chromosomes of specimens from four Hoplias malabaricus populations from headwaters of adjacent river basins at Ponta Grossa, southern Brazil, were investigated using differential staining techniques (C-banding, AgNO3 and CMA3) and fluorescent in situ hybridization (FISH) with an 18S rDNA probe. The diploid chromosome number in representatives of all four populations was invariably 2n = 42, with karyotypes composed of 12 pairs of metacentrics and 9 pairs of submetacentrics, without heteromorphic sex chromosomes. This kind of karyotype represents cytotype A in regard to cytotypes identified previously in H. malabaricus, exhibiting however, at the same time, some differences in the distribution of constitutive heterochromatin segments and in the locations...

Cytogenetic and morphological diversity in populations of Astyanax fasciatus (Teleostei, Characidae) from Brazilian northeastern river basins Genet. Mol. Biol.
Medrado,Aline Souza; Figueiredo,Alba Vivian Amaral; Waldschmidt,Ana Maria; Affonso,Paulo Roberto Antunes de Mello; Carneiro,Paulo Luiz Souza.
In the present work, morphometric and cytogenetic analyses were carried out in populations of the fish Astyanax fasciatus (Characidae) from Contas and Recôncavo Sul River basins (State of Bahia, Brazil), providing new data on the genetic structure of this species along the region. Based on morphologic measurements, we observed that populations from the same hydrographic basin were more similar to each other (Contas and Preto do Costa Rivers), and remarkably divergent from Recôncavo Sul (Mineiro Stream), as indicated by clustering analysis. Cytogenetic data revealed a same diploid number for all populations (2n = 48), but distinct karyotype formulae (8M+24SM+12ST+4A, FN = 92 in Contas River, 8M+24SM+10ST+6A, FN = 90 in Preto do Costa River, and...

The karyotype of the critically endangered Lear's macaw, Anodorhynchus leari Bonaparte 1856 (Aves, Psittaciformes) Genet. Mol. Biol.
Nogueira,Denise Monnerat; Souza,Lucia Moreno de; Goldschmidt,Beatriz; Silva,Christiano Pinheiro da; Monsores,Denise Wilches.
We used conventional chromosomal staining to describe the karyotype of the critically endangered Lear's macaw, Anodorhynchus leari Bonaparte 1856. A diploid number of 2n = 70 and a karyotype similar to that of its congener Anodorhynchus hyacinthinus suggests that chromosomal rearrangements were not the main evolutionary mechanism in the genus.

Description of the karyotypes of Stejneger's beaked whale (Mesoplodon stejnegeri) and Hubbs’ beaked whale (M. carlhubbsi) Genet. Mol. Biol.
Kurihara,Nozomi; Tajima,Yuko; Yamada,Tadasu K.; Matsuda,Ayaka; Matsuishi,Takashi.
Abstract The genus Mesoplodon (Cetacea: Odontoceti: Ziphiidae) is one of the few cetacean genera with the karyotype 2n = 42. The 2n = 42 karyotype of M. europaeus and M. carlhubbsi is largely consistent with the general cetacean karyotype 2n = 44, although other 2n = 42 karyotypes do not exhibit clear homologies with the general cetacean karyotype. Therefore, the chromosomes of Mesoplodon species may be the key to understanding cetacean karyological evolution. In the present study, the male karyotypes of M. stejnegeri and M. carlhubbsi were examined. In both species, the diploid number of the male karyotype was 42. Both species had the following characteristics: 1) a huge subtelocentric X chromosome with a large C-block; 2) a small metacentric Y...

Spermatogenesis and nucleolar activity in Triatoma klugi (triatomine, Heteroptera) Genet. Mol. Biol.
Costa,Laiana Cristina da; Azeredo-Oliveira,Maria Tercília Vilela de; Tartarotti,Ester.
Triatoma klugi is a Chagas disease vector in the Rio Grande do Sul State. Triatominae chromosomes are holocentric and sex chromosomes segregation is post-reductional. In this paper we describe the karyotype of male T. klugi and a meiotic analysis including the nucleolar behavior during spermatogenesis. Testis cells were analyzed after lacto-acetic orcein and silver nitrate staining. Two autosomes and the heterochromosomes presented nucleolar activity (Ag-NORs) during diplotene-diakinesis. The analysis of metaphase I and II revealed a karyotype with 2n = 20+XY. In metaphase I a prominent nucleolar mass was observed in the cell periphery and small silver grains were detected in metaphase II. During anaphase, the chromosomes segregated in parallel and a...

Karyotype and nuclear DNA content of Trichomycterus areolatus (Siluriformes, Trichomycteridae) Genet. Mol. Biol.
Colihueque,Nelson; Corrales,Olga; Parraguez,Margarita.
Cytogenetic analysis of Trichomycterus areolatus, collected from the Tijeral and Huilma Rivers in southern Chile has shown a diploid chromosome number of 2n = 54, a fundamental number of FN = 106, and a karyotypic formula of 44m + 8sm + 2st. Intra-individual polymorphism of chromosome number (2n = 54, 55 and 56) in specimens from the Huilma River has also been documented, providing further evidence of the occurrence of this phenomenon in Trichomycterus. The karyotype exhibited large chromosome pairs: metacentric pairs 1 (relative length 7.54%), 2 (5.75%) and 3 (5.09%), submetacentric pair 23 (5.25%), and subtelocentic pair 27 (5.28%). Nuclear DNA content analysis showed an average value of 5.04 ± 1.09 pg/nucleus. This DNA content is higher than the mean...

Multiple interstitial ribosomal sites (NORs) in the Brazilian squirrel Sciurus aestuans ingrami (Rodentia, Sciuridae) with 2n = 40: an overview of Sciurus cytogenetics Genet. Mol. Biol.
Fagundes,Valéria; Christoff,Alexandre Uarth; Amaro-Ghilard,Renata Cecília; Scheibler,Daniel R.; Yonenaga-Yassuda,Yatiyo.
This is the first time the karyotype of the Brazilian squirrel Sciurusaestuans ingrami, with 2n = 40, is described. The karyotype of this species comprises 18 pairs of biarmed and one minute pair of acrocentric autosomes, a medium-sized submetacentric X and a medium-sized acrocentric Y. Four pairs have an interstitial secondary constriction, co-located with nucleolar organizer regions (NORs), identified by silver-staining technique and fluorescent in situ hybridization (FISH) with ribosomal 18S/28S probes. The occurrence of multiple interstitial NORs is rare in rodents, and this is one of the few examples, identified by a molecular cytogenetics approach.

Differentiation of mouse embryonic stem cells and their hybrids during embryoid body formation Genet. Mol. Biol.
Mittmann,Josane; Kerkis,Irina; Kawashima,Cinthia; Sukoyan,Marina; Santos,Enrico; Kerkis,Alexandre.
We studied the karyotypes of three hybrid clones of mouse embryonic stem cells and murine splenocytes (two having near diploid and one having near tetraploid chromosome numbers) and the characteristics of their differentiation during the formation of embryoid bodies. The X chromosome originating from embryonic stem cells may be lost in hybrids with a near diploid chromosome number and reprogramming of the "somatic" X may occur. The morphological data we obtained using light and electron microscopy revealed a correlation between the karyotype constitution of hybrid cells and their differentiation during the formation of embryoid bodies. At the beginning of development, the embryoid bodies derived from hybrid cells already showed an advanced degree of...

Baeza,Carlos M.; Finot,Víctor; Ruiz,Eduardo; Carrasco,Pedro; Novoa,Patricio; Rosas,Marcelo; Toro-Núñez,Oscar.
Abstract Alstroemeria L. (Alstroemeriaceae) represents one of the most diverse genera of vascular plants in Chile. It contains approximately 54 taxa, 40 of which are endemic. The “complex” Alstroemeria magnifica is endemic to Chile, and it comprises four varieties: A. magnifica var. magenta, A. magnifica var. magnifica, A. magnifica var. sierrae, and A. magnifica var. tofoensis. It is distributed from Coquimbo to the Valparaíso Region. We analyzed karyotypes of 10 populations along its natural distribution. All the populations presented an asymmetric karyotype, with 2n = 16 chromosomes but with three different karyotypic formulae. Alstroemeria magnifica var. magnifica and A. magnifica var. sierrae presented the same karyotypic fomula, and A. magnifica var....

Benzaquem,Denise Corrêa; Feldberg,Eliana; Porto,Jorge Ivan Rebelo; Gross,Maria Claudia; Zuanon,Jansen Alfredo Sampaio.
Karyotypes of six cichlid species of the genus Crenicichla were investigated. The species C. cincta, C. inpa, C. reticulata, C. lugubris, and C. cf. johanna were collected from Amazon basin, and C. britskii was collected from the Paraná-Paraguai basin. All of the analysed species showed 2n = 48 chromosomes; however, C. cincta, C. lugubris, C. cf. johanna, and C. britskii had a karyotype formula of 8M-SM+40ST-A, FN = 56, while C. inpa and C. reticulata exhibited the formula 6M-SM+42ST-A, FN = 54. Analysis of active Ag-NORs revealed two NOR-bearing chromosomes in all species; however, theses cistrons were located on different chromosome pairs and/or in different chromosome locations in each species. This condition bears evolutionary significance, since it is...

Genetic identification of bucktooth parrotfish Sparisoma radians (Valenciennes, 1840) (Labridae, Scarinae) by chromosomal and molecular markers Genet. Mol. Biol.
Paim,Fabilene Gomes; Brandão,José Henrique Souza Galdino; Sampaio,Iracilda; Affonso,Paulo Roberto Antunes de Mello; Diniz,Débora.
Parrotfishes (Labridae, Scarinae) comprise a large marine fish group of difficult identification, particularly during juvenile phase when the typical morphology and coloration of adults are absent. Therefore, the goal of this study was to test cytogenetic markers and DNA barcoding in the identification of bucktooth parrtotfish Sparisoma radians from the northeastern coast of Brazil. Sequencing of cytochrome c oxidase subunit I (COI) confirmed all studied samples as S. radians, and all showed high similarity (99-100%) with Caribbean populations. The karyotype of this species was divergent from most marine Perciformes, being composed of 2n = 46 chromosomes. These consisted of a large number of metacentric and submetacentric pairs with small amounts of...

Abstract The order Charadriiformes comprises three major clades: Lari and Scolopaci as sister group to Charadrii. Until now, only three Charadriiformes species have been studied by chromosome painting: Larus argentatus (Lari), Burhinus oedicnemus and Vanellus chilensis (Charadrii). Hence, there is a lack of information concerning the third clade, Scolapaci. Based on this, and to gain a better understanding of karyotype evolution in the order Charadriiformes, we applied conventional and molecular cytogenetic approaches in a species belonging to clade Scolopaci - the wattled jacana (Jacana jacana) - using Gallus gallus and Zenaida auriculata chromosome-specific probes. Cross-species evaluation of J. jacana chromosomes shows extensive genomic reshuffling...

The subgenus Ceratochloa of the genus Bromus includes a number of closely related allopolyploid forms or species that present a difficult taxonomic problem. The present work combines data concerning chromosome length, heterochromatin distribution and nuclear genome size of different 6x, 8x and 12x accessions in this subgenus. Special attention is paid to the karyotype structure and genomic constitution of duodecaploid plants recently found in South America. Hexaploid lineages possess six almost indistinguishable genomes and a nuclear DNA content between 12.72 pg and 15.10 pg (mean 1Cx value = 2.32 pg), whereas octoploid lineages contain the same six genomes (AABBCC) plus two that are characterized by longer chromosomes and a greater DNA content (1Cx = 4.47...

Cytogenetics of three Brazilian species of Eleutherodactylus (Anura, Leptodactylidae) with 22 chromosomes and re-analysis of multiple translocations in E. binotatus Genet. Mol. Biol.
Siqueira-Jr.,Sérgio; Ananias,Fernando; Recco-Pimentel,Shirlei M..
In this paper, we provide a cytogenetic analysis of Eleutherodactylus guentheri, E. parvus and E. binotatus. All of the species had a diploid chromosomal number of 2n = 22. The karyotypes of E. guentheri and E. parvus were very similar and differed only slightly in the morphology of pair 2. These two species also had an NOR-bearing secondary constriction on the long arms of pair 6. The karyotype of E. binotatus differed from those of E. guentheri and E. parvus in the morphology and size of the chromosomes, in the number of chromosomal arms, in the NOR location (detected on the short arms of pair 1), and in the pattern of heterochromatin. These results reinforce the differences between E. guentheri and E. binotatus and support the existence of two species...

Chromosome mapping of 5S rRNA genes differentiates Brazilian populations of Leporellus vittatus (Anostomidae, Characiformes) Genet. Mol. Biol.
Aguilar,Cecilia Teixeira de; Galetti Junior,Pedro Manoel.
Among the anostomid fishes, the genus Leporellus is represented by only three species: L. nattereri, endemic of the Amazon River, L. retropinnis, endemic of the Piracicaba River, and L. vittatus, widely distributed in rivers from Peru, Colombia, Guianas, and different major hydrographic basins of Brazil. A cytogenetic study carried out on specimens of Leporellus vittatus from three major Brazilian hydrographic basins evidenced a karyotype of 54 metacentric and submetacentric chromosomes. C-banding analysis revealed the presence of large pericentromeric heterochromatic segments in all chromosomes and a telomeric block coincident with the NOR sites. Ag, CMA3 or MM staining, and FISH with ribosomal probes located the 45S ribosomal genes on the terminal region...
